    Projections for future radiocarbon content in dissolved inorganic carbon in hardwater lakes: a retrospective approach

    Inland water bodies contain significant amounts of carbon in the form of dissolved inorganic carbon (DIC) derived from a mixture of modern atmospheric and pre-aged sources, which needs to be considered in radiocarbon-based dating and natural isotope tracer studies. While reservoir effects in hardwater lakes are generally considered to be constant through time, a comparison of recent and historical DI14C data from 2013 and 1969 for Lake Constance reveals that this is not a valid assumption. We hypothesize that changes in atmospheric carbon contributions to lake water DIC have taken place due to anthropogenically forced eutrophication in the 20th century. A return to more oligotrophic conditions in the lake led to reoxygenation and enhanced terrigenous organic matter remineralization, contributing to lake water DIC. Such comparisons using DI14C measurements from different points in time enable nonlinear changes in lake water DIC source and signature to be disentangled from concurrent anthropogenically induced changes in atmospheric 14C. In the future, coeval changes in lake dynamics due to climate change are expected to further perturb these balances. Depending on the scenario, Lake Constance DI14C is projected to decrease from the 2013 measured value of 0.856 Fm to 0.54–0.62 Fm by the end of the century

    Platelet-activating factor receptor affects food intake and body weight

    “Let’s Move!” is a comprehensive initiative, launched by the First Lady, Michelle Obama, dedicates to solving problems of obesity, which is growing in child. The life behaviors do affect obesity; however, the mechanistic insight in molecular level is still not clear. In this study, by continually monitoring mouse body weight under chow and high fat western diets as well as metabolic, physical activity and food intake behaviors assessed in a CLAMS Comprehensive Lab Animal Monitoring System, we demonstrated that the platelet-activating factor receptor (PTAFR) contributes to modification of life behaviors. PTAFR does not affect metabolism of ingested dietary fat and carbohydrate in young animals; however, Ptafr ablation dramatically increased weight gain without affecting adipose tissue accumulation. Ptafr−/− mice possess new habits that increased food intake and decreased movement. Our studies suggest that regulation of PTAFR activity may be a novel strategy to control obesity in children or young adults

    Petrogenic organic carbon retention in terrestrial basins: a case study from perialpine Lake Constance

    Inland waters play a major role in the global carbon cycle, with particulate organic carbon (POC) burial in terrestrial wetlands surpassing that in ocean sediments. Lake Constance, the second largest lake at the periphery of the European Alps, receives POC sourced from both aquatic and terrestrial productivity as well as petrogenic OC (OCpetro) from bedrock erosion. Distinguishing POC inputs to lake sediments is key to assessing carbon flux and fate as reworked OCpetro represents neither a net sink of atmospheric CO2 nor source of O2. New stable and radiocarbon isotopic data indicate that 11 (9–12) Gg/yr of OCpetro is buried in Lake Constance with underlying sediments on average containing 0.3 (0.25–0.33) wt% OCpetro. Extrapolation of these results suggests that 27 TgOCpetro/yr (12–54 TgOC/yr) could be subject to temporary geological storage in lakes globally, which is comparable to estimates of 43−25+61 TgOCpetro/yr delivered to the ocean by rivers (Galy et al., 2015). More studies are needed to quantify OCpetro burial in inland sedimentary reservoirs in order to accurately account for atmospheric carbon sequestration in terrestrial basins

    Prioritizing ecological restoration among sites in multi‐stressor landscapes

    Most ecosystems are impacted by multiple local and long‐distance stressors, many of which interact in complex ways. We present a framework for prioritizing ecological restoration efforts among sites in multi‐stressor landscapes. Using a simple model, we show that both the economic and sociopolitical costs of restoration will typically be lower at sites with a relatively small number of severe problems than at sites with numerous lesser problems. Based on these results, we propose using cumulative stress and evenness of stressor impact as complementary indices that together reflect key challenges of restoring a site to improved condition. To illustrate this approach, we analyze stressor evenness across the world’s rivers and the Laurentian Great Lakes. This exploration reveals that evenness and cumulative stress are decoupled, enabling selection of sites where remediating a modest number of high‐intensity stressors could substantially reduce cumulative stress. Just as species richness and species evenness are fundamental axes of biological diversity, we argue that cumulative stress and stressor evenness constitute fundamental axes for identifying restoration opportunities in multi‐stressor landscapes. Our results highlight opportunities to boost restoration efficiency through strategic use of multi‐stressor datasets to identify sites that maximize ecological response per stressor remediated.     Integrin-dependent control of translation: engagement of integrin alphaIIbbeta3 regulates synthesis of proteins in activated human platelets.

