    Several proteins from lily bulb for foodstuffs showed strong inhibitory activity toward bovine trypsin. A trypsin inhibitor (LTI-II-4) was purified from lily bulb by a method including column chromatographies on CM-Sepharose CL-6B and DEAE-Sepharose GL-6B and high-pressure liquid chromatography. Purified LTI-II-4 had a molecular weight of 21,000. The amino acid composition was characterized by high contents of glycine, aspartic acid and serine. The inhibitor contained 4 halfcystines in its constituent amino acids. The N-terminal 25 and the C-terminal 3 residues of LTI-II-4 were sequenced. The Inhibitor was stable in a wide pH range under 50℃ and was unstable at the higher temperatures than 80℃. The reactivesite amino acid of LTI-II-4 was assumed to be lysine. The inhibitor didn\u27t inhibit elastase and subtilisin. From these results, the inhibitor LTI-II-4 is assumed to belong to the Kunitz soybean trypsin inhibitor family

    IgE binding antigen of the major proteins in egg white, which is hen (H), quail (Q) and duck (D) was determined using the sera of patients with egg allergy. The major proteins of egg white were ovotransferrin (OT), ovoinhibitor (OI), ovalbumin (OA), ovomucoid (OM) and lysozyme (LY). The age of egg allergy patients is used this present studey were 3 years 4 months-old to 4 months-old (RAST 2~6). After SDS-PAGE analysis of egg whites, Western-blotting and enzyme-immunoassay was examined using the serum of 33 patoents with egg allergy. The positive blots of IgE binding proteins were ranked score 1~5 by positive color. The blots from score 2 to score 5 were positive of IgE binding. The positive percent of IgE blotting to hen OM was shown to have higher positive percent relatively to the other major proteins with the serum of 33 patients. And, on the sera of RAST 3 and RAST 4~6, ratio of IgE binding to hen OM indicated high positive percent. Therefore, from this study, these findings suppose that hen OM is the most important allergen of egg white proteins

    Influence of LINE-Assisted Provision of Information about Human Papillomavirus and Cervical Cancer Prevention on HPV Vaccine Intention: A Randomized Controlled Trial

    We conducted a prospective, randomized two-arm, parallel group, and open label trial to investigate whether the use of LINE would increase HPV vaccine intention among not completely vaccinated university students. In June 2020, we recruited students aged between 18 and 35 years from four universities in Japan. Among the 357 enrollees (female, 53%), 178 and 179 participants were randomized into the LINE and Mail groups, respectively. At baseline, within three years, vaccine intention was observed in 40% vs. 42% of participants, respectively. At the first intervention, which provided similar PDF leaflets about HPV vaccine and cervical cancer prevention, there was no significant difference in vaccine intention between the two groups. However, at the second intervention of LINE-assisted knowledge intervention for 5 days per week for 7 weeks, the LINE group had a higher proportion of vaccine intention than the no intervention group (66% vs. 44%, OR: 2.62, 95% confidence interval (CI): 1.59-4.35) in per-protocol analysis. The significance remained in the intention-to-treat analysis of multiply imputed datasets. Although LINE did not directly increase HPV vaccine intention compared to conventional posts, the LINE-assisted provision of information was effective in improving HPV vaccine intention among Japanese university and college students

    Cytotoxic T Lymphocytes Regenerated from iPS Cells Have Therapeutic Efficacy in a Patient-Derived Xenograft Solid Tumor Model

    Current adoptive T cell therapies conducted in an autologous setting are costly, time consuming, and depend on the quality of the patient's T cells. To address these issues, we developed a strategy in which cytotoxic T lymphocytes (CTLs) are regenerated from iPSCs that were originally derived from T cells and succeeded in regenerating CTLs specific for the WT1 antigen, which exhibited therapeutic efficacy in a xenograft model of leukemia. In this study, we extended our strategy to solid tumors. The regenerated WT1-specific CTLs had a strong therapeutic effect in orthotopic xenograft model using a renal cell carcinoma (RCC) cell line. To make our method more generally applicable, we developed an allogeneic approach by transducing HLA-haplotype homozygous iPSCs with WT1-specific TCR α/β genes that had been tested clinically. The regenerated CTLs antigen-specifically suppressed tumor growth in a patient-derived xenograft model of RCC, demonstrating the feasibility of our strategy against solid tumors

    An overview of the current status of the 8.2 m Subaru Telescope constructed and operated at Mauna Kea, Hawaii, by the National Astronomical Observatory of Japan is presented. The basic design concept and the verified performance of the telescope system are described. Also given are the status of the instrument package offered to the astronomical community, the status of operation, and some of the future plans. The status of the telescope reported in a number of SPIE papers as of the summer of 2002 are incorporated with some updates included as of 2004 February.     Characterization of autonomous Dart1 transposons belonging to the hAT superfamily in rice

    An endogenous 0.6-kb rice DNA transposon, nDart1-0, was found as an active nonautonomous element in a mutable virescent line, pyl-v, displaying leaf variegations. Here, we demonstrated that the active autonomous element aDart in pyl-v corresponds to Dart1-27 on chromosome 6 in Nipponbare, which carries no active aDart elements, and that aDart and Dart1-27 are identical in their sequences and chromosomal locations, indicating that Dart1-27 is epigenetically silenced in Nipponbare. The identification of aDart in pyl-v was first performed by map-based cloning and by detection of the accumulated transposase transcripts. Subsequently, various transposition activities of the cloned Dart1-27 element from Nipponbare were demonstrated in Arabidopsis. Dart1-27 in Arabidopsis was able to excise nDart1-0 and Dart1-27 from cloned sites, generating footprints, and to integrate into new sites, generating 8-bp target site duplications. In addition to Dart1-27, Nipponbare contains 37 putative autonomous Dart1 elements because their putative transposase genes carry no apparent nonsense or frameshift mutations. Of these, at least four elements were shown to become active aDart elements in transgenic Arabidopsis plants, even though considerable sequence divergence arose among their transposases. Thus, these four Dart1 elements and Dart1-27 in Nipponbare must be potential autonomous elements silenced epigenetically. The regulatory and evolutionary implications of the autonomous Dart1 elements and the development of an efficient transposon-tagging system in rice are discussed
