In vivo-Studien zur ClpP-Maschinerie in Streptomyceten am Beispiel des ADEP-Produzenten Streptomyces hawaiiensis NRRL 15010 und des Modellorganismus Streptomyces lividans
Streptomyces hawaiiensis NRRL 15010 ist der natĂŒrliche Produzentenstamm des SekundĂ€rmetabolitkomplexes A54556, einem Extrakt mit bisher sechs strukturell aufgeklĂ€rten antibiotisch wirksamen Acyldepsipeptid-Derivaten (ADEPs). Diese Antibiotikaklasse zeigt eine vielversprechende AktivitĂ€t gegen verschiedenste Gram-positive Bakterien und weist einen vollkommen neuartigen Wirkmechanismus auf. Das ADEP-Target ist die proteolytische Kernkomponente ClpP der bakteriellen caseinolytischen Protease, einer ATP-abhĂ€ngigen Serinprotease, die eine wichtige Rolle fĂŒr die Proteinhomöostase und -qualitĂ€tskontrolle sowie die regulatorische Proteolyse spielt. Durch seine Bindung an ClpP verdrĂ€ngt ADEP die mit ClpP assoziierten Clp-ATPasen von ihren Bindestellen und inhibiert dadurch sĂ€mtliche natĂŒrlichen Funktionen der Protease. Des Weiteren induziert die ADEP-Bindung eine KonformationsĂ€nderung der ClpP-Fassstruktur, welche die Eintrittspore zum katalytischen Zentrum erweitert, so dass essentielle Proteine hineingelangen und abgebaut werden, was einen letalen Effekt auf die bakterielle Zelle hat. Im Rahmen dieser Arbeit wurde das bisher unbekannte ADEP-Biosynthesegencluster in S. hawaiiensis identifiziert und analysiert. In der NĂ€he des Genclusters wurde zudem ein zusĂ€tzliches clpP-Homolog, clpPADEP, gefunden, welches als Resistenzfaktor gegen ADEP bestĂ€tigt werden konnte. Die FunktionalitĂ€t von ClpPADEP sowie der durch dieses ClpP-Homolog vermittelte Resistenzmechanismus konnten nicht durch in vivo-Experimente im Produzentenstamm selbst untersucht werden, da es trotz einer Vielzahl angewandter Verfahren nicht gelang, den Stamm genetisch zu modifizieren. Allerdings konnte das Resistenzprinzip erfolgreich in den genetisch gut zugĂ€nglichen Modellorganismus S. lividans ĂŒbertragen und dort untersucht werden. Bioassay- und Western Blot-Analysen ergaben deutliche Hinweise auf einen neuartigen, dualen Resistenzmechanimus, bei dem ClpPADEP sowohl die Detoxifizierung von durch ADEP ĂŒberaktivierten ClpP-Komplexen, als auch die Bildung von funktionalen, ADEP-resistenten Komplexen bewirkt, und so das Ăberleben der bakteriellen Zelle sichert
Inducing Secondary Metabolite Production by the Endophytic Fungus <i>Fusarium tricinctum</i> through Coculture with <i>Bacillus subtilis</i>
Coculturing the fungal endophyte <i>Fusarium tricinctum</i> with the bacterium <i>Bacillus
subtilis</i> 168 trpC2
on solid rice medium resulted in an up to 78-fold increase in the
accumulation in constitutively present secondary metabolites that
included lateropyrone (<b>5</b>), cyclic depsipeptides of the
enniatin type (<b>6</b>â<b>8</b>), and the lipopeptide
fusaristatin A (<b>9</b>). In addition, four compounds (<b>1</b>â<b>4</b>) including (â)-citreoisocoumarin
(<b>2</b>) as well as three new natural products (<b>1</b>, <b>3</b>, and <b>4</b>) were not present in discrete
fungal and bacterial controls and only detected in the cocultures.
The new compounds were identified as macrocarpon C (<b>1</b>), 2-(carboxymethylamino)Âbenzoic acid (<b>3</b>), and (â)-citreoisocoumarinol
(<b>4</b>) by analysis of the 1D and 2D NMR and HRMS data. Enniatins
B1 (<b>7</b>) and A1 (<b>8</b>), whose production was
particularly enhanced, inhibited the growth of the cocultivated <i>B. subtilis</i> strain with minimal inhibitory concentrations
(MICs) of 16 and 8 ÎŒg/mL, respectively, and were also active
against <i>Staphylococcus aureus</i>, <i>Streptococcus
pneumoniae</i>, and <i>Enterococcus faecalis</i> with
MIC values in the range 2â8 ÎŒg/mL. In addition, lateropyrone
(<b>5</b>), which was constitutively present in <i>F. tricinctum</i>, displayed good antibacterial activity against <i>B. subtilis</i>,<i> S. aureus</i>, <i>S. pneumoniae</i>, and <i>E. faecalis,</i> with MIC values ranging from 2 to 8 ÎŒg/mL.
All active compounds were equally effective against a multiresistant
clinical isolate of <i>S. aureus</i> and a susceptible reference
strain of the same species
