    Targeting effect of folate on cancer cell through curcumin carrier nano-system

    Folate receptor (FR) is well known for its overexpression on surface of various cancer cell lines, which is identical to normal tissue. Folic-based targeting drug delivery systems, therefore, are one of the most effective targeting carriers that effectively bind to FR up-regulated cancer cells. Curcumin was used both for labeling and chemotherapy. The materials were characterized and structurally confirmed by FT-IR spectra, fluorescent images and FE-SEM images. Bioassays were conducted on HeLa and HT29 cancer cell lines after 4 and 12 hours. Results show that folic acid significantly enhanced both targeting efficiency and internalization of curcumin to FR-expressing cancer cells

    Selective breeding of saline-tolerant striped catfish (Pangasianodon hypophthalmus) for sustainable catfish farming in climate vulnerable Mekong Delta, Vietnam

    peer reviewedStriped catfish (Pangasianodon hypophthalmus), a freshwater species cultured mainly in the Mekong Delta region in Southern Vietnam, is facing a significant challenge due to salinity intrusion as a result of climatic changes. Given these evolving environmental conditions, selecting new strains with a higher salinity tolerance could make production of striped catfish economically feasible in brackish environments. In this study, we carried out a selection program aimed at developing a striped catfish strain able to survive and grow fast in a saline environment. To implement the selection program, we first collected males and females from different provinces in the Mekong delta. We next performed a factorial cross of these breeders to produce half- and full-sib families. When fish reached fry stage (47 dph), we put them in a saline environment (10 ppt) and subsequently kept 50 % of the fastest-growing fish after 143 days post hatching (dph). We repeated this mass selection procedure after 237 dph and 340 dph. We maintained in parallel a randomly selected group in saline conditions and a group of fish reared in freshwater to serve as controls. After crossing the selected individuals, we performed several tests on the next generation of fish to evaluate the effectiveness of selection after one generation in saline conditions. Average direct responses to selection were 18.0 % for growth and 11.4 % for survival rate after one generation of selection. We estimated a moderate realized heritability (0.29) for body weight. The genetic gains obtained in our study for body weight and survival rate after one generation of selection under saline conditions suggest that selection can be effective to improve ability of striped catfish to cope with saline stress. We conclude that our selection program has succeeded in developing a productive strain of striped catfish with better tolerance to salinity. © 2022 The Author

    Multifunctional nanocarriers of Fe3O4@PLA-PEG/curcumin for MRI, magnetic hyperthermia and drug delivery

    Background: Despite medicinal advances, cancer is still a big problem requiring better diagnostic and treatment tools. Magnetic nanoparticle (MNP)-based nanosystems for multiple-purpose applications were developed for these unmet needs. Methods: This study fabricated novel trifunctional MNPs of Fe3O4@PLA-PEG for drug release, MRI and magnetic fluid hyperthermia. Result: The MNPs provided a significant loading of curcumin (∼11%) with controllable release ability, a high specific absorption rate of 82.2 W/g and significantly increased transverse relaxivity (r2 = 364.75 mM-1 s-1). The in vivo study confirmed that the MNPs enhanced MRI contrast in tumor observation and low-field magnetic fluid hyperthermia could effectively reduce the tumor size in mice bearing sarcoma 180. Conclusion: The nanocarrier has potential for drug release, cancer treatment monitoring and therapy.The authors are grateful for the financial support by AOARD under award FA2386-17-1-4042.     Age-time-specific transmission of hand-foot-and-mouth disease enterovirus serotypes in Vietnam: a catalytic model with maternal immunity

    Hand, foot and mouth disease (HFMD) is highly prevalent in the Asia Pacific region, particularly in Vietnam. To develop effective interventions and efficient vaccination programs, we inferred the age-time-specific transmission patterns of HFMD serotypes enterovirus A71 (EV-A71), coxsackievirus A6 (CV-A6), coxsackievirus A10 (CV-A10), coxsackievirus A16 (CV-A16) in Ho Chi Minh City, Vietnam from a case data collected during 2013–2018 and a serological survey data collected in 2015 and 2017. We proposed a catalytic model framework with good adaptability to incorporate maternal immunity using various mathematical functions. Our results indicate the high-level transmission of CV-A6 and CV-A10 which is not obvious in the case data, due to the variation of disease severity across serotypes. Our results provide statistical evidence supporting the strong association between severe illness and CV-A6 and EV-A71 infections. The HFMD dynamic pattern presents a cyclical pattern with large outbreaks followed by a decline in subsequent years. Additionally, we identify the age group with highest risk of infection as 1-2 years and emphasise the risk of future outbreaks as over 50% of children aged 6-7 years were estimated to be susceptible to CV-A16 and EV-A71. Our study highlights the importance of multivalent vaccines and active surveillance for different serotypes, supports early vaccination prior to 1 year old, and points out the potential utility for vaccinating children older than 5 years old in Vietnam

    The methanol extract of the soft coral Sarcophyton pauciplicatum afforded four sterols as (24S)-ergostane-3β,5α,6β,25-tetraol 25-monoacetate (1), (24S)-ergostane-3β,5α,6β,25-tetraol (2), (24S)-ergostane-1β,3β,5α,6β,25-pentaol 25-monoacetate (3), and (24S)-ergost-25-ene-1β,3β,5α,6β-tetraol (4) after subjecting it to various chromatographic experiments. The structures of isolated compounds were elucidated by 1D and 2D-NMR experiments and comparison of their NMR data with reported values. This is the first report of these compounds from                       S. pauciplicatum

    Molecular epidemiology of Japanese encephalitis in northern Vietnam, 1964?2011: genotype replacement

    Background: Japanese encephalitis virus (JEV) is an arthropod-borne virus causing serious public health issues in Asia. JEV consists of five genotypes and recent studies have shown the emergence of JEV genotype I (GI) and its replacement of genotype III (GIII). Using an archival JEV collection, we investigated the molecular evolution of JEV in Vietnam over the last 48 years (1964-2012) in humans, mosquitoes, and pigs, within the global context. Methods: The nine JEV isolates from humans, pigs, and mosquitoes sequenced in this study and 29 sequences available in GenBank were used to analyze the envelope (E) protein of the Vietnamese JEVs. A collection of 225 cerebrospinal fluid specimens from patients with suspected Japanese encephalitis (JE) was also tested and genotyped with real-time RT-PCR. Results: The 38 E genes identified with sequencing and nine Vietnamese JEV strains genotyped with real-time RT-PCR, belonging to two lineages, evolved in accordance with those in the rest of the world. The first GIII strain was detected in humans in Vietnam in 1964, and in mosquitoes in 1979, whereas GI strains were first detected in humans and mosquitoes in 1990 and 1994, respectively. After 2004, GI was the only genotype detected in Vietnam, demonstrating that the GIIII strains had been displaced by GI strains. Five haplotypes were identified in the Vietnamese JEVs, with SKSS predominant. The S123N and S123R substitutions in the E protein were already present in the Vietnamese JEVs. Conclusion: This study describes the long evolutionary history of JEV in Vietnam over 34 years, which correlates well with the global evolution of JEV. The Vietnamese GIII strains have been replaced by GI strains in mosquitoes, pigs, and humans. The predominant haplotypes of the Vietnamese strains support this genotype displacement in Vietnam. Further surveillance is required to confirm the disappearance of the GIII strains in nature and the emergence of new pathogens causing encephalitis in Vietnam, after the long-term use of JEV vaccines in that country
