    Unusual Presentation of a Left Testicular Carcinoid

    Testicular carcinoid generally presents either with a palpable (painful or painless) or rarely a nonpalpable mass (when it is diagnosed incidentally on scrotal imaging) of the involved testis. An ipsilateral testicular carcinoid presenting exclusively with contralateral testicular symptoms has never been described in the literature to date. We report a case of nonpalpable left testicular carcinoid diagnosed incidentally on a sonogram done for right testicular pain and swelling, with conspicuous absence of left testicular symptoms. This case highlights the importance of recognizing such an atypical presentation of testicular carcinoid

    Dispersive detection of radio-frequency-dressed states

    We introduce amethod to dispersively detect alkali-metal atoms in radio-frequency-dressed states. In particular, we use dressed detection tomeasure populations and population differences of atoms prepared in their clock states. Linear birefringence of the atomic medium enables atom number detection via polarization homodyning, a form of common path interferometry. In order to achieve low technical noise levels, we perform optical sideband detection after adiabatic transformation of bare states into dressed states. The balanced homodyne signal then oscillates independently of field fluctuations at twice the dressing frequency, thus allowing for robust, phase-locked detection that circumvents low-frequency noise. Using probe pulses of two optical frequencies, we can detect both clock states simultaneously and obtain population difference as well as the total atom number. The scheme also allows for difference measurements by direct subtraction of the homodyne signals at the balanced detector, which should technically enable quantum noise limited measurements with prospects for the preparation of spin squeezed states. The method extends to other Zeeman sublevels and can be employed in a range of atomic clock schemes, atom interferometers, and other experiments using dressed atoms

    Transcriptional activators in yeast

    Eukaryotic transcription activation domains (ADs) are not well defined on the proteome scale. We systematicallly tested ∼6000 yeast proteins for transcriptional activity using a yeast one-hybrid system and identified 451 transcriptional activators. We then determined their transcription activation strength using fusions to the Gal4 DNA-binding domain and a His3 reporter gene which contained a promoter with a Gal4-binding site. Among the 132 strongest activators 32 are known transcription factors while another 35 have no known function. Although zinc fingers, helix–loop–helix domains and several other domains are highly overrepresented among the activators, only few contain characterized ADs. We also found some striking correlations: the stronger the activation activity, the more acidic, glutamine-rich, proline-rich or asparagine-rich the activators were. About 29% of the activators have been found previously to specifically interact with the transcription machinery, while 10% are known to be components of transcription regulatory complexes. Based on their transcriptional activity, localization and interaction patterns, at least six previously uncharacterized proteins are suggested to be bona fide transcriptional regulators (namely YFL049W, YJR070C, YDR520C, YGL066W/Sgf73, YKR064W and YCR082W/Ahc2)

    Co-ingestion of black tea reduces the indispensable amino acid digestibility of hens’ egg in Indian adults

    Background: Tea, a commonly consumed beverage, contains high amounts of polyphenols that can impair protein digestibility, as demonstrated in vitro. There are no human studies examining the inhibitory influence of tea polyphenols (TPP) on high-quality protein digestibility. Objective: The aim of this study was to determine the effect of black tea on the true indispensable amino acid (IAA) digestibility of whole boiled egg protein, in healthy adult humans, through use of a dual isotope tracer approach. Methods: The effect of black TPP (4.6 mg/mL, ingested as a beverage with the meal) on 2H-labeled whole boiled egg protein, administered with ghee rice and tomato curry, was measured with reference to 13C-spirulina protein in healthy Indian adults aged 20–27 y of both sexes with BMI of 22.0 ± 2.8 kg/m2. The results were then compared to previously determined whole egg mean IAA digestibility measured by the same method, without black tea, in the same subjects (n = 5). To correct for any independent effect of TPP on spirulina protein (used as a standard protein), the true IAA digestibility of 13C-spirulina protein was independently measured with reference to a 2H-amino acid mixture, with and without co-ingestion of black tea, in 3 of the same subjects. Results: The true IAA digestibility of whole boiled egg protein significantly decreased by 17% when co-ingested with black tea. However, there was no significant reduction in the true IAA digestibility of spirulina protein when co-ingested with black tea. Conclusions: TPP protein interactions reduced whole egg digestibility in healthy Indian adults but had minimal effect on spirulina protein digestibility. In populations who are at risk of dietary quality protein inadequacy, the consumption of tea during or after a meal can further increase the risk of inadequacy.     We describe our progress in the development of an atom based rotation sensor, which employs state-dependent trapping potentials to transport ultracold atoms along a closed path and perform Sagnac interferometry. Whilst guided atom interferometers are sought after to build miniaturized devices that overcome size restrictions fromfree-falling atoms, fully trapped interferometers also remove free-propagation along an atomic waveguide. This provides additional control of motion, e.g. removing wave-packet dispersion and enabling operation that remains independent of external acceleration. Our experimental scheme relies on radio-frequency and microwave-fields,which are partly generated via atom-chip technology, providing a step towards implementing a small, robust, and eventually portable atomic-gyroscope

    True ileal digestibility of legumes determined by dual isotope tracer method in Indian adults

