    Ticks and Tick-Borne Pathogens in Domestic Animals, Wild Pigs, and Off-Host Environmental Sampling in Guam, USA

    Background: Guam, a United States of America (USA) island territory in the Pacific Ocean, is known to have large populations of ticks; however, it is unclear what the risk is to wildlife and humans living on the island. Dog (Canis familiaris), cat (Felis catus), and wild pig (Sus scrofa) sentinels were examined for ticks, and environmental sampling was conducted to determine the ticks present in Guam and the prevalence of tick-borne pathogens in hosts. Methods and Results: From March 2019-November 2020, ticks were collected from environmental sampling, dogs, cats, and wild pigs. Blood samples were also taken from a subset of animals. A total of 99 ticks were collected from 27 environmental samples and all were Rhipicephalus sanguineus, the brown dog tick. Most ticks were collected during the dry season with an overall sampling success rate of 63% (95% CI: 42.4–80.6). 6,614 dogs were examined, and 12.6% (95% CI: 11.8–13.4) were infested with at least one tick. One thousand one hundred twelve cats were examined, and six (0.54%; 95% CI: 0.20–1.1) were found with ticks. Sixty-four wild pigs were examined and 17.2% (95% CI: 9.5–27.8) had ticks. In total, 1,956 ticks were collected and 97.4% of ticks were R. sanguineus. A subset of R. sanguineus were determined to be the tropical lineage. The other tick species found were Rhipicephalus microplus (0.77%), Amblyomma breviscutatum (0.77 %), and a Haemaphysalis sp. (0.51%). Blood samples from 136 dogs, four cats, and 64 wild pigs were tested using polymerase chain reaction (PCR) and DNA sequencing methods. Five different tick-borne pathogens with the following prevalences were found in dogs: Anaplasma phagocytophilum 5.9% (95% CI: 2.6–11.3); Anaplasma platys 19.1% (95% CI: 12.9–26.7); Babesia canis vogeli 8.8% (95% CI: 4.6–14.9); Ehrlichia canis 12.5% (95% CI: 7.5–19.3); Hepatozoon canis 14.7% (95% CI: 9.2–28.8). E. canis was detected in one cat, and no tick-borne pathogens were detected in wild pigs. Overall, 43.4% (95% CI: 34.9–52.1) of dogs had at least one tick-borne pathogen. Serological testing for antibodies against Ehrlichia spp. and Anaplasma spp. showed prevalences of 14.7% (95% CI: 9.2–28.8) and 31.6% (95% CI: 23.9–40), respectively. Conclusion: Four different tick species were found in Guam to include a Haemaphysalis sp., which is a previously unreported genus for Guam. Dogs with ticks have a high prevalence of tick-borne pathogens which makes them useful sentinels

    Editor's Choice - Re-interventions After Repair of Ruptured Abdominal Aortic Aneurysm: A Report From the IMPROVE Randomised Trial.

    OBJECTIVE/BACKGROUND: The aim was to describe the re-interventions after endovascular and open repair of rupture, and investigate whether these were associated with aortic morphology. METHODS: In total, 502 patients from the IMPROVE randomised trial (ISRCTN48334791) with repair of rupture were followed-up for re-interventions for at least 3 years. Pre-operative aortic morphology was assessed in a core laboratory. Re-interventions were described by time (0-90 days, 3 months-3 years) as arterial or laparotomy related, respectively, and ranked for severity by surgeons and patients separately. Rare re-interventions to 1 year, were summarised across three ruptured abdominal aortic aneurysm trials (IMPROVE, AJAX, and ECAR) and odds ratios (OR) describing differences were pooled via meta-analysis. RESULTS: Re-interventions were most common in the first 90 days. Overall rates were 186 and 226 per 100 person years for the endovascular strategy and open repair groups, respectively (p = .20) but between 3 months and 3 years (mid-term) the rates had slowed to 9.5 and 6.0 re-interventions per 100 person years, respectively (p = .090) and about one third of these were for a life threatening condition. In this latter, mid-term period, 42 of 313 remaining patients (13%) required at least one re-intervention, most commonly for endoleak or other endograft complication after treatment by endovascular aneurysm repair (EVAR) (21 of 38 re-interventions), whereas distal aneurysms were the commonest reason (four of 23) for re-interventions after treatment by open repair. Arterial re-interventions within 3 years were associated with increasing common iliac artery diameter (OR 1.48, 95% confidence interval [CI] 0.13-0.93; p = .004). Amputation, rare but ranked as the worst re-intervention by patients, was less common in the first year after treatment with EVAR (OR 0.2, 95% CI 0.05-0.88) from meta-analysis of three trials. CONCLUSION: The rate of mid-term re-interventions after rupture is high, more than double that after elective EVAR and open repair, suggesting the need for bespoke surveillance protocols. Amputations are much less common in patients treated by EVAR than in those treated by open repair

