    Supporting Data mining of large databases by visual feedback queries

    In this paper, we describe a query system that provides visual relevance feedback in querying large databases. Our goal is to support the process of data mining by representing as many data items as possible on the display. By arranging and coloring the data items as pixels according to their relevance for the query, the user gets a visual impression of the resulting data set. Using an interactive query interface, the user may change the query dynamically and receives immediate feedback by the visual representation of the resulting data set. Furthermore, by using multiple windows for different parts of a complex query, the user gets visual feedback for each part of the query and, therefore, may easier understand the overall result. Our system allows to represent the largest amount of data that can be visualized on current display technology, provides valuable feedback in querying the database, and allows the user to find results which, otherwise, would remain hidden in the database

    Tropomyosin (Tm) is an evolutionarily conserved ?-helical coiled-coil protein, dimers of which form end-to-end polymers capable of associating with and stabilising actin-filaments and regulate myosin function. The fission yeast, Schizosaccharomyces pombe, possesses a single essential Tm, Cdc8, which can be acetylated on its amino terminal methionine to increase its affinity for actin and enhance its ability to regulate myosin function. We have designed and generated a number of novel Cdc8 mutant proteins with amino terminal substitutions to explore how stability of the Cdc8-polymer overlap region affects the regulatory function of this Tm. By correlating the stability of each protein, its propensity to form stable polymers, its ability to associate with actin and to regulate myosin, we have shown the stability of the amino terminal of the Cdc8 ?-helix is crucial for Tm function. In addition we have identified a novel Cdc8 mutant with increased amino-terminal stability, dimers of which are capable of forming Tm-polymers significantly longer than the wild-type protein. This protein had a reduced affinity for actin with respect to wild type, and was unable to regulate actomyosin interactions. The data presented here are consistent with acetylation providing a mechanism for modulating the formation and stability of Cdc8 polymers within the fission yeast cell. The data also provide evidence for a mechanism in which Tm dimers form end-to-end polymers on the actin-filament, consistent with a cooperative model for Tm binding to actin

    s- and d-wave superconductivity in a two-band model

    Superconductivity in strongly correlated systems is a remarkable phenomenon that attracts a huge interest. The study of this problem is relevant for materials as the high Tc oxides, pnictides and heavy fermions. In this work we study a realistic model that includes the relevant physics of superconductivity in the presence of strong Coulomb correlations. We consider a two-band model, since most of these correlated systems have electrons from at least two different atomic orbitals coexisting at their Fermi surface. The Coulomb repulsion is taken into account through a local repulsive interaction. Pairing is considered among quasi- particles in neighbouring sites and we allow for different symmetries of the order parameter. In order to deal with the strong local correlations, we use the well known slave boson approach that has proved very successful for this problem. Here we are interested in obtaining the zero temperature properties of the model, specifically its phase diagram and the existence and nature of superconducting quantum critical points. We show that these can arise by increasing the mixing between the two bands. Since this can be controlled by external pressure or doping, our results have a direct relation with experiments. We show that the superconductor-to-normal transition can be either to a metal, a correlated metal or to an insulator. Also we compare the relative stability of s and d-wave paired states for different regions of parameter space and investigate the BCS- BEC crossover in the two-band lattice model as function of the strength of the pairing interaction.Comment: 21 pages, 14 figure

    Deactivation of TEM-1 beta-Lactamase investigated by isothermal batch and non-isothermal continuous enzyme membrane reactor methods

    The thermal deactivation of TEM-1 ÎČ-lactamase was examined using two experimental techniques: a series of isothermal batch assays and a single, continuous, non-isothermal assay in an enzyme membrane reactor (EMR). The isothermal batch-mode technique was coupled with the three-state Equilibrium Model of enzyme deactivation, while the results of the EMR experiment were fitted to a four-state molten globule model . The two methods both led to the conclusions that the thermal deactivation of TEM-1 ÎČ -lactamase does not follow the Lumry-Eyring model and that the Teq of the enzyme (the point at which active and inactive states are present in equal amounts due to thermodynamic equilibrium) is at least 10 °C from the Tm (melting temperature), contrary to the idea that the true temperature optimum of a biocatalyst is necessarily close to the melting temperature

    The 13C-NMR spectra of gem quality and industrial diamonds show two resonances with the more intense resonance at high field. Two resonances are also shown in 13C-NMR spectra of various graphites; however, the low field resonance is of greater intensity than the high field resonance in the graphites. The resonances are very broad and they are assigned to graphite type (sp2) carbon and diamond type (sp3) carbon
