Povezanost između radiografskih nalaza, magnetske rezonancije i histopatoloških nalaza kod pokusne rupture prednje križne sveze u kunića.
Experimental osteoarthritis (OA) was induced in the knee joints of rabbits and the trend of changes were compared by radiography, Magnetic Resonance Imaging (MRI) and histopathology. Twenty rabbits were randomly divided into two equal groups based on short (30 days) and long-term (180 days) follow ups. In half of the animals in each group (n = 5) OA was induced by sectioning the cranial cruciate ligament and in the other half, only arthrotomy was performed as a sham operation. Radiography and MRI were carried out on days 0 and 30 in the group of short term studies, and on days 0, 90 and 180 in the other group. Histopathological examinations were performed on day 30 in the short-term group after the animals had been sacrificed and in the other group on day 180. The slope of changes over the course of the study between all 3 methods and the grade of changes, were both highest in histopathology, and then in MRI and radiology respectively. The slope of changes was 0.01 for histopathology, 0.009 for MRI and 0.004 for radiology. The ratios of slopes, when compare to each other, were as follows: His./MRI = 1.1, His./Rad. = 2.5, MRI/Rad. = 2.2. Comparison of MRI with radiology revealed that radiology would not show signs of OA when the MRI grade is less than a grade of 0.27. Comparing both imaging techniques with histopathology showed that whenever the histopathological grade was below 0.22, radiology would not show signs of OA involvement, while MRI was capable of showing signs of OA involvement whenever it was more than 0.018 on histopathological grade.Nakon što je u kunića pokusno izazvan osteoartritis koljenog zgloba, trend promjena promatran je pomoću radiografije, magnetske rezonancije (MRI) i patohistoloških nalaza. Dvadeset kunića bilo je metodom slučajnog odabira podijeljeno u dvije jednake skupine od kojih je jedna promatrana tijekom kratkotrajnog (30 dana), a druga tijekom dugotrajnog (180 dana) razdoblja. U polovice životinja iz svake skupine (n = 5) osteoartroza je bila uzrokovana sekcijom prednje križne sveze, a kod druge polovice primijenjena je samo artrotomija kao lažna operacija. Radiografija i magnetska rezonancija obavljene su 0-ti i 30. dan tijekom kratkotrajnog promatranja te 0-ti, 90. i 180. dan kod dugotrajnog promatranja. Histopatološke pretrage obavljene su 30. dan kod kratkotrajnog praćenja nakon što su životinje bile usmrćene, a kod dugotrajnog 180. dan. Slabljenje promjena tijekom promatranja svima trima metodama kao i stupanj promjena pokazali su se najvećima kod histopatoloških nalaza, zatim kod magnetske rezonancije te nakon toga kod radioloških snimki. Slabljenje promjena bilo je 0,01 za histopatološke nalaze, 0,009 za magnetsku rezonanciju i 0,004 za radiološke snimke. Kad se uzmu u obzir međusobni odnosi slabljenja su bila sljedeća: His./MRI = 1,1, His./Rad. = 2,5, MRI/Rad. = 2,2. Usporedba magnetske rezonancije i radiologije potvrdila je da radiologija neće pokazati znakove osteoartritisa kada je stupanj magnetske rezonance niži od 0,27. Usporedbom obje tehnike snimanja s histopatološkim nalazima utvrđeno je da radiologija neće pokazati znakove osteoartritisa kad je stupanj histopatoloških nalaza ispod 0,22. Primjenom magnentske rezonancije moguće je utvrditi znakove osteoartritisa svaki put kada je njezin stupanj veći od 0,018 u odnosu na stupanj histopatoloških nalaza
Benign prostatic hyperplasia treatment using plasmonic nanoparticles irradiated by laser in a rat model
