    A cell factory of Bacillus subtilis engineered for the simple bioconversion of myo-inositol to scyllo-inositol, a potential therapeutic agent for Alzheimer's disease

    <p>Abstract</p> <p>Background</p> <p>A stereoisomer of inositol, <it>scyllo</it>-inositol, is known as a promising therapeutic agent for Alzheimer's disease, since it prevents the accumulation of beta-amyloid deposits, a hallmark of the disease. However, this compound is relatively rare in nature, whereas another stereoisomer of inositol, <it>myo</it>-inositol, is abundantly available.</p> <p>Results</p> <p><it>Bacillus subtilis </it>possesses a unique inositol metabolism involving both stereoisomers. We manipulated the inositol metabolism in <it>B. subtilis </it>to permit the possible bioconversion from <it>myo</it>-inositol to <it>scyllo</it>-inositol. Within 48 h of cultivation, the engineered strain was able to convert almost half of 10 g/L <it>myo</it>-inositol to <it>scyllo</it>-inositol that accumulated in the culture medium.</p> <p>Conclusions</p> <p>The engineered <it>B. subtilis </it>serves as a prototype of cell factory enabling a novel and inexpensive supply of <it>scyllo</it>-inositol.</p

    Many slope failures and debris flows triggered by heavy rainfall associated with Typhoon 0421 occurred in Seisan area, Kagawa Prefecture, Japan. Critical rainfall to trigger the slope failure is 65mm/h, 150mm/3h, total precipitation 250mm, and soil water index 180. Many slope failures occurred at side-slope and/or debris filled floor of 1-order valley in mountain slopes consisting of alternating beds of sandstone and mudstone (Izumi Group). Field evidence suggests that rainwater penetrated into permeable weathered sandstone layer lying on poor permeable mudstone layer. Many slope failures occurred at weathered sandstone layer because of low strength and a rise in the groundwater level. The occurrences of slope failures were controlled by weathered profile of alternating beds of sandstone and mudstone

    Acoustic stiffness and change in plug cartilage over time after autologous osteochondral grafting: correlation between ultrasound signal intensity and histological score in a rabbit model

    We investigated quantitative changes over time in ultrasound signal intensity (an index of stiffness), signal duration (an index of surface irregularity), and interval between signals (an index of thickness) of plug cartilage in an animal model of autologous osteochondral grafting. A full-thickness osteochondral plug was surgically removed and replaced in male Japanese white rabbits (n = 22). Specimens obtained at day 0 and weeks 2, 4, 8, 12 and 24 postoperatively were assessed using an ultrasound system and by macroscopic and histological evaluation (modified Mankin's score). Histology revealed that the plug sank until 2 weeks postoperatively, and that newly formed cartilage-like tissue covered the plug, but at 24 weeks the tissue detached. The plug itself survived well throughout the period of observation. Although the signal intensity at the plug site was same as that in the sham operated contralateral knee at day 0, from 2 to 24 weeks postoperatively it was less than that in the sham knee. At 8 weeks, this difference was significant (P < 0.05). Modified Mankin's score revealed early degenerative changes at the site, but macroscopic examination did not. Signal intensity correlated significantly with score (both at day 0 and at the five postoperative time points [P < 0.05, r = -0.91] and as a whole [P < 0.05, r = -0.36]). Signal intensity also significantly correlated with the individual subscores for 'cartilage structure' (P < 0.05, r = -0.32) and 'cartilage cells' (P < 0.05, r = -0.30) from the modified Mankin's score, but not significantly with subscores for 'staining' and 'tidemark'. Signal duration correlated significantly with total score (as a whole [P < 0.05, r = 0.34]), but not significantly with the score for cartilage structure (P = 0.0557, r = 0.29). The interval between signals reflected well the actual thickness of the plug site. The significant relationships between ultrasound signal intensity and scores suggest that early degenerative changes in plug cartilage and cartilage-like tissue, especially in the superficial layer, are detectable by high-frequency ultrasound assessment

    In-situ Neutron Tomography on Mixing Behavior of Supercritical Water and Room Temperature Water in a Tubular Flow Reactor

    We have synthesized metal oxide nanoparticles through hydrothermal reaction at around 400 °C and 25 MPa by mixing the stream of metal ion solution at room temperature with another stream of supercritical water in a continuous flow-type reactor. In order to visualize the mixing behavior of the two streams, we performed neutron tomography measurements. By performing tomography measurements while rotating the mixing piece with supplying supercritical water and room temperature water, we succeeded in obtaining the three dimensional distribution of neutron attenuation. The results clearly showed how the two streams mix, which serves as a reference for numerical simulation

    Importance of Second-look Endoscopy on an Empty Stomach for Finding Gastric Bezoars in Patients with Gastric Ulcers

     Most gastric bezoars can be treated with endoscopic fragmentation combined with or without cola dissolution, whereas laparotomy or laparoscopic surgery is generally inevitable for small intestinal bezoars because they cause small bowel obstruction. Therefore, early diagnosis and management of gastric bezoars are necessary to prevent bezoar-induced ileus. To investigate the incidence of overlooked gastric bezoars during the initial esophagogastroduodenoscopy, we retrospectively reviewed the cases of 27 patients diagnosed with gastrointestinal bezoars. The bezoars were diagnosed using esophagogastroduodenoscopy (n=25), abdominal ultrasonography (n=1), and barium follow-through examination (n=1). Bezoars were overlooked in 9/25 patients (36.0%) during the initial endoscopy examination because the bezoars were covered with debris in the stomach. Of the 9 patients, 8 had concomitant gastric ulcers, and the other patient had gastric lymphoma. Although a computed tomography (CT) scan was performed before the second-look endoscopy in 8 of the 9 patients, the bezoars were mistaken as food debris on CT findings and were overlooked in these patients. In conclusion, gastric bezoars may not be discovered during the initial esophagogastroduodenoscopy and CT scan. In cases with debris in the stomach, second-look endoscopy is essential to detect bezoars
