    Performen Reproduksi dan Produksi Susu Sapi Perah di Kecamatan Tegalombo Kabupaten Pacitan Provinsi Jawa Timur

    Kabupaten Pacitan merupakan salah satu daerah di Provinsi Jawa Timur yang akan dijadikan salah satu konsentrasi peternakan sapi perah di Jawa Timur, yang masih menyimpan potensi untuk pengembangannya, karena dukungan iklim dan topografi yang sesuai dengan habitat sapi perah Peranakan Frisien Holstein (PFH). Jenis sapi perah yang banyak dipelihara oleh peternak di Kabupaten Pacitan adalah jenis sapi Friesian Holstein yang memiliki produk utama susu. Penelitian mengenai produktivitas sapi perah FH ini dimaksudkan untuk mendapatkan gambaran potensi yang dimiliki oleh sapi perah yang dipelihara para peternak di daerah Pacitan. Hasil penelitian diperoleh performans sifat produksi susu yang meliputi rataan produksi susu setiap laktasi 4175,89 ± 980,33 kg/ekor/hari; lama laktasi 315,97 ± 25,17 hari; dan lama kering sebesar 65,53 ± 14,59 hari. Performans sifat reproduksi yang meliputi rataan kawin pertama setelah beranak 85,25 ± 19,64 hari; lama masa kosong 117,10 ± 30,52 hari; jumlah kawin perkebuntingan 2,10 ± 0,88 kali; lamanya bunting 284,19 ± 8,12 hari; dan selang beranak 390,54 ± 26,60 hari

    Motilitas dan Abnormalitas Spermatozoa X dan Y Kambing Lokal pada Berbagai Persentase Gradient Percoll Menggunakan Metode Swim Down

    The purpose of this study was to determine the quality of spermatozoa X and Y from semen of domestic goat which separated with various concentration of percoll. The metode of this study is experimental laboratory. The treatment consist of several concentration of percoll and diluter of percoll. R1 treated by 30% percoll : 70% diluter, R2 treated by 40% percoll : 70% diluter, R3 treated by 50% percoll : 70% diluter, and R4 treated by 60% percoll : 70% diluter. Experimental disign used in this experimental was RCBD (Randomize Complete Block Disain). Each treatment was replicated six times with the size of circle scrotum as the block. Result showed that the treatment R1 for X and Ybearing sperm, sperm motility, was 77,50 ± 5,00 and 70,00 ± 14,14, sperm abnormality was 10,50 ± 1,52 and 8,91 ± 1,31. The treatment R2 for X and Y-bearing sperm, sperm motility and sperm abnormality was 75,00 ± 5,77, and 75,00 ± 5,77, abnormality was 10,44 ± 1,09 and 11,4 ± 1,47. The treatment R3 for X and Y-bearing sperm, sperm motility was and sperm abnormality was 67,5 ± 5,00 and 62,50 ± 5,00, abnormality was 8,07 ± 1,40 and 9,37 ± 1,35. The treatment R4 for X and Y-bearing sperm, sperm motility, and sperm abnormality was 72,50 ± 9,57 and 60,00 ± 8,16, abnormality was 10,87 ± 2,84 and 9,83 ± 2,20. Sperm separation with the different concentration of percoll were hightly significant (P < 0.01) to identify spermatozoa X and Y chromosome, but there was no significantly different (P > 0.05) in the sperm motility and sperm abnormality. It is concluded that semen selected by variouse concentration of percoll and diluter increased the presentage of X and Y-bearing sperm. However motility and abnormality of X and Y-bearing sperm were not affected by the treatment

    Pengembangan Metode Identifikasi Kerusakan DNA Spermatozoa Ternak

    The success of artificial insemination is very much determined by the quality of spermatozoa. The detection or identification of damaged chromatin of spermatozoa DNA is very important to forsee the adverse clinical outcome. However, the method of identification is still depended on expensive imported kits. Therefore, the objective of this research was to developed an identification kit to determine the quality of livestock spermatozoa DNA chromatin.This study consist of three step. Step 1) Determination of low melting point agarose (LMP-agarose) concentration which is 0,6%, 0,7% and 0,8%. 2) Comparison of three lysis solution (LS) which is LS I (0.4M Tris, 0.8M DTT, 1% SDS, pH 7.5), LS II (0.4M Tris,2 M NaCl, 1% SDS , pH 7.5), and LS III (0.4M Tris-HCl, 2M NaCl, 1% SDS 0,05 M EDTA, pH 7.5). 3) Comparison different staining which is Eosin yellow and Methylene blue. The results showed that 0.6% LMP-agarose demonstrated the best concentration to “trapped the spermatozoa” compared for sheep and goats. whereas the three concentration of spermatozoa cows can not be used to trap spermatozoa cow. The best formulation to lysis the membrane was LS III (0.4M Tris -HCl, 2M NaCl, 1% SDS 0,05 M EDTA). The best staining was eosin yellow and methylene blue with 2:1 ratio

