The roles of the microtubular system in the transport and secretion of surfactant phospholipids in the lung were studied using colchicine-treated rats, who were administered in vivo by (methyl-3H) choline and (2-3H) glycerol. The radioactivities of phosphatidylcholine, disaturated phosphatidylcholine and phosphatidylglycerol were measured in four lung fractions, i.e., lung parenchyma, microsomes, lamellar bodies and alveolar wash, of colchicine-treated animals. The prior injection of colchicine resulted in marked increases in the labels of the phospholipids in microsomes as well as in lamellar bodies, and in the decrease in alveolar wash. These findings suggest that the microtubular system participates not only in the secretion of lamellar bodies into the alveolar space, but also in the translocation of surfactant phospholipids from the synthetic site, i.e., microsomes to lamellar bodies of the store house
Pathological Role of Tonsillar B Cells in IgA Nephropathy
Although impaired immune regulation along the mucosa-bone marrow axis has been postulated to play an important role, the pathogenesis of IgA nephropathy (IgAN) is unknown; thus, no disease-specific therapy for this disease exists. The therapeutic efficacy of tonsillectomy or tonsillectomy in combination with steroid pulse therapy for IgAN has been discussed. Although randomized control trials for these therapies are ongoing in Japan, the scientific rationale for these therapies remains obscure. It is now widely accepted that abnormally glycosylated IgA1 and its related immune complex (IC) are probably key molecules for the pathogenesis, and are thus considered possible noninvasive biomarkers for this disease. Emerging evidence indicates that B cells in mucosal infections, particularly in tonsillitis, may produce the nephritogenic IgA. In this paper, we