    Christian Mindfulness and Mental Health: Coping Through Sacred Traditions and Embodied Awareness

    Mindfulness is increasingly implemented as a tool in mental health practice for coping and self-care. Some Christians worry that these practices might be in conflict with their own tradition, while other Christian contexts are reclaiming the contemplative aspects of the faith. Though clinicians are not trained to teach on religious topics and ethically must avoid pushing religion onto clients, conceptualization and research extend the benefits of mindfulness practices for religious clients. This paper will discuss the evidence for using mindfulness in mental health treatment and connect mindfulness to the Christian tradition. The authors explore how intentional awareness and embodiment of the present moment are supported in Christian theology through the incarnation of Jesus and God’s attention of the physical body in the Christian scriptures. The authors also discuss how sacraments and prayer naturally overlap with mindfulness practices for the dual purposes of emotional healing and spiritual growth. To bolster the benefits of mindfulness in the psychological and religious realms, the purpose of this paper is to empower therapists to address client concerns of whether mindfulness is in conflict with Christianity, support clients in expanding current Christian religious coping, and provide Christian leaders with more information about how mindfulness elements are already present in Christian rituals and beliefs

    Approximate exploitability: Learning a best response in large games

    A standard metric used to measure the approximate optimality of policies in imperfect information games is exploitability, i.e. the performance of a policy against its worst-case opponent. However, exploitability is intractable to compute in large games as it requires a full traversal of the game tree to calculate a best response to the given policy. We introduce a new metric, approximate exploitability, that calculates an analogous metric using an approximate best response; the approximation is done by using search and reinforcement learning. This is a generalization of local best response, a domain specific evaluation metric used in poker. We provide empirical results for a specific instance of the method, demonstrating that our method converges to exploitability in the tabular and function approximation settings for small games.     Striated Rootlet and Nonfilamentous Forms of Rootletin Maintain Ciliary Function

    SummaryPrimary cilia are microtubule-based sensory organelles whose structures and functions must be actively maintained throughout animal lifespan to support signal transduction pathways essential for development and physiological processes such as vision and olfaction [1]. Remarkably, few cellular components aside from the intraflagellar transport (IFT) machinery are implicated in ciliary maintenance [2]. Rootletin, an evolutionarily conserved protein found as prominent striated rootlets or a nonfilamentous form, both of which are associated with cilium-anchoring basal bodies, represents a likely candidate given its well-known role in preventing ciliary photoreceptor degeneration in a mouse model [3, 4]. Whether rootletin is universally required for maintaining ciliary integrity, and if so, by what mechanism, remains unresolved. Here, we demonstrate that the gene disrupted in the previously isolated C. elegans chemosensory mutant che-10 encodes a rootletin ortholog that localizes proximally and distally to basal bodies of cilia harboring or lacking conspicuous rootlets. In vivo analyses reveal that CHE-10/rootletin maintains ciliary integrity partly by modulating the assembly, motility, and flux of IFT particles, which are critical for axoneme length control. Surprisingly, CHE-10/rootletin is also essential for stabilizing ciliary transition zones and basal bodies, roles not ascribed to IFT. Unifying these findings, we provide evidence that the underlying molecular defects in the che-10 mutant stem from disrupted organization/function of the periciliary membrane, affecting the efficient delivery of basal body-associated and ciliary components and resulting in cilium degeneration. Together, our cloning and functional analyses of C. elegans che-10 provide the first mechanistic insights into how filamentous and nonfilamentous forms of rootletin play essential roles in maintaining ciliary function in metazoans

    Relationship between nondestructive firmness measurements and commercially important ripening fruit stages for peaches, nectarines and plums

    Fruit firmness measurement is a good way to monitor fruit softening and to predict bruising damage during harvest and postharvest handling. Ripening protocols traditionally utilize a destructive penetrometer-type fruit firmness measure to monitor ripening. Until recently, methods of assessing fruit texture properties nondestructively were not commercially available. The nondestructive Sinclair iQ™ firmness tester was investigated to monitor ripening and predict bruising susceptibility in stone fruit. This work was carried out on four peach, three plum, and five nectarine cultivars over two seasons. The correlations between destructive and nondestructive firmness measurements were significant (p-value = 0.0001), although too low for commercial applications as they varied from r2 = 0.60–0.71 according to fruit type. Using a different approach, the relationship between destructive and nondestructive firmness measures was characterized in terms of segregating these fruit according to their stages of ripening. This was done by using discriminant analysis (66–90% agreement in ripeness stage classification was observed in validation tests). Discriminant analysis consistently segregated nondestructive firmness measured fruit into commercially important classes (“ready to eat”, “ready to buy”, “mature and immature”). These represented key ripening stages with different bruising potentials and consumer acceptance. This work points out the importance to relate nondestructive measurements directly to important commercial physiological stages rather than to correlate them with the current standard penetrometer values. Thus, destructive and nondestructive firmness measurements can be directly used to identify the stage of ripeness and potential susceptibility to bruising during postharvest changes. Further work is recommended to evaluate the performance of this nondestructive sensor in segregating fruit according to their stage of ripeness under packinghouse or processing plant conditions

