SAM-dependent enzyme-catalysed pericyclic reactions in natural product biosynthesis.
Pericyclic reactions-which proceed in a concerted fashion through a cyclic transition state-are among the most powerful synthetic transformations used to make multiple regioselective and stereoselective carbon-carbon bonds. They have been widely applied to the synthesis of biologically active complex natural products containing contiguous stereogenic carbon centres. Despite the prominence of pericyclic reactions in total synthesis, only three naturally existing enzymatic examples (the intramolecular Diels-Alder reaction, and the Cope and the Claisen rearrangements) have been characterized. Here we report a versatile S-adenosyl-l-methionine (SAM)-dependent enzyme, LepI, that can catalyse stereoselective dehydration followed by three pericyclic transformations: intramolecular Diels-Alder and hetero-Diels-Alder reactions via a single ambimodal transition state, and a retro-Claisen rearrangement. Together, these transformations lead to the formation of the dihydropyran core of the fungal natural product, leporin. Combined in vitro enzymatic characterization and computational studies provide insight into how LepI regulates these bifurcating biosynthetic reaction pathways by using SAM as the cofactor. These pathways converge to the desired biosynthetic end product via the (SAM-dependent) retro-Claisen rearrangement catalysed by LepI. We expect that more pericyclic biosynthetic enzymatic transformations remain to be discovered in naturally occurring enzyme 'toolboxes'. The new role of the versatile cofactor SAM is likely to be found in other examples of enzyme catalysis
Application of the indirect fluorescent antibody assay in the study of malaria infection in the Yangtze River Three Gorges Reservoir, China
<p>Abstract</p> <p>Background</p> <p>China Yangtze Three Gorges Project (TGP) is one of the biggest construction projects in the world. The areas around the Three Gorge Dam has a history of tertian malaria and subtertian malaria epidemic, but there are no overall data about malaria epidemics before the completion of the project. The objective of this study was to get a reliable baseline on malaria infection in the Yangtze River Three Gorges reservoir area and to provide reference data for future studies about the impact of the project on malaria epidemics.</p> <p>Methods</p> <p>Two surveys of malaria infection were carried out in area, at six-month intervals in May and October 2008. About 3,600 dual specimens blood film samples for parasite diagnosis and filter paper blood spots for serology (using the immunofluorescence antibody test) were collected from the general population, including school populations, whenever possible.</p> <p>Results</p> <p>The overall percentage of positive response of the same population during post-transmission periods was about twice (1.40/0.72) of that in pre-transmission. Positive individuals under 15 years of age were detected in all the localities.</p> <p>Conclusion</p> <p>A certain extent of malaria infection existed in this area. Additional studies are needed to determine the length of malaria experience, and chemotherapeutic intervention as well as the distribution of main vectors for transmission in this area.</p
Entanglement control in one-dimensional random XY spin chain
The entanglement in one-dimensional random XY spin systems where the
impurities of exchange couplings and the external magnetic fields are
considered as random variables is investigated by solving the different
spin-spin correlation functions and the average magnetization per spin. The
entanglement dynamics near particular locations of the system is also studied
when the exchange couplings (or the external magnetic fields) satisfy three
different distributions(the Gaussian distribution, double-Gaussian
distribution, and bimodal distribution). We find that the entanglement can be
controlled by varying the strength of external magnetic field and the different
distributions of impurities. Moreover, the entanglement of some
nearest-neighboring qubits can be increased for certain parameter values of the

Entanglement dynamics of two-qubit system in different types of noisy channels
In this paper, we study entanglement dynamics of a two-qubit extended
Werner-like state locally interacting with independent noisy channels, i.e.,
amplitude damping, phase damping and depolarizing channels. We show that the
purity of initial entangled state has direct impacts on the entanglement
robustness in each noisy channel. That is, if the initial entangled state is
prepared in mixed instead of pure form, the state may exhibit entanglement
sudden death (ESD) and/or be decreased for the critical probability at which

Malignancy in dermatomyositis: a mono-centric retrospective study of 134 patients in China and a potential predictive model
