    Study of the Local Interstellar Medium using Pulsar Scintillation

    We present here the results from an extensive scintillation study of twenty pulsars in the dispersion measure (DM) range 3 - 35 pc cm^-3 carried out using the Ooty Radio Telescope, to investigate the distribution of ionized material in the local interstellar medium (LISM). Our analysis reveals several anomalies in the scattering strength, which suggest that the distribution of scattering material in the Solar neighborhood is not uniform. Our model suggests the presence of a low density bubble surrounded by a shell of much higher density fluctuations. We are able to put some constraints on geometrical and scattering properties of such a structure, and find it to be morphologically similar to the Local Bubble known from other studies.Comment: 5 pages, 3 figure

    Domain walls between gauge theories

    Noncommutative U(N) gauge theories at different N may be often thought of as different sectors of a single theory: the U(1) theory possesses a sequence of vacua labeled by an integer parameter N, and the theory in the vicinity of the N-th vacuum coincides with the U(N) noncommutative gauge theory. We construct noncommutative domain walls on fuzzy cylinder, separating vacua with different gauge theories. These domain walls are solutions of BPS equations in gauge theory with an extra term stabilizing the radius of the cylinder. We study properties of the domain walls using adjoint scalar and fundamental fermion fields as probes. We show that the regions on different sides of the wall are not disjoint even in the low energy regime -- there are modes penetrating from one region to the other. We find that the wall supports a chiral fermion zero mode. Also, we study non-BPS solution representing a wall and an antiwall, and show that this solution is unstable. We suggest that the domain walls emerge as solutions of matrix model in large class of pp-wave backgrounds with inhomogeneous field strength. In the M-theory language, the domain walls have an interpretation of a stack of branes of fingerstall shape inserted into a stack of cylindrical branes.Comment: Final version; minor corrections; to appear in Nucl.Phys.

    Observational constraints on the neutron star mass distribution

    Radio observations of neutron star binary pulsar systems have constrained strongly the masses of eight neutron stars. Assuming neutron star masses are uniformly distributed between lower and upper bounds mlm_l and mum_u, the observations determine with 95\% confidence that 1.01<ml/M⊙<1.341.01 < m_l/\text{M}_\odot < 1.34 and 1.43<mu/M⊙<1.641.43 < m_u/\text{M}_\odot < 1.64. These limits give observational support to neutron star formation scenarios that suggest that masses should fall predominantly in the range 1.3<m/M⊙<1.61.3<m/\text{M}_\odot<1.6, and will also be important in the interpretation of binary inspiral observations by the Laser Interferometer Gravitational-wave Observatory.Comment: Postscript, 4 pages, NU-GR-

    Preventing empirical antibiotic treatment failure in migrant populations: screening by infection risk, not ethnic background

    Multidrug-resistant organisms (MDROs) are a major international health threat. In many low and middle-income countries poorly regulated antibiotic use, limited surveillance, and inadequate sanitation give rise to high rates of antibiotic resistance. A resulting reliance on last-line antibiotic options further contributes to the emergence of MDROs. The potential for these pathogens to spread across international borders is a matter of considerable concern. However, this problem is commonly framed as primarily a threat to the health security of countries where resistance is not yet endemic. In fact, it is little acknowledged that those at greatest risk from antibiotic treatment failure are individuals who move from regions of high MDRO prevalence to settings where standard empirical treatment options remain largely effective. In this perspective, we highlight the poor treatment outcomes for disseminated bacterial infections in individuals who have moved from settings in which MDROs are common to those where MDROs are currently less common. We discuss MDRO screening strategies that could avoid stigmatizing vulnerable populations by focusing on future risk of disseminated infection, rather than past risk of acquisition. In practical terms, this means screening individuals before childbirth, immunosuppressive treatments, major surgery, or other events associated with disseminated infection risk, rather than prioritizing screening for individuals from regions with high carriage rates. We argue that such measures would reduce antibiotic treatment failure and improve outcomes while protecting migrant populations from the divisive consequences of targeted screening programs.Steven L.     The bacterial biome of ticks and their wildlife hosts at the urban–wildland interface