Objective: In the current study we have stimulated the efficacy of plasmonic nanoparticles (NPs) by laser hyperthermia to achieve a less invasive method for tumor photothermal therapy of benign prostatic hyperplasia (BPH). Methods: The levels of apoptosis on induced BPH in rats were assessed after treatment and revealed and recorded by various assayed. Moreover, the expression of caspases was considered to demonstrate the apoptotic pathways due to laser induced plasmonic NPs. Results: In the Laser + NPs group prostate size of induced BPH decreased. Laser + NPs also decreased prostate specific antigen in comparison with the BPH groups. Furthermore, Laser + NPs attenuated BPH histopathologic indices in the rats. Laser + NPs induced apoptosis in prostatic epithelial cells via caspase-1 pathway. Conclusions: Altogether, the approach and findings from this study can be applied to introduce the laser irritated NPs method as a novel and less invasive therapy for patients suffering from BP
Anti-inflammatory and healing action of oral gel containing borneol monoterpene in chemotherapy-induced mucositis in rats ( Rattus norvegicus )
ABSTRACT The aim of this study was to investigate the effect of gels containing the monoterpene borneol in induced oral mucositis using an animal model. Gels were prepared with borneol at 1.2% and 2.4% (w/w). Oral mucositis was induced by administration of three doses of 5-fluorouracil (30 mg/kg, i.p.) and injury with acetic acid (50%, v/v) soaked in filter paper applied to right cheek mucosa for 60s. Four subgroups comprising 12 animals each were formed. Six animals from each group were sacrificed at days seven and fourteen after oral mucositis induction. Mucous samples were processed and stained with hematoxylin-eosin and Masson’s Trichrome. The semiquantitative evaluation involved observation of inflammatory parameters. ImageJ® software was used in the quantitative evaluation. For statistical analyses, Two-way ANOVA, followed by Tukey’s post-test (p <0.05), were employed. Borneol 2.4% gel proved effective in the treatment of oral mucositis with statistically significant differences between groups for angiogenesis control, inflammatory cell count reduction and percentage neoformed collagen increase. The confirmation of anti-inflammatory and healing action of borneol in oral mucositis in rats renders it a good marker for predicting this activity for plant extracts rich in this substance
Antioxidant effects of betaine against Indomethacin-induced gastric damage in rats
Introduction: Betaine (trimethyl glycine) is known as methyl group donor and antioxidant in previous reports. The aim of this study was to assess the antioxidant effects of betaine in Indomethacin-induced gastric damages.
Methods: Thirty-two adult male Sprague–Dawley rats in an experimental study were divided into four equal groups as follow: Control, Indomethacin, Betaine-indomethacin and Ascorbic acid-indomethacin. Control and indomethacin groups received normal saline and betaine and ascorbic acid-pretreated rats were administrated betaine (1.5% of the total diet) and ascorbic acid (50 mg/kg body weight) for 15 consecutive days, respectively. After 24 h fasting, all of the groups received indomethacin (48 mg/kg body weight) and control group received distilled water.
Results: Indomethacin administration increased gastric ulcer occurrence (%) in comparison with control group and betaine pretreatment significantly decreased ulcer occurrence (%) when compared to the other groups (P=0.0017). Gastric wall glutathione peroxidase (GPx) activity was significantly lower in indomethacin group in comparison with the other groups (P=0.0012) while, betaine and ascorbic acid pretreatment increased GPx activity in comparison with indomethacin group (P=0.0012). Catalase activity was significantly higher in betaine-pretreated rats in comparison with indomethacin and ascorbic acid-indomethacin groups (P=0.0015). Lipid peroxidation significantly decreased in betaine and ascorbic acid pretreated groups (P=0.0013).
