    Characterization of arsenic-resistant endophytic Priestia megaterium R2.5.2 isolated from ferns in an arsenic-contaminated multi-metal mine in Vietnam

    Bioremediation is a biological process to remove or neutralize environmental pollutants. This study was carried out to investing at the efficacy of arsenic resistant endophytic bacteria isolated from Pteris vittata, Pityrogramma calomelanos, Blenchum orientale, and Nephrolepis exaltata, which grow in a highly arsenic (As) contamination mining site in Vietnam. Their segmented roots, stems, and leaves were homogenized separately and inoculated on LB agar plates containing 5mM As(III) and As(V). A total of 31 arsenic resistant endophytic strains were selected, in which strain R2.5.2 isolated from the root of P. calomelanos had the highest arsenic resistant capability. Strain R2.5.2 tolerated up to 320 mM and 160 mM of arsenate and arsenite, respectively. The strain developed well on a media of 0.1 5% NaCl, at 20-40ºC and pH 5 9, and actively utilized most of the sugar sources. It had a high IAA biosynthesis capacity with an average concentration of 19.14 mg/L, tolerated to 0.5-16 mM concentration of Ag+, Hg2+, Co2+, Ni2+, Cu2+, Cr4+, and reduced As(V). Based on 16s rDNA, R2.5.2 was identified as Priestia megaterium. The ars C gene coding for arsenate reductase catalyzing reduction of As(V) was successfully amplified in P. megaterium R2.5.2.  The selected strain may have potential use for bioremediation practice

    Evaluation of Ursolic Acid as the Main Component Isolated from Catharanthus Roseus against Hyperglycemia

    ABSTRACT. Ursolic acid with large amount (0.67% of dried plant weight) along with 7 compounds, namely as spatozoate (1), kaurenoic acid (2), ursonic acid (3), 3-hydroxy-11-ursen-28,13-olide (4), ursolic acid (5), vindoline (6) and mixture of β-sitosterol and stigmasterol were isolated from dichloromethane and ethyl acetate extracts which have shown anti-glucosidase activity of the whole plant of C.roseus. Some isolated compounds and their derivatives were also tested for anti-glucosidase and cytotoxicity.Ursolic acid was examined for hypoglycemic activity in alloxan-induced diabetic mice with dose of 200 and 300 mg/kg/day, respectively. The results have shown that the blood glucose level were reduced by 45.75% and 51.31% to compare with the control group. This study has confirmed that the main component of Vietnamese C. roseus has had significant anti-hyperglycemia activity

    Simultaneous quantitative analyses of Tanshinone I, Cryptotanshinone, and Tanshinone IIA in Danshen (Salvia miltiorrhiza Bunge) cultivated in Vietnam using LC-MS/MS

    74-83By using chromatography methods, the principal compounds tanshinon I, cryptotanshinone, tanshinone IIA were isolated from danshen (Salvia miltiorrhiza Bunge). Based on the spectroscopic data (1H-NMR, 13C-NMR and ESI-MS mass spectra), the structures were determined. The compound was purified (purity > 99.8%) by Agilent 218 purification system, which was used as the standard for analyzing tanshinon I, cryptotanshinone, tanshinone IIA in six samples. In this study, one LC-MS/MS method was developed for the simultaneous quantitative determination of three bioactive principles, tanshinone I, cryptotanshinone, and tanshinone IIA in Radix Salviae miltiorrhizae (RSM, the root of S. miltiorrhiza). The quantification of these diterpenoids is based on the fragments of [M+H]+ under collision-activated conditions and in selected reaction monitoring (SRM) mode. The quantitative method is validated by determining the mean recovery from fortified samples of tanshinone I, cryptotanshinone, and tanshinone IIA as higher than 98%. The established method is successfully applied to the quality assessment of six batches of RSM samples collected from different regions of Vietnam. The results show that Lam Dong sample has the highest tanshinone I content (4.4286±0.0009 µg/mg), meanwhile Muong Long sample has the lowest (1.2717±0.0013µg/mg). Lam Dong sample has the highest cryptotanshinone content (8.1589±0.0006 µg/mg), whereas Guangxi-China sample has the lowest (2.8630±0.0008 µg/mg). Ha Giang sample has the highest tanshinone IIA content (13.0252±0.0004 µg/mg), whereas Muong Long sample has the lowest (3.8278±0.0003 µg/mg)

    When Intervention Becomes Imperative: A Case Report of Spontaneous Vulvar Edema During Pregnancy