    Advances in sequencing technologies have revealed the complex and diverse microbial communities present in ticks (Ixodida). As obligate blood-feeding arthropods, ticks are responsible for a number of infectious diseases that can affect humans, livestock, domestic animals and wildlife. While cases of human tick-borne diseases continue to increase in the northern hemisphere, there has been relatively little recognition of zoonotic tick-borne pathogens in Australia. Over the past 5 years, studies using high-throughput sequencing technologies have shown that Australian ticks harbour unique and diverse bacterial communities. In the present study, free-ranging wildlife (n=203), representing ten mammal species, were sampled from urban and peri-urban areas in New South Wales (NSW), Queensland (QLD) and Western Australia (WA). Bacterial metabarcoding targeting the 16S rRNA locus was used to characterize the microbiomes of three sample types collected from wildlife: blood, ticks and tissue samples. Further sequence information was obtained for selected taxa of interest. Six tick species were identified from wildlife: Amblyomma triguttatum, Ixodes antechini, Ixodes australiensis, Ixodes holocyclus, Ixodes tasmani and Ixodes trichosuri. Bacterial 16S rRNA metabarcoding was performed on 536 samples and 65 controls, generating over 100 million sequences. Alpha diversity was significantly different between the three sample types, with tissue samples displaying the highest alpha diversity (P<0.001). Proteobacteria was the most abundant taxon identified across all sample types (37.3 %). Beta diversity analysis and ordination revealed little overlap between the three sample types (P<0.001). Taxa of interest included Anaplasmataceae, Bartonella, Borrelia, Coxiellaceae, Francisella, Midichloria, Mycoplasma and Rickettsia. Anaplasmataceae bacteria were detected in 17.7% (95/536) of samples and included Anaplasma, Ehrlichia and Neoehrlichia species. In samples from NSW, ‘Ca. Neoehrlichia australis’, ‘Ca. Neoehrlichia arcana’, Neoehrlichia sp. and Ehrlichia sp. were identified. A putative novel Ehrlichia sp. was identified from WA and Anaplasma platys was identified from QLD. Nine rodent tissue samples were positive for a novel Borrelia sp. that formed a phylogenetically distinct clade separate from the Lyme Borrelia and relapsing fever groups. This novel clade included recently identified rodent-associated Borrelia genotypes, which were described from Spain and North America. Bartonella was identified in 12.9% (69/536) of samples. Over half of these positive samples were obtained from black rats (Rattus rattus), and the dominant bacterial species identified were Bartonella coopersplainsensis and Bartonella queenslandensis. The results from the present study show the value of using unbiased high-throughput sequencing applied to samples collected from wildlife. In addition to understanding the sylvatic cycle of known vector-associated pathogens, surveillance work is important to ensure preparedness for potential zoonotic spillover events

    Absence of plastin 1 causes abnormal maintenance of hair cell stereocilia and a moderate form of hearing loss in mice

    Hearing relies on the mechanosensory inner and outer hair cells (OHCs) of the organ of Corti, which convert mechanical deflections of their actin-rich stereociliary bundles into electrochemical signals. Several actin-associated proteins are essential for stereocilia formation and maintenance, and their absence leads to deafness. One of the most abundant actin-bundling proteins of stereocilia is plastin 1, but its function has never been directly assessed. Here, we found that plastin 1 knock-out (Pls1 KO) mice have a moderate and progressive form of hearing loss across all frequencies. Auditory hair cells developed normally in Pls1 KO, but in young adult animals, the stereocilia of inner hair cells were reduced in width and length. The stereocilia of OHCs were comparatively less affected; however, they also showed signs of degeneration in ageing mice. The hair bundle stiffness and the acquisition of the electrophysiological properties of hair cells were unaffected by the absence of plastin 1, except for a significant change in the adaptation properties, but not the size of the mechanoelectrical transducer currents. These results show that in contrast to other actin-bundling proteins such as espin, harmonin or Eps8, plastin 1 is dispensable for the initial formation of stereocilia. However, the progressive hearing loss and morphological defects of hair cells in adult Pls1 KO mice point at a specific role for plastin 1 in the preservation of adult stereocilia and optimal hearing. Hence, mutations in the human PLS1 gene may be associated with relatively mild and progressive forms of hearing loss

