Status of MSBS study at NAL
Two Magnetic Suspension and Balance Systems (MSBS) at the National Aerospace Laboratory (NAL) in Japan are introduced. They are the 10 cm MSBS and the 60 cm MSBS. They have 10 cm x 10 cm and 60 cm x 60 cm test sections. The control of suspending a model at the 10 cm MSBS is six degrees of freedom including the rolling moment control. The model for the rolling moment control has two pairs of small extra permanent magnets at both its ends plus a main cylindrical magnet. The rolling moment is generated by the magnetic forces acting on the extra magnets by controlled current passing through the four side coils independently. Test results show the roll angle of the model is controlled in this way. The dynamic calibration test was carried out at the MSBS in five degrees of freedom without the rolling moment control. The model is a simple cylindrical magnet magnetized along its axis. The obtained results show that the dynamic calibration with measured magnetic field intensity is much superior to that with the coil currents. The 60 cm MSBS was designed with some data obtained at the 10 cm one. It is fundamentally proportional to the 10 cm one in size and coil positions. The measured magnetic field intensity is not so strong as expected at design. It was operated first in 1993. The control is three degrees of freedom in the longitudinal direction
Dermoscopic Features of Pigmented Bowen's Disease in a Japanese Female Mimicking Malignant Melanoma
Various structures have been reported for dermoscopic features of pigmented Bowen's disease (BD), which could be a mimic of various pigmented skin lesions. A 79-year-old Japanese woman presented with a 3-year history of brown-black macule on her right upper arm without symptom. Dermoscopic examination demonstrated irregular flossy streaks, irregular brown dots/globules, blue-whitish regression structures, and overlaying whitish scaly areas. We suspected pigmented skin lesions including seborrheic keratosis, pigmented eccrine poroma, and malignant melanoma and excised completely with a 5 mm margin. Histopathological features were consistent with a diagnosis of pigmented BD. Although similar dermoscopic features might be revealed in pigmented skin lesions and it may occasionally be difficult to distinguish between pigmented BD and other pigmented skin lesions, dermoscopy would be useful in speculating pathologic features of pigmented BD
Glycated albumin suppresses glucose-induced insulin secretion by impairing glucose metabolism in rat pancreatic β-cells
<p>Abstract</p> <p>Background</p> <p>Glycated albumin (GA) is an Amadori product used as a marker of hyperglycemia. In this study, we investigated the effect of GA on insulin secretion from pancreatic β cells.</p> <p>Methods</p> <p>Islets were collected from male Wistar rats by collagenase digestion. Insulin secretion in the presence of non-glycated human albumin (HA) and GA was measured under three different glucose concentrations, 3 mM (G3), 7 mM (G7), and 15 mM (G15), with various stimulators. Insulin secretion was measured with antagonists of inducible nitric oxide synthetase (iNOS), and the expression of iNOS-mRNA was investigated by real-time PCR.</p> <p>Results</p> <p>Insulin secretion in the presence of HA and GA was 20.9 ± 3.9 and 21.6 ± 5.5 μU/3 islets/h for G3 (<it>P </it>= 0.920), and 154 ± 9.3 and 126.1 ± 7.3 μU/3 islets/h (<it>P </it>= 0.046), for G15, respectively. High extracellular potassium and 10 mM tolbutamide abrogated the inhibition of insulin secretion by GA. Glyceraldehyde, dihydroxyacetone, methylpyruvate, GLP-1, and forskolin, an activator of adenylate cyclase, did not abrogate the inhibition. Real-time PCR showed that GA did not induce iNOS-mRNA expression. Furthermore, an inhibitor of nitric oxide synthetase, aminoguanidine, and NG-nitro-L-arginine methyl ester did not abrogate the inhibition of insulin secretion.</p> <p>Conclusion</p> <p>GA suppresses glucose-induced insulin secretion from rat pancreatic β-cells through impairment of intracellular glucose metabolism.</p
Contributions of Three Starch Branching Enzyme Isozymes to the Fine Structure of Amylopectin in Rice Endosperm
Three starch branching enzyme (BE) isozymes, BEI, BEIIa, and BEIIb, are involved in starch biosynthesis in rice endosperm. Past in vivo and in vitro studies have suggested that each BE isozyme plays a distinct role in forming the fine structure of amylopectin. To elucidate more details of their roles, we prepared DNA constructs in which all the possible combinations of the expressions of these three isozymes were suppressed in developing rice endosperm. Analysis of the chain-length distributions of amylopectin produced under these various conditions confirmed the contributions of the individual BE isozymes to the fine structure of amylopectin in rice endosperm. Among these isozymes, the impact of loss of BEIIb activity on amylopectin fine structure was most remarkable and indicated that it plays a specific role in the synthesis of short chains with a 6–13 degree of polymerization (DP). The contribution of BEI to the amylopectin synthesis was unclear when only BEI activity was reduced. It was clear, however, when both BEI and BEIIb activities