ObjectivesTo describe the demographics and phenotypes of malignancies-associated dermatomyositis (MADM) in east China and pinpoint potential factors indicative of malignancies in patients with dermatomyositis and establish a predictive model.MethodsWe retrospectively analyzed clinical data from 134 patients with adult-onset dermatomyositis hospitalized between January 2019 and May 2022 in one comprehensive hospital. Clinical data including disease course, initial symptoms and signs, and demographic information were retrieved from the Electronic Medical Records System. Other parameters including myositis-specific autoantibodies profiles, ferritin, sedimentation, etc. were all referable. Multivariable multinomial logistic regression was employed to simulate a model to predict cancer risks. Receiver operating characteristic curve was adopted to evaluate the potency of the model.Results134 patients with adult-onset dermatomyositis were aptly enrolled in this study based on inclusive and exclusive criteria: 12 (8.96%) with malignancies, 57 (42.53%) with aberrant tumor biomarkers but no malignancies, 65 (48.51%) with neither malignancies nor abnormal tumor biomarkers. Senior diagnostic age, higher LDH, higher ferritin, positive anti-TIF1γ and anti-Mi2 rather than anti-NXP2 autoantibodies were positive indicators of malignancies. Additionally, neither initial complaints nor signs were found to be correlated to a tendency towards malignancies. Digestive system, nasopharyngeal, and lung malignancies were mostly documented in east China. One multivariable multinomial logistic regression model was established to predict the phenotypes of dermatomyositis on the basis of potential malignancies and the overall sensitivity and specificity was satisfactory.ConclusionPositivity of anti-TIF1γ and anti-Mi2 autoantibodies are highly indicative of malignancies while the role of anti-NXP2 autoantibody in MADM in the Chinese population remains unclear. The phenotypes of malignancies can be predicted through the model and the predictive power is sufficient. More attention should be paid to malignancies screening in patients with aberrant tumor biomarkers but no malignancies, particularly digestive system, nasopharyngeal, and lung malignancies in patients with dermatomyositis but without malignancies
Unconventional Origin and Hybrid System for Construction of Pyrrolopyrrole Moiety in Kosinostatin Biosynthesis
SummaryKosinostatin (KST), an antitumor antibiotic, features a pyrrolopyrrole moiety spirally jointed to a five-membered ring of an anthraquinone framework glycosylated with a γ-branched octose. By a combination of in silico analysis, genetic characterization, biochemical assay, and precursor feeding experiments, a biosynthetic pathway for KST was proposed, which revealed (1) the pyrrolopyrrole moiety originates from nicotinic acid and ribose, (2) the bicyclic amidine is constructed by a process similar to the tryptophan biosynthetic pathway, and (3) a discrete adenylation enzyme and a peptidyl carrier protein (PCP) are responsible for producing a PCP-tethered building block parallel to type II polyketide synthase (PKS) rather than for the PKS priming step by providing the starter unit. These findings provide an opportunity to further explore the inexplicable enzymatic logic that governs the formation of pyrrolopyrrole moiety and the spirocyclic skeleton
More microbial manipulation and plant defense than soil fertility for biochar in food production: A field experiment of replanted ginseng with different biochars
The role of biochar–microbe interaction in plant rhizosphere mediating soilborne disease suppression has been poorly understood for plant health in field
conditions. Chinese ginseng ( Panax ginseng C. A. Meyer) is widely cultivated in
Alfisols across Northeast China, being often stressed severely by pathogenic
diseases. In this study, the topsoil of a continuously cropped ginseng farm was
amended at 20 t ha − 1, respectively, with manure biochar (PB), wood biochar
(WB), and maize residue biochar (MB) in comparison to conventional manure
compost (MC). Post-amendment changes in edaphic properties of bulk
topsoil and the rhizosphere, in root growth and quality, and disease incidence
were examined with field observations and physicochemical, molecular, and
biochemical assays. In the 3 years following the amendment, the increases
over MC in root biomass were parallel to the overall fertility improvement,
being greater with MB and WB than with PB. Differently, the survival rate
of ginseng plants increased insignificantly with PB but significantly with WB
(14%) and MB (21%), while ginseng root quality was unchanged with WB but
improved with PB (32%) and MB (56%). For the rhizosphere at harvest following
3 years of growing, the total content of phenolic acids from root exudate
decreased by 56, 35, and 45% with PB, WB, and MB, respectively, over MC.
