    Reproductive Organ of Male Sulawesi Giant Rat (Paruromys dominator)

    Abstract Indonesia has a biodiversity in fauna and one of them is the rat. The methodology to identify animal character is mostly based on morphological structure, the example of body lenght and hair color. However, reproductive organs of every species are diferent in location, shape, color and weight. Paruromys dominator is one of rats species in Sulawesi. Although, many informations about the rat's morphology is already provided , but no data about reproductive organ reported yet. Knowing, identifying and understanding the reproductive organs in that species must be done by sampling reproductive organs and measure all of them,. All of the reproductive organs were then collected for anatomical sample and processed for histological staining and were analyzed with light microscope and SEM. Based on the present study, Paruromys dominator has reproductive organs which are like other murids, namely seminal vesicle, testis, prostat glands, Cowper's glans and glans penis. There is no diference about histological stucture of rat's testis with other rats species, but based on the result of SEM, the penis has many penile spines. The sperm head is hook shape, it is like Rattus norvegicus. The average of head length and tail lenght of sperm are 1,28±0,101 μm and 12,95±1,139 μm, respectively.

    Kajian terhadap profil hormon-hormon steroid merupakan kunci penting dalam upaya memahami aspek fisiologis satwa. Beberapa dekade terakhir telah dikembangkan metode alternatif untuk mengetahui profil hormon steroid, yaitu melalui pengukuran metabolit hormon steroid yang diekskresikan melalui ekskreta tubuh seperti feses. Metode tersebut dikenal sebagai metode non-invasif. Metode ini memungkinkan pengumpulan sampel secara terus menerus dalam jangka panjang dengan meminimalisasi gangguan terutama pada satwa liar. Kajian terhadap profil metabolit hormon steroid yang terukur dapat diaplikasikan antara lain untuk mengetahui status reproduksi, penentuan jenis kelamin, studi perilaku hingga monitoring tingkat stres satwa. Berbagai kajian dengan memanfaatkan metabolit hormon steroid telah berhasil diaplikasikan pada berbagai taksa vertebrata.     MEASUREMENT OF SERUM TESTOSTERONE IN KACANG GOAT BYUSING ENZYME-LINKED IMMUNOSORBENT ASSAY (ELISA) TECHNIQUE: THE IMPORTANCE OF KIT VALIDATION (Pengukuran Testosteron Serum Kambing Kacang dengan Teknik Enzyme-Linked Immunosorbent Assay (ELISA): Pentingnya Validasi Kit)

    This study was conducted to validate a commercial testosterone enzyme-linked immunosorbent assay (ELISA) kits (DRG EIA-1559) inanalytic and biological manner for measuring serum testosterone concentrations in kacang goats. This study used 18 healthy kacang goats, six bucks (2 years), six kids (6 months), and six does (2 years). Blood samples were collected from jugular vein and prepared as serum. Two validation tests were performed, an analytical validation comprises a parallelism, accuracy, precision and sensitivity and a biological validation by comparing testosterone concentration from bucks, kids, and does. Testosterone concentrations were measured using ELISA technique. Data of analytical validation were analyzed descriptively and test of equality of slope was performed to see the parallelism between samples and standard curves. Analysis of variance (ANOVA) was used for biological validation data. Results of parallelism showed that sample curve was parallel to the standard curve. Accuracy, precision (% CV of intra-and inter-assay) and sensitivity of the assay were 99.65±4.27%, 10%, 15% and 0.083 ng/ml, respectively. Results of biological validation showed that the assay used were accurately measured testosterone which testosterone concentrations in bucks were significantly higher compared to kids and does (P0.05). In conclusion, a commercial testosterone ELISA kits (DRG EIA-1559) is a reliable assay for measuring serum testosterone concentration in kacang goats.     Genus Tragulus in Indonesia is consisted of three species, namely Tragulus javanicus, T. napu, and T.kanchil. These three species are distributed in Kalimantan (Borneo), Java, Sumatra and Lesser Sundaislands. Taxonomically, based on morphological characteristic,T. javanicus is the same species as T. kanchil,while T. napu have differences in body size compared to the previous species. The status of genus Tragulusin Indonesia are protected by the law and categorized as least concern for T. napu and data deficient for T.javanicus in the IUCN Redlist. The aims of this study was to look at genetic variation among species andkinship based on mitochondrial DNA gene genus Tragulus. Genes used in this study was 12S ribosomalRNA with 300 bp long, using eight samples orginated from Singkep Islands (four individuals), MountHalimun, West Java (three individuals) and East Kalimantan (one individual). As a comparisons, publishednucleotide sequence of each species from GeneBank database were used. The results showed that thegenetic distances (d) between individuals is high d = 0.322 ± 0.023 with the nucleotide diversity (ð) =0.2402 which indicates that samples was consisted of samples from different species. Phylogenetic treebased on the nucleotide analysis indicatedthat these samples composed of three species namely T. javanicus,T. kanchil and T. napu. Samples from Mt. Halimun and East Kalimantan are clustered in one group,namely T. kanchil (Borneo) and T. javanicus (Mt. Halimun). Two samples from Singkep are clustered indifferent groups with the possibility as T. napu

    Antlers are deer’s bony organ that follows a cycle of growing, hardening, casting and regroupingwithin a certain period. The effect of consuming velvet antler from temperate origin has beenknown scientifically to have positive effect for rheumatism and metabolic disorder sickness therapy.However, the role of velvet antler originated from tropical deer has not yet been explored. Thisstudy aimed to assess the potential of the velvet antler of sambar deer (Rusa unicolor) which wasexperimentally fed to laboratory rats. The assessment was made based on the animals growth rate(i.e. femur length, weight of testicle, body eight) and physical endurance (i.e. swimming test).Laboratory rats at 21 days old were allocated into four different groups and each group consisted offive rats were fed with powder of soft and hard parts of velvet antler at dose of 0, 1, 2, and 3 g/kgbody weight, respectively. Animals were examined for eight weeks the body weight was examinedweekly and the dose at velvet antler supplement was adjusted accordingly. At the end of the studythe rat were put on endurance swimming test and then euthanized, for measurement of femurbone length and weight of testis. The results showed that there were no differences in the bodyweight. However at dose of 2 g soft part/kg BW indicating a consistently higher live weight gainsacross the observation time. Testis weight showed no significant differences between the treatments,but the length of femur bone showed a significant effect (p <0.05) with the doses level, with thehighest score being at 3 g hard part / kg BW. Physical endurance showed a significant effect (p<0.05) with the doses level, with the level of 1 g soft part/kg BW gave the best performance

    Maintenance of wildlife outside its original habitat is usually for the purposes of the exhibition in zoos, for research and captive breeding effort, especially for endangered species which eventually return to nature. The adaptability of the animals outside their original habitat can be seen from their health and their ability to reproduce in ex situ (captive) environment. One of the parameter that can be used to determine a health an animal is by observing their blood parameters and compared it with normal reference, however for many wild animals reference data for normal blood parameters are limited or not available. This study was conducted to determine a normal reference of blood parameters of slow lories that are kept in captive breeding condition, consisted of routine hematology and blood chemistry. This study used five animals consisting of four of N. coucang (1 male and three female) and one female N. javanicus. The results of the analysis showed that the value of hemoglobin, MCV, PDW, MPC, and erythrocytes (RBC) were higher on the male slow lories, while leukocytes (WBC), hematocrit, and MCHC were higher on the female. There were no significant differences between blood parameters of N. coucang and N. javanicus
