Principais métodos terapêuticos para peixes em cultivo.

    Get PDF

Macrophage-derived neutrophil chemotactic factor is involved in the neutrophil recruitment inhibitory activity present in the supernatants of LPS-stimulated macrophages

In a previous study, we demonstrated the presence of a neutrophil recruitment inhibitory factor (NRIF) in the supernatants of LPS-stimulated macrophages. Recently, the purification of a 54 kDa protein, identified as the macrophage-derived neutrophil chemotactic factor (MNCF) was reported. Since NRIF and MNCF are obtained under the same conditions, and, since the intravenous administration of TNF-α and IL-8 inhibits neutrophil migration, we have investigated whether MNCF could be responsible for this inhibitory activity. After affinity chromatography of the macrophage supernatants on a D-galactose column, the inhibitory activity was recovered in both the unbound (D-gal−) and bound (D-gal+) fractions, with MNCF being found in the D-gal+ fraction. Further gel filtration of the latter on Superdex 75 yielded a single peak containing both activities. In a cytotoxicity assay, most of the TNF found in the crude supernatants was recovered in the D-gal− fraction. Furthermore, the incubation of the D-gal− fraction with anti-TNF-α plus anti-IL-8 antisera partially prevents its inhibitory effect on neutrophil migration, but had no effect on the D-gal+ activity. Overall, these results suggest that the D-gal− inhibitory effect is partially mediated by TNF-α and IL-8, and that MNCF accounts for the inhibition of neutrophil migration in vivo by the D-gal+ fraction

Immunostimulants in farmed fish diets has been considered an effective approach to health management. This study aimed to assess antioxidant effects and resistance against pathogens of Colossoma macropomum (tambaqui) fed essential oil (EO) of M. piperita at 0%, 0.5%, 1.0% and 1.5% kg−1 commercial feed during 30 days. Tambaqui were challenged with Aeromonas hydrophila and did not present clinical signs of alterations. Higher hepatic catalase activity was observed in fish fed 1.0% of M. piperita EO, compared with 1.5%. Renal superoxide dismutase and glutathione peroxidase decreased in the fish fed M. piperita EO. Hepatic and renal lipid hydroperoxide increased among fish fed diets with 1.0 and 1.5% of EO. Tambaqui fed 1.0% of M. piperita EO showed respiratory activity increase, compared with 0.5%. Monocytes and PAS-GL were abundant in fish fed higher EO concentrations. Reduction in monogenoideans abundance was observed in fish fed 0.5 and 1.0% of EO. Although anti-helminth effects were observed, M. piperita EO in tambaqui diet did not promote leukocytes and lysozyme activity increases at the concentrations assessed. It can be suggested that other concentrations should be tested in future studies, as the combined use with other products, such as immunostimulants and probiotics

Hematological parameters of three freshwater stingray species (Chondrichthyes: Potamotrygonidae) in the middle Rio Negro, Amazonas state.

This paper aimed to study and compare the hematology of newborns, young, subadults, adult males, adult females and pregnant females of Potamotrygon wallacei (cururu stingray), Potamotrygon motoro and Paratrygon aiereba. Newborn cururu stingrays had lower red blood parameters than those of other development stages. Thrombograms and leukograms showed a conservative pattern between development stage, sexual dimorphism and pregnancy. In P. motoro and P. aiereba, variables relating to red blood parameters, biochemistry and leukograms showed little variation between the species' biological characteristics, thus showing that these variables are not good criteria for differentiating them within the same species. In conclusion, the development stage is an important factor for differentiating hematological properties in the cururu stingray, while this has not been observed in P. motoro and P. aiereba stingrays