For the rhizosphere microbiome, total fungal and bacterial abundance both
was unchanged under WB but significantly increased under MB (by 200 and
38%), respectively, over MC. At the phyla level, abundances of arbuscular
mycorrhizal and Bryobacter as potentially beneficial microbes were elevated while those of Fusarium and Ilyonectria as potentially pathogenic microbes
were reduced, with WB and MB over MC. Moreover, rhizosphere fungal
network complexity was enhanced insignificantly under PB but significantly
under WB moderately and MB greatly, over MC. Overall, maize biochar exerted
a great impact rather on rhizosphere microbial community composition and
networking of functional groups, particularly fungi, and thus plant defense
than on soil fertility and root growth
Genetic study of congenital bile-duct dilatation identifies de novo and inherited variants in functionally related genes
Background:
Congenital dilatation of the bile-duct (CDD) is a rare, mostly sporadic, disorder that results in bile retention with severe associated complications. CDD affects mainly Asians. To our knowledge, no genetic study has ever been conducted.
Methods:
We aim to identify genetic risk factors by a “trio-based” exome-sequencing approach, whereby 31 CDD probands and their unaffected parents were exome-sequenced. Seven-hundred controls from the local population were used to detect gene-sets significantly enriched with rare variants in CDD patients.
Results:
Twenty-one predicted damaging de novo variants (DNVs; 4 protein truncating and 17 missense) were identified in several evolutionarily constrained genes (p < 0.01). Six genes carrying DNVs were associated with human developmental disorders involving epithelial, connective or bone morphologies (PXDN, RTEL1, ANKRD11, MAP2K1, CYLD, ACAN) and four linked with cholangio- and hepatocellular carcinomas (PIK3CA, TLN1 CYLD, MAP2K1). Importantly, CDD patients have an excess of DNVs in cancer-related genes (p < 0.025). Thirteen genes were recurrently mutated at different sites, forming compound heterozygotes or functionally related complexes within patients.
Conclusions:
Our data supports a strong genetic basis for CDD and show that CDD is not only genetically heterogeneous but also non-monogenic, requiring mutations in more than one genes for the disease to develop. The data is consistent with the rarity and sporadic presentation of CDD
Entanglement and quantum phase transition in alternating XY spin chain with next-nearest neighbour interactions
By using the method of density-matrix renormalization-group to solve the
different spin-spin correlation functions, the nearest-neighbouring
entanglement(NNE) and next-nearest-neighbouring entanglement(NNNE) of
one-dimensional alternating Heisenberg XY spin chain is investigated in the
presence of alternating nearest neighbour interactions of exchange couplings,
external magnetic fields and next-nearest neighbouring interactions. For
dimerized ferromagnetic spin chain, NNNE appears only above the critical
dimerized interaction, meanwhile, the dimerized interaction effects quantum
phase transition point and improves NNNE to a large value. We also study the
effect of ferromagnetic or antiferromagnetic next-nearest neighboring (NNN)
interactions on the dynamics of NNE and NNNE. The ferromagnetic NNN interaction
increases and shrinks NNE below and above critical frustrated interaction
respectively, while the antiferromagnetic NNN interaction always decreases NNE.
The antiferromagnetic NNN interaction results to a larger value of NNNE in