    Routine handling methods affect behaviour of three-spined sticklebacks in a novel test of anxiety

    Fish are increasingly popular subjects in behavioural and neurobiological research. It is therefore important that they are housed and handled appropriately to ensure good welfare and reliable scientific findings, and that species-appropriate behavioural tests (e.g. of cognitive/affective states) are developed. Routine handling of captive animals may cause physiological stress responses that lead to anxiety-like states (e.g. increased perception of danger). In fish, these may be particularly pronounced when handling during tank-to-tank transfer involves removal from water into air. Here we develop and use a new combined scototaxis (preference for dark over light areas) and novel-tank-diving test, alongside conventional open-field and novel-object tests, to measure the effects of transferring three-spined sticklebacks (Gasterosteus aculeatus) between tanks using a box or net (in and out of water respectively). Preference tests for dark over light areas confirmed the presence of scototaxis in this species. Open-field and novel-object tests failed to detect any significant differences between net and box-handled fish. However, the combined diving and scototaxis detected consistent differences between the treatments. Net-handled fish spent less time on the dark side of the tank, less time in the bottom third, and kept a greater distance from the ‘safe’ bottom dark area than box-handled fish. Possible explanations for this reduction in anxiety-like behaviour in net-handled fish are discussed. The combined diving and scototaxis test may be a sensitive and taxon-appropriate method for measuring anxiety-like states in fish

    Engineering of an environmental isolate of bacillus megaterium for biochemical production under supercritical CO2

    Continuous processing is a mainstay for chemical production but is far less common for biochemical processes. The increase in productivity and corresponding decrease in costs make continuous processing an intriguing option for bulk chemicals where price is a major consideration. Among the various challenges of continuous bioprocessing are the risks of contamination and the toxicity of the target products. Supercritical carbon dioxide (scCO2) may provide a means to address both of these issues. scCO2 is an attractive substitute for conventional organic solvents due to its unique transport and thermodynamic properties, its renewability and labile nature, and its high solubility for compounds such as alcohols, ketones and aldehydes. scCO2 is also known for its broad microbial lethality. The isolation and engineering of a microbe that is capable of growth and production in the presence of scCO2 thus represents an opportunity to create a production environment that is both resist to contamination and capable of sequestering toxic products through phase separation. Using a targeted bioprospecting approach by sampling fluid from a natural, deep-subsurface scCO2 well, a strain of Bacillus megaterium was isolated that is able to germinate and grow in the presence of scCO2. Transformation is possible using a protoplast-based method, which permitted the identification of promoters capable of inducible heterologous protein expression in both aerobic and anaerobic conditions. A xylose-inducible promoter was evaluated under scCO2 and found to have similar expression under both conditions. We engineered the B. megaterium strain to produce isobutanol from 2-ketoisovalerate by introducing a two-enzyme pathway (2- ketoisovalerate decarboxylase (KivD) and alcohol dehydrogenase (Adh)). Due to the strong partition of the aldehyde to the scCO2 phase, we tested five homologous Adh enzymes and found that YqhD from E. coli resulted in greater than 85% conversion when grown aerobically. Isobutanol production was also observed when our recombinant strain was cultured under scCO2. Finally, we have developed a process model for an integrated bioprocess and have found conditions that are comparable if not better than existing in situ extraction techniques such as gas stripping.