    Spontaneous idiopathic vulvar edema during the second trimester is a rare condition. The approach to managing this condition involves relieving symptoms, identifying underlying causes, and implementing appropriate treatment. Managing such cases during pregnancy is challenging because of concerns for potential adverse fetal outcomes. Conservative management expects the condition to be relieved spontaneously postpartum, whereas invasive treatment offers a more rapid resolution. Treatment choices are controversial because each method has its pros and cons and influences the delivery process to a certain extent. Surgical drainage becomes a viable option when patients are not responsive to medications. We report a case of spontaneous massive vulvar edema in a 22-year-old primigravida in her 23rd week of pregnancy. After ruling out other notable causes of vulvar edema, we decided to intervene using an invasive procedure because she complained of progressive symptoms and discomfort. Subsequently, the edema subsided postprocedure, and the patient experienced successful labor with no complications. This report aims to alert clinicians that drainage attempts should be considered in pregnant patients with worsening symptoms

    Successful Psoriasis Treatment Using NB-UVB with Methotrexate: The Vietnamese Experience

    AIM: To compare the effectiveness of narrowband ultraviolet B (NBUVB) and oral methotrexate (MTX) to oral MTX alone in Vietnamese psoriasis patients, from May 2016 to May 2018. METHODS: We conducted a non-randomized trial on 70 patients with plaque-type psoriasis of moderate to severe. Thirty-five patients apply NBUVB once/day in 5 days/week for 4 weeks plus oral MTX 7.5 mg/week and 35 patients oral MTX 7.5 mg/week and both two groups treatment for 3 months. The extent of the lesion was assessed by the Psoriasis Area and Severity Index (PASI). RESULTS: The proportion of decreasing PASI was comparable (68.49% in NBUVB and MTX versus 57.62% in MTX alone); p < 0.05. Inside, good 28.58%, moderate 68.57% and poor 2.85% in NBUVB and MTX better than good 2.85%, moderate 71.4% and poor 25.72% in MTX alone; p < 0.05. The recurrence rate after 24 months of the NBUVB and MTX group (42.9%) was lower than the MTX alone group (71.4%); p < 0.05. CONCLUSION: NBUVB and oral MTX have affected treatment with chronic plaque psoriasis better than oral MTX alone

    The Efficacy of a Two-Fold Increase of H1-Antihistamine in the Treatment of Chronic Urticaria - the Vietnamese Experience

    BACKGROUND: Chronic urticaria, a mast cell-driven condition, is common, debilitating and hard to treat. H1-antihistamines are the first line treatment of chronic urticaria, but often patients do not get satisfactory relief with the recommended dose. European guidelines recommend increased antihistamine doses up to four-fold. AIM: We conducted this study to evaluate the efficacy of increased H1-antihistamine doses up to two-fold in Vietnamese chronic urticaria patients. METHODS: One hundred and two patients with chronic urticaria were recruited for treatment with levocetirizine (n = 52) or fexofenadine (n = 50). Treatment started at the conventional daily dose of 5 mg levocetirizine or 180 mg fexofenadine for 2 weeks and then increased to 10 mg levocetirizine or 360 mg fexofenadine for 2 weeks if patients did not have an improvement in symptoms. At week 0, week 2 and week 4 wheal, pruritus, size of the wheal, total symptom scores, and associated side-effects were assessed. RESULTS: With the conventional dose, the total symptom scores after week 2 decreased significantly in both groups compared to baseline figures, i.e. 7.4 vs 2.3 for levocetirizine group and 8.0 vs 2.6 for fexofenadine group (p < 0.05). However, there were still 26 patients in each group who did not have improvements. Of these 26 patients, after having a two-fold increase of the conventional dose, 11.5% and 38.5% became symptom-free at week 4 in levocetirizine group and fexofenadine group, respectively. At week 4 in both groups, the total symptom scores had significantly decreased when compared with those at week 2 (2.8 ± 1.5 versus 4.7 ± 1.6 in levocetirizine group; 2.1 ± 1.9 versus 5.1 ± 1.4 in fexofenadine group). In both groups, there was no difference in the rate of negative side effects between the conventional dose and the double dose. CONCLUSION: This study showed that increasing the dosages of levocetirizine and fexofenadine by two-fold improved chronic urticaria symptoms without increasing the rate of negative side effects

    Experimental cultivation of Spirulina platensis using My An mineral water, Thua Thien Hue province

    Spirulina platensis was experimentally cultivated by semi-continuous method at 20m2 scale pilot to effectively use the available bicarbonate and minerals of the My An mineral water, Phu Vang district, Thua Thien Hue province, and reduce production cost. Analyses of My An water quality showed that this mineral water source was suitable to cultivate Spirulina Platensis after removing H2S. The average algal yield reached to 10gDW/m2.day. The obtained algal biomass contained high nutrient compositions and was a suitable source for functional food (68.32 % protein, 7.32 % lipids containing Omega-3 and Omega-6, and low heavy metal concentration). The obtained results indicated that the My An mineral water source was indeed a suitable water source for producing Spirulina platensis biomass as a functional food