were substantially inhibited. The DP11-22 intermediate chains were markedly reduced in the ΔBEI/BEIIb line compared with the ΔBEIIb line, indicating that BEI plays a distinct role in the synthesis of these intermediate chains. Although no substantial change in amylopectin chain profile was detected in the ΔBEIIa line, the role of BEIIa could be deciphered by analyzing amylopectin fine structure from the ΔBEI/BEIIa/BEIIb line in comparison to that from ΔBEI/BEIIb line. This strongly suggests that BEIIa compensates for the role of BEI, rather than that of BEIIb, by forming intermediate chains of DP11-22. In addition, the new possibility that BEIIa is involved in the formation of starch granules in rice endosperm was suggested because the onset temperature for gelatinization of starch granules in the ΔBEIIa/BEIIb line was significantly higher than that in the ΔBEIIb line. In summary, the present study highlights the distinct roles of BEI, BEIIa, and BEIIb in the synthesis of amylopectin in developing rice endosperm
Analysis of pRb Family Binding Regions of Adenovirus Type 12 E1A
Adenovirus early 1A (E1A) products can form complexes with cellular pro-teins including the pRb tumor suppressor, pRb-related p107 and p130, and p300 proteins related to the CREB-binding protein (CBP) . Within the E1A sequence, CR2 and CR1 mediate interaction with pRb family proteins, and the amino termi-nus and CR1 are involved in association with p300 protein. These interactions are essential for the transforming activity of ElA proteins of various adenovirus serotypes. Since E1A of highly oncogenic adenovirus type 12 (Ad12) have some differences in transformation efficiency and oncogenicity from non-oncogenic Ad5 E1A, we have analyzed the role of CR1 and CR2 sequences of Ad12 E1A in trans-formation and pRb family binding using specific mutations. On the basis of the characteristic phenotypes of some CR1 mutations we suggest a modification of the previously proposed consensus regarding CR1 sequence for pRb binding. Combinations of CR1 and CR2 mutations revealed different roles for noncon-served residues within the conserved pRb binding motif LXCXE of CR2, and interaction of CR1 and CR2 of Ad12 E1A through specific amino acid residue is suggested. These structural features of Ad12 E1A proteins may explain at least in part the functional differences from Ad5 E1A proteins
Trends in Self-Rated Oral Health and Its Associations with Oral Health Status and Oral Health Behaviors in Japanese University Students: A Cross-Sectional Study from 2011 to 2019
Self-rated oral health (SROH) is a valid, comprehensive indicator of oral health status. The purpose of this cross-sectional study was to analyze how oral health behaviors and clinical oral status were associated with SROH and how they had changed over the course of nine years in Japanese university students. Data were obtained from 17,996 students who underwent oral examinations and completed self-questionnaires from 2011 to 2019. Oral status was assessed using the decayed and filled teeth scores, bleeding on probing (BOP), probing pocket depth, the Oral Hygiene Index-Simplified (OHI-S), oral health behaviors, and related factors. SROH improved from 2011 to 2019. The logistic regression model showed that university students who were female and had a high daily frequency of tooth brushing, no BOP, no decayed teeth, no filled teeth, and a low OHI-S score and were significantly more likely to report very good, good, or fair SROH. An interaction effect was observed between survey year and regular dental check-ups (year x regular dental check-ups). The improvement trend in SROH might be associated with changes in oral health behaviors and oral health status
NIASGBdb: NIAS Genebank databases for genetic resources and plant disease information
The National Institute of Agrobiological Sciences (NIAS) is implementing the NIAS Genebank Project for conservation and promotion of agrobiological genetic resources to contribute to the development and utilization of agriculture and agricultural products. The project’s databases (NIASGBdb; http://www.gene.affrc.go.jp/databases_en.php) consist of a genetic resource database and a plant diseases database, linked by a web retrieval database. The genetic resources database has plant and microorganism search systems to provide information on research materials, including passport and evaluation data for genetic resources with the desired properties. To facilitate genetic diversity research, several NIAS Core Collections have been developed. The NIAS Rice (Oryza sativa) Core Collection of Japanese Landraces contains information on simple sequence repeat (SSR) polymorphisms. SSR marker information for azuki bean (Vigna angularis) and black gram (V. mungo) and DNA sequence data from some selected Japanese strains of the genus Fusarium are also available. A database of plant diseases in Japan has been developed based on the listing of common names of plant diseases compiled by the Phytopathological Society of Japan. Relevant plant and microorganism genetic resources are associated with the plant disease names by the web retrieval database and can be obtained from the NIAS Genebank for research or educational purposes