    Assessing Health Concerns & Obstacles to Diesel Exposure Reduction in Vermont Diesel Vehicle Operators

    Background and Objectives: Diesel vehicle idling reduction is an important national environmental and legislative issue. Exposure to diesel exhaust is associated with significant morbidity and mortality, including: • Lung & esophageal cancer • Asthma • Cardiovascular disease • Neurotoxicity • Decreased sperm count & testosterone deficiency Drivers of diesel vehicles have specifically been shown to have increased incidence and death from lung cancer. Diesel engines emit a number of known hazardous chemicals, including carbon monoxide, nitric oxide, sulfur dioxide, benzene, formaldehyde, and acrolein, into the air supply. While public health efforts to reduce diesel idling in Vermont and elsewhere have identified employers’ significant financial incentives in fuel conservation, perhaps there is also a role for appealing to drivers themselves: the people who are incurring the most direct exposure. It is unknown, however, whether Vermont diesel vehicle operators are aware of the health effects of diesel exhaust – or, more significantly, whether they are concerned about it. In order to identify potential targets for future interventions to reduce diesel idling in Vermont, this study aims to probe the following: • Have Vermont drivers been educated about exhaust exposure? • Are they concerned about potential health effects of diesel? • Are they satisfied with their understanding of the health impact of diesel fuel? • What are their health concerns, more generally? • What resources for health information do they respect? • What are their specific obstacles to idling reduction?https://scholarworks.uvm.edu/comphp_gallery/1046/thumbnail.jp

    Solving Common-Payoff Games with Approximate Policy Iteration

    For artificially intelligent learning systems to have widespread applicability in real-world settings, it is important that they be able to operate decentrally. Unfortunately, decentralized control is difficult -- computing even an epsilon-optimal joint policy is a NEXP complete problem. Nevertheless, a recently rediscovered insight -- that a team of agents can coordinate via common knowledge -- has given rise to algorithms capable of finding optimal joint policies in small common-payoff games. The Bayesian action decoder (BAD) leverages this insight and deep reinforcement learning to scale to games as large as two-player Hanabi. However, the approximations it uses to do so prevent it from discovering optimal joint policies even in games small enough to brute force optimal solutions. This work proposes CAPI, a novel algorithm which, like BAD, combines common knowledge with deep reinforcement learning. However, unlike BAD, CAPI prioritizes the propensity to discover optimal joint policies over scalability. While this choice precludes CAPI from scaling to games as large as Hanabi, empirical results demonstrate that, on the games to which CAPI does scale, it is capable of discovering optimal joint policies even when other modern multi-agent reinforcement learning algorithms are unable to do so.     Loss of SMAD4 Is Associated With Poor Tumor Immunogenicity and Reduced PD-L1 Expression in Pancreatic Cancer

    Transforming Growth Factor β (TGFβ) is a key mediator of immune evasion in pancreatic ductal adenocarcinoma (PDAC), and the addition of TGFβ inhibitors in select immunotherapy regimens shows early promise. Though the TGFβ target SMAD4 is deleted in approximately 55% of PDAC tumors, the effects of SMAD4 loss on tumor immunity have yet to be fully explored. Using a combination of genomic databases and PDAC specimens, we found that tumors with loss of SMAD4 have a comparatively poor T-cell infiltrate. SMAD4 loss was also associated with a reduction in several chemokines with known roles in T-cell recruitment, which was recapitulated using knockdown of SMAD4 in PDAC cell lines. Accordingly, JURKAT T-cells were poorly attracted to conditioned media from PDAC cells with knockdown of SMAD4 and lost their ability to produce IFNγ. However, while exogenous TGFβ modestly reduced PD-L1 expression in SMAD4-intact cell lines, SMAD4 and PD-L1 positively correlated in human PDAC samples. PD-L1 status was closely related to tumor-infiltrating lymphocytes, particularly IFNγ-producing T-cells, which were more abundant in SMAD4-expressing tumors. Low concentrations of IFNγ upregulated PD-L1 in tumor cells in vitro, even when administered alongside high concentrations of TGFβ. Hence, while SMAD4 may have a modest inhibitory effect on PD-L1 in tumor cells, SMAD4 indirectly promotes PD-L1 expression in the pancreatic tumor microenvironment by enhancing T-cell infiltration and IFNγ biosynthesis. These data suggest that pancreatic cancers with loss of SMAD4 represent a poorly immunogenic disease subtype, and SMAD4 status warrants further exploration as a predictive biomarker for cancer immunotherapy