Conclusion: These results showed beneficial antioxidant effects of betaine against gastric damages induced by indomethacin in rats
Spilt bile and gallstones effect during laparoscopic cholecystectomy: an experimental study for adhesion formation
Laparoscopic cholecystectomy (LC) is recognized as the standard treatment for symptomatic gallstone disease. In comparison with open techniques, LC results in a higher rate of iatrogenic biliary tract damage and spillage of bile and stones. The present study was designed to compare intraperitoneal adhesion formation following LC due to the bile and stone spillage. In this randomized experimental study, 15 dogs were randomly allocated to three groups. Each group contained five dogs. The control group underwent LC, while in the group of bile and stone spillage (BS), 5 cc of a mixture of dog bile and a sterile ground human gallstone was placed in the bed of the gallbladder following LC. The bile spillage (B) group underwent LC, followed by adding 5 cc of the bile in the bed of the gallbladder. Postsurgical adhesion formation was measured, using Nair and Zühlke classifications for inflammation, fibrosis, and neovascularization. Adhesion formation was noted in one dog from the control and B groups, whereas all animals in group 2 were found to develop high-grade adhesion. Bile cultures were negative for the control group and group B with adhesion. Based on the findings, bile spillage, as well as gallbladder stone spillage, is related to a significant increase in adhesion formation after LC. Comparing the healing properties of intra-articular injection of human dental pulp stem cells and cell-free-secretome on induced knee osteoarthritis in male rats
Objective: Osteoarthritis (OA) is a common and painful joint disease with multifactorial causes. Stem cells, due to their high ability to reproduce and differentiate, have created a new horizon in tissue engineering of cartilage and bone. Secretions are one of the new therapies that can be used with stem cells or separately. This study aimed to compare the healing effects of human dental pulp stem cells, cell-free secretome, and human dental pulp mesenchymal stem cells with secretome in the induced OA in male rats. Methods: Dental pulp mesenchymal stem cells were isolated and prepared from human dental pulp. The collagenase type II was injected into the knee of twenty-five male Sprague-Dawley rats, and after 10 weeks, OA was confirmed. Rats were divided into five groups (n = 5): 1) Human dental pulp stem cells plus secretome (HDP+Sec); 2) Human dental pulp stem cells (HDP); 3) Secretome (Sec); 4) Hyalgan as the positive control (Hya); 5) No treatment as the negative control (Ctrl). After 12 weeks since OA was confirmed, the healing process was examined by histopathology and radiology evaluations. Results: Histopathological evaluations, radiological assessments, and matrix indexes in three treatment groups significantly improved compared to the Ctrl and Hya groups. Surface in HDP+Sec was significantly better than the Ctrl group. In radiological evaluations, a significant decrease in OA was observed in the three treatment groups in comparison with the Ctrl groups. There was no significant difference between the treatment groups in any radiological and histopathological evaluations. HDP + Sec group slightly records better results compared to Sec or HDP treatment groups. Conclusion: It was concluded that human dental pulp stem cells and their secretome promote cartilage regeneration due to their cell protective potential as well as matrix degeneration reduction capacity. © 2023 Elsevier Lt
The Effect of Weight-Bearing Exercise on the Strength of Femur Bone in Ovariectomized Rats
Introduction & Objective: Fractures due to osteoporosis after menopause in women is widespread. Osteoporosis may occur in case of inadequate lack of physical activity .The aim of this study was to determine the effect of running training on femur bone strength in ovariectomized rats.
Materials & Methods Forty matured Sprague Dawley rats were chosen for this study. A group of 10 were killed randomly to measure their initial femur strength. The remaining rats had ovarian surgery. After three months, in order to reach menopause period, they were randomly divided into 3 groups, including pre test, running training and control groups. The running training program was carried out for one hour a day, five days a week, for eight weeks. Femur bone strength was measured by HOUNSFIELD system. Data was analyzed by using one-way analysis of variance and dependent T- tests by the SPSS software.
Results: Results of this study showed that ovariectomy leads to significant decrease of femur bone strength. On the other hand the eight weeks running training lead to significant increase of femur bone strength.
Conclusion: The results of this study suggest that life style is important factors in preventing of osteoporosis and running training program had an inhibitory or reversal effect on decrease of menopause-induced femur bone strength