    Background: Good-quality plant protein sources are important for protein adequacy in a balanced diet. Legumes are known to be a source of good quality plant protein, but the true ileal digestibility of indispensable amino acids (IAAs) of commonly consumed legumes is not known in humans. Objectives: In this study we measured the true ileal IAA digestibility of 2H-intrinsically labeled chickpea, yellow pea, and mung bean (hulled and dehulled) protein, using the dual-isotope tracer technique referenced to a standard protein ([U-13C] spirulina). The study also aimed to validate the use of [U-13C] spirulina as a reference protein in this method. Methods: 2H-intrinsically labeled legumes, obtained by watering plants with deuterium oxide (2H2O), were administered in a plateau feeding method to healthy Indian adults to measure their true ileal IAA digestibility with the dual-isotope tracer technique, using [U-13C] spirulina protein or a 13C-algal IAA mixture as the standard. Result: The true ileal IAA digestibilities (mean ± SD) of chickpea, yellow pea, and mung bean were 74.6 ± 0.8%, 71.6 ± 1.3%, and 63.2 ± 1.5%, respectively. The true mean ileal IAA digestibility of mung bean when referenced to [U-13C] spirulina protein or a 13C-algal IAA mixture did not differ significantly (63.2 ± 1.5% versus 64.0 ± 2.4%, P > 0.05). The true ileal IAA digestibility of mung bean improved to 70.9 ± 2.1% after dehulling. Conclusions: The true mean ileal IAA digestibility of legumes in healthy Indian adults was lower than expected. Traditional processing techniques such as dehulling improve protein digestibility by about 8%.     Heterodera glycines utilizes promiscuous spliced leaders and demonstrates a unique preference for a species-specifc spliced leader over C. elegans SL1

    Spliced leader trans-splicing (SLTS) plays a part in the maturation of pre-mRNAs in select species across multiple phyla but is particularly prevalent in Nematoda. The role of spliced leaders (SL) within the cell is unclear and an accurate assessment of SL occurrence within an organism is possible only after extensive sequencing data are available, which is not currently the case for many nematode species. SL discovery is further complicated by an absence of SL sequences from high-throughput sequencing results due to incomplete sequencing of the 5\u27-ends of transcripts during RNA-seq library preparation, known as 5\u27-bias. Existing datasets and novel methodology were used to identify both conserved SLs and unique hypervariable SLs within Heterodera glycines, the soybean cyst nematode. In H. glycines, twenty-one distinct SL sequences were found on 2,532 unique H. glycines transcripts. The SL sequences identified on the H. glycines transcripts demonstrated a high level of promiscuity, meaning that some transcripts produced as many as nine different individual SL-transcript combinations. Most uniquely, transcriptome analysis revealed that H. glycines is the first nematode to demonstrate a higher SL trans-splicing rate using a species-specific SL over well-conserved Caenorhabditis elegans SL-like sequences

    Protein-quality evaluation of complementary foods in Indian children

    Background: The types of food in complementary feeding of infants and young children are important for growth and development. Food protein quality, as measured by the Digestible Indispensable Amino Acid Score (DIAAS), requires the determination of true ileal digestibility of indispensable amino acids (IAAs) in children. Objectives: First, the aim of this study was to measure the true ileal IAA digestibility of 4 (rice, finger millet, mung bean, and hen egg) commonly consumed complementary foods in children aged <2 y using the dual-isotope tracer method. Second, we calculated the DIAAS of complementary feeding diets and their relation to stunting in a representative Indian rural population. Design: Rice, finger millet, and mung bean were intrinsically labeled with deuterium oxide (2H2O), whereas egg was labeled through oral dosing of hens with a uniformly 2H-labeled amino acid mixture. True ileal IAA digestibility was determined by the dual-isotope tracer technique. The DIAAS of complementary food protein was calculated in children aged 1–3 y from a nationally representative survey to evaluate its relation with stunting. Results: True ileal IAA digestibility was lowest in mung bean (65.2% ± 7.1%), followed by finger millet (68.4 %± 5.3%) and rice (78.5% ± 3.5%), and was highest for egg (87.4% ± 4.0%). There was a significant inverse correlation of complementary food DIAAS with stunting in survey data (r = −0.66, P = 0.044). The addition of egg or milk to nationally representative complementary diets theoretically improved the DIAAS from 80 to 100. Conclusions: The true ileal IAA digestibility of 4 foods commonly consumed in complementary diets showed that the DIAAS was associated with stunting and reinforces the importance of including animal source food (ASF) in diets to improve growth.     Comparative evaluation of the therapeutic efficacy of rumen buffers and direct fed microbials (DFM) in sub-acute ruminal acidosis in dairy cows

    Sub-acute ruminal acidosis (SARA) in dairy cattle is a disorder of ruminal fermentation caused by the ingestion of large amounts of concentrates with inadequate amounts of fibre. The condition is characterized by sustained and marked decrease in rumen liquor pH ≤5.5 for at least three hours of each day. Rumen buffers and direct fed microbials (DFM) are dietary supplements that had been used in the management of SARA. Rumen liquor samples of dairy cows from the organized farms and households of Wayanad district which were at the risk of SARA were collected by rumenocentesis for rumen pH estimation. Blood samples were collected from cows positive for SARA and subjected to haemato-biochemical evaluations. Three different therapeutic protocols involving oral administration of sodium bicarbonate, Lithothamnium calcareum extracts as rumen buffers and a mixture Saccharomyces cerevisiae and Aspergillus oryzae as direct fed microbial were compared in SARA positive cows for 14 days and the efficacy of the treatment was evaluated by the improvement in rumen liquor pH after the treatment period. Rumen buffer like sodium bicarbonate was found to be more efficient than Lithothamnium calcareum and DFMs like Saccharomyces cerevisiae and Aspergillus oryzae mixture in improving the rumen pH values in cattle positive for SARA