    A Nonparametric Method for the Derivation of α/ÎČ Ratios from the Effect of Fractionated Irradiations

    Multifractionation isoeffect data are commonly analysed under the assumption that cell survival determines the observed tissue or tumour response, and that it follows a linear-quadratic dose dependence. The analysis is employed to derive the α/ÎČ ratios of the linear-quadratic dose dependence, and different methods have been developed for this purpose. A common method uses the so-called Fe plot. A more complex but also more rigorous method has been introduced by Lam et al. (1979). Their method, which is based on numerical optimization procedures, is generalized and somewhat simplified in the present study. Tumour-regrowth data are used to explain the nonparametric procedure which provides α/ÎČ ratios without the need to postulate analytical expressions for the relationship between cell survival and regrowth delay

    The Laser Interferometer Space Antenna (LISA) mission will use advanced technologies to achieve its science goals: the direct detection of gravitational waves, the observation of signals from compact (small and dense) stars as they spiral into black holes, the study of the role of massive black holes in galaxy evolution, the search for gravitational wave emission from the early Universe. The gravitational red-shift, the advance of the perihelion of Mercury, deflection of light and the time delay of radar signals are the classical tests in the first order of General Relativity (GR). However, LISA can possibly test Einstein's theories in the second order and perhaps, it will show some particular feature of non-linearity of gravitational interaction. In the present work we are seeking a method to construct theoretical templates that limit in the first order the tensorial structure of some metric fields, thus the non-linear terms are given by exponential functions of gravitational strength.     Impact of long-term erythromycin therapy on the oropharyngeal microbiome and resistance gene reservoir in non-cystic fibrosis bronchiectasis

    Published 18 April 2018Long-term macrolide therapy reduces rates of pulmonary exacerbation in bronchiectasis. However, little is known about the potential for macrolide therapy to alter the composition and function of the oropharyngeal commensal microbiota or to increase the carriage of transmissible antimicrobial resistance. We assessed the effect of long-term erythromycin on oropharyngeal microbiota composition and the carriage of transmissible macrolide resistance genes in 84 adults with bronchiectasis, enrolled in the Bronchiectasis and Low-dose Erythromycin Study (BLESS) 48-week placebo-controlled trial of twice-daily erythromycin ethylsuccinate (400 mg). Oropharyngeal microbiota composition and macrolide resistance gene carriage were determined by 16S rRNA gene amplicon sequencing and quantitative PCR, respectively. Long-term erythromycin treatment was associated with a significant increase in the relative abundance of oropharyngeal Haemophilus parainfluenzae (P = 0.041) and with significant decreases in the relative abundances of Streptococcus pseudopneumoniae (P = 0.024) and Actinomyces odontolyticus (P = 0.027). Validation of the sequencing results by quantitative PCR confirmed a significant decrease in the abundance of Actinomyces spp. (P = 0.046). Erythromycin treatment did not result in a significant increase in the number of subjects who carried erm(A), erm(B), erm(C), erm(F), mef(A/E), and msrA macrolide resistance genes. However, the abundance of erm(B) and mef(A/E) gene copies within carriers who had received erythromycin increased significantly (P < 0.05). Our findings indicate that changes in oropharyngeal microbiota composition resulting from long-term erythromycin treatment are modest and are limited to a discrete group of taxa. Associated increases in levels of transmissible antibiotic resistance genes within the oropharyngeal microbiota highlight the potential for this microbial system to act as a reservoir for resistance.IMPORTANCE Recent demonstrations that long-term macrolide therapy can prevent exacerbations in chronic airways diseases have led to a dramatic increase in their use. However, little is known about the wider, potentially adverse impacts of these treatments. Substantial disruption of the upper airway commensal microbiota might reduce its contribution to host defense and local immune regulation, while increases in macrolide resistance carriage would represent a serious public health concern. Using samples from a randomized controlled trial, we show that low-dose erythromycin given over 48 weeks influences the composition of the oropharyngeal commensal microbiota. We report that macrolide therapy is associated with significant changes in the relative abundances of members of the Actinomyces genus and with significant increases in the carriage of transmissible macrolide resistance. Determining the clinical significance of these changes, relative to treatment benefit, now represents a research priority.Jocelyn M. Choo, Guy C. J.