    Integrins are widely expressed plasma membrane adhesion molecules that tether cells to matrix proteins and to one another in cell-cell interactions. Integrins also transmit outside-in signals that regulate functional responses of cells, and are known to influence gene expression by regulating transcription. In previous studies we found that platelets, which are naturally occurring anucleate cytoplasts, translate preformed mRNA transcripts when they are activated by outside-in signals. Using strategies that interrupt engagement of integrin alphaIIbbeta3 by fibrinogen and platelets deficient in this integrin, we found that alphaIIbbeta3 regulates the synthesis of B cell lymphoma 3 (Bcl-3) when platelet aggregation is induced by thrombin. We also found that synthesis of Bcl-3, which occurs via a specialized translation control pathway regulated by mammalian target of rapamycin (mTOR), is induced when platelets adhere to immobilized fibrinogen in the absence of thrombin and when integrin alphaIIbbeta3 is engaged by a conformation-altering antibody against integrin alphaIIbbeta3. Thus, outside-in signals delivered by integrin alphaIIbbeta3 are required for translation of Bcl-3 in thrombin-stimulated aggregated platelets and are sufficient to induce translation of this marker protein in the absence of thrombin. Engagement of integrin alpha2beta1 by collagen also triggered synthesis of Bcl-3. Thus, control of translation may be a general mechanism by which surface adhesion molecules regulate gene expression

    Fish Distributions and Nutrient Cycling in Streams: Can Fish Create Biogeochemical Hotspots?

    Rates of biogeochemical processes often vary widely in space and time, and characterizing this variation is critical for understanding ecosystem functioning. In streams, spatial hotspots of nutrient transformations are generally attributed to physical and microbial processes. Here we examine the potential for heterogeneous distributions of fish to generate hotspots of nutrient recycling. We measured nitrogen (N) and phosphorus (P) excretion rates of 47 species of fish in an N-limited Neotropical stream, and we combined these data with population densities in each of 49 stream channel units to estimate unit- and reach-scale nutrient recycling. Species varied widely in rates of N and P excretion as well as excreted N:P ratios (6–176 molar).     Monitoring outdoor tobacco policies of Virginia colleges: A descriptive analysis

    Purpose: The purpose of this study was to monitor current outdoor tobacco policies of colleges within the state of Virginia. Methods: The tobacco policies of 2-year public colleges, 4-year public colleges, and 4-year private non-profit colleges in Virginia were located online. The policies were then categorized according to the types of tobacco products that were prohibited (Smoking Policies, Tobacco Policies, and E-cigarette Policies) and where those products were prohibited outdoors (No Policy, Entrance Policy, Perimeter Policy, Designated Smoking Areas Policy, All Grounds Policy). Findings: From a final sample of 62 college policies, 2 (3%) had No Policy, 29 (47%) had an Entrance Policy, 12 (19%) had a Perimeter Policy, 10 (16%) had a Designated Smoking Areas Policy, and 9 (15%) had an All Grounds Policy. Conclusions: The far majority of colleges do not meet national recommendations for outdoor tobacco policies. Practical implications and suggestions for future research are discussed in the article

    Online 13C and 14C gas measurements by EA-IRMS–AMS at ETH Zürich

    Studies using carbon isotopes to understand the global carbon cycle are critical to identify and quantify sources, sinks, and processes and how humans may impact them. 13C and 14C are routinely measured individually; however, there is a need to develop instrumentation that can perform concurrent online analyses that can generate rich data sets conveniently and efficiently. To satisfy these requirements, we coupled an elemental analyzer to a stable isotope mass spectrometer and an accelerator mass spectrometer system fitted with a gas ion source. We first tested the system with standard materials and then reanalyzed a sediment core from the Bay of Bengal that had been analyzed for 14C by conventional methods. The system was able to produce %C, 13C, and 14C data that were accurate and precise, and suitable for the purposes of our biogeochemistry group. The system was compact and convenient and is appropriate for use in a range of fields of research
