    Peptide-modified Substrate for Modulating Gland Tissue Growth and Morphology In Vitro

    In vitro fabricated biological tissue would be a valuable tool to screen newly synthesized drugs or understand the tissue development process. Several studies have attempted to fabricate biological tissue in vitro. However, controlling the growth and morphology of the fabricated tissue remains a challenge. Therefore, new techniques are required to modulate tissue growth. RGD (arginine-glycine-aspartic acid), which is an integrin-binding domain of fibronectin, has been found to enhance cell adhesion and survival; it has been used to modify substrates for in vitro cell culture studies or used as tissue engineering scaffolds. In addition, this study shows novel functions of the RGD peptide, which enhances tissue growth and modulates tissue morphology in vitro. When an isolated submandibular gland (SMG) was cultured on an RGD-modified alginate hydrogel sheet, SMG growth including bud expansion and cleft formation was dramatically enhanced. Furthermore, we prepared small RGD-modified alginate beads and placed them on the growing SMG tissue. These RGD-modified beads successfully induced cleft formation at the bead position, guiding the desired SMG morphology. Thus, this RGD-modified material might be a promising tool to modulate tissue growth and morphology in vitro for biological tissue fabrication

    Magnetic Phase Diagrams with Possible Field-induced Antiferroquadrupolar Order in TbB2_2C2_2

    Magnetic phase diagrams of a tetragonal antiferromagnet TbB2_2C2_2 were clarified by temperature and field dependence of magnetization. It is noticeable that the N{\'e}el temperature in TbB2_2C2_2 is anomalously enhanced with magnetic fields, in particular the enhancement reaches 13.5 K for the {} direction at 10 T. The magnetization processes as well as the phase diagrams are well interpreted assuming that there appear field-induced antiferroquadrupolar ordered phases in TbB2_2C2_2. The phase diagrams of the AFQ compounds in RB2_2C2_2 are systematically understood in terms of the competition with AFQ and AFM interactions.Comment: 4 pages, 4 figures, RevTeX

    児童養護施設における自立支援計画の全国実態調査 : 日常生活支援への適用に向けて

    近年, 児童養護施設では自立支援計画の策定が義務化され,重要視されてきている。支援計臨の策定・確認・評価に関する業務過程の実態を明らかにするために,我々は全国の児童養護施設を対象に質問紙調査を行った。合計470名の職員から回答を得て、統計的分析を実施した。調査の結果,職員は特定の条件で自立支援計画に対して肯定的な評定を示した。1つ目に,多職種から助言を受ける時に職員はより役立っていると評価する傾向が見られた。2つ目に、ケース会議で支援目標を話し合う機会がある隊員はより自立支援計画について肯定的であった。3つ目に, 自立支援計画が高頻度で見返される場合に、職員は自立支援計画が役立っていると考える傾向が見られた。調査の結果から,児童養護施設の生活支援に自立支援計画を活用するためには,児童の支援目標を高頻度で時認して、達成状況を効率的に記録・共有できるような業務環境の整備が重要である可能性が示唆された。In late years it is mandatory for children\u27s homes in Japan to write up an independent living service (ILS) plan for each child in care, which is considered significantly important. To reveal the actual work process of making, monitoring, and evaluating ILS plans for children in care, we conducted a fact-finding survey nationwide. The participants were 470 workers at children\u27s homes, who completed a questionnaire. As a result of statistical analysis, the workers showed positive attitudes toward ILS plans mainly in 3 circumstances. First, they tended to think that ILS plans were more helpful for their work when receiving advice from multidisciplinary professionals. Second, the workers who had opportunities to share their cases with others at case conferences were more positive about ILS plans. Third, the workers tended to consider ILS plans useful when ILS plans were monitored at high frequency, which is once a month. In conclusion, in order to fully utilize ILS plans for children at children\u27s homes, it is suggested that we should prepare a work environment, where we can monitor case goals at high frequency, record, and share case goals with other workers.In conclusion, preparing work environments in that the children\u27s performances were monitored frequently as well as that the staffs were able to efficiently record and share each other\u27s cases were shown to be important when utilizing the self-reliance support plans in residential cares in Japan

    Purification and Characterization of Thermostable Endo-1,5-α-l-Arabinase from a Strain of Bacillus thermodenitrificans

    A strain of a thermophilic bacterium, tentatively designated Bacillus thermodenitrificans TS-3, with arabinan-degrading activity was isolated. It produced an endo-arabinase (ABN) (EC 3.2.1.99) and two arabinofuranosidases (EC 3.2.1.55) extracellularly when grown at 60°C on a medium containing sugar beet arabinan. The ABN (tentatively called an ABN-TS) was purified 7,417-fold by anion-exchange, hydrophobic, size exclusion, and hydroxyapatite chromatographies. The molecular mass of ABN-TS was 35 kDa as determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and the isoelectric point was pH 4.5. The enzyme was observed to be more thermostable than known ABNs; it had a half-life of 4 h at 75°C. The enzyme had optimal activity at 70°C and pH 6.0. The enzyme had apparent K(m) values of 8.5 and 45 mg/ml and apparent V(max) values of 1.6 and 1.1 mmol/min/mg of protein against debranched arabinan (α-1,5-arabinan) and arabinan, respectively. The enzyme had no pectin-releasing activity (protopectinase activity) from sugar beet protopectin, differing from an ABN (protopectinase-C) from mesophilic Bacillus subtilis IFO 3134. The pattern of degradation of debranched arabinan by ABN-TS indicated that the enzyme was an endo-acting enzyme and the main end products were arabinobiose and arabinose. The results of preliminary experiments indicated that the culture filtrate of strain TS-3 is suitable for l-arabinose production from sugar beet pulp at high temperature