    Kejadian Fascioliasis pada Sapi Perah di Kecamatan Tegalombo, Kabupaten Pacitan

    Fascioliasis adalah penyakit cacing hati yang dapat menyerang sapi dan sangat merugikan, karena dapat menurunkan produktivitas. Peternakan sapi perah di Tegalombo, Pacitan merupakan peternakan rakyat yang pengelolaannya banyak melibatkan perempuan. Penelitian ini bertujuan mengidentifikasi fascioliasis pada sapi perah di kecamatan Tegalombo, kabupaten Pacitan. Sebanyak 50 ekor sapi perah digunakan sebagai sampel di dalam penelitian ini. Semua sapi diperiksa secara fisik, diambil fesesnya dan diperiksa adanya telur cacing Fasciola sp. Hasil pemeriksaan fisik sapi secara klinis menunjukkan bahwa semua sapi pada penelitian ini menunjukkan kondisi umum normal, nafsu makan baik, serta tidak ada abnormalitas yang signifikan pada semua bagian tubuh, sehingga dinyatakan sehat secara fisik. Hasil pemeriksaan sampel feses didapatkan adanya telur cacing Fasciola sp. pada 2 dari 50 ekor sapi (4%), sedangkan 48 ekor sapi lainnya (96 %) tidak ditemukan telur cacing. Kesimpulan dari peneltian ini adalah kejadian fascioliasis pada sapi perah di Tegalombo, kabupaten Pacitan rendah. Pemeriksaan secara periodik terhadap kemungkinan fascioliasis perlu dilakukan, sebagai langkah penanggulangan fascioliasis di daerah tersebut

    Investigation of Honeycomb Sandwich Panel Structure using Aluminum Alloy (AL6XN) Material under Blast Loading

    In this study, we focused on the large inelastic behavior of a sandwich panel made of two solid plates as a stiffener and a honeycomb core shell subjected to blast load. The loading scheme was carried out using an explosive charge bullet mounted at a standoff distance of 100 mm with three mass variations of trinitrotoluene: 1, 2, and 3 kg TNT. The numerical simulations performed using ABAQUS/CAE were validated with the experimental results of a previous study. The geometrical effects of the sandwich panel on intact and damaged models were also numerically investigated. The panel was designed using a square and hexagonal honeycomb core. The effect of honeycomb core height was also observed by modeling the core using three height variations: 31, 51, and 71 mm. The results showed that the hexagonal core was more resistant to blast loads than the square design. The core height parameter determines the energy absorption based on these results. The structural strength is also affected by the damage.     Can Farmers' Wives Adopt New Technology? Case: Milk Processing during the Covid-19 Pandemic in the Tahunan Village, Pacitan Regency, Indonesia

    This paper draws from our experience in assisting the midwives of milk farmers in learning how to process milk to raise their family income. We focus on 136 housewives of dairy farmers (all members of dairy farmer groups) in Tahunan village, Tegalombo sub-regency, Pacitan regency in Indonesia. There is only one collector in the area, PT DAS, sub-contracting from Nestle Indonesia. The problem happened when there is an over-supply, and the milk collector refuses to process the farmer's milk. For this reason, we have been assisting the dairy farm wives to process the milk and sell it to the local market since 2018. We start with the introduction of the plan to all dairy farm members (both the husband and the wives), then survey the availability of time the wives have. We focus on the wives because the husband is responsible for taking care of the cows. Also, we investigate the potential of local products (empon-empon, traditional roots such as ginger, turmeric) that can be processed together with the milk. The local product is chosen because of the familiarity of the potential customer with the flavor of the product. Introduction of the new technology to process milk at the beginning and the accompaniment are the keys to the acceptance of the activities

    Kepincangan Akibat Kuku Abnormal Sapi Perah di Kandang dengan Alas Karet dan Beton

    Kepincangan pada sapi perah yang dapat diebabkan kuku abnormal dapat mempengaruhi kesehatan dan produksi susu. Penelitian ini bertujuan membandingkan kasus kepincangan akibat kuku abnormal pada sapi perah yang dipelihara di kandang dengan alas karet dan beton. Penelitian ini menggunakan 104 ekor sapi perah dari 23 peternak, yang terdiri dari 72 ekor dipelihara dengan alas kandang karet dan 32 beton. Semua sapi diperiksa kukunya, kemampuan berdiri dan berjalan. Hasil penelitian menunjukkan bahwa prevalensi kasus kuku abnormal sebanyak 26 dari 105 ekor sapi (25%) yang terdiri dari 16/72 ekor (22,2%) pada kandang alas karet dan 10/32 ekor (31,3%) pada kandang alas beton. Kondisi kuku tersebut menyebabkan sebanyak 8 ekor sapi (30,8%) kesulitan berdiri atau kesulitan berjalan dan (69,2%) masih dapat berdiri dan berjalan dengan normal. Sebanyak 6/16 ekor (37,5%) sapi pada kandang alas karet dan 2/10 ekor (20%) sapi pada kandang alas beton menunjukkan kesulitan berdiri dan berjalan. Abnormalitas kuku pada penelitian ini kemungkinan disebabkan kuku tidak dipotong tepat waktu karena peternak kurang berpengalaman. Kesimpulan dari peneltian ini adalah prevalensi problem kuku abnormal adalah 25% yang didapatkan lebih banyak terjadi pada kandang alas beton. Kepincangan akibat kuku abnormal terjadi pada 30,8% sapi perah kuku abnormal dan kejadian didapatkan lebih banyak pada sapi yang dipelihara di kandang dengan alas karet

Education for Sustainability as a teaching strategy for resolving community challenges through formal, informal, or non-formal education is expected to benefit from various community service best practices by academics, researchers, and students. The 3rd ICCEESD has “Strengthening Education for Sustainability Towards Better Community Engagement” as its theme this year.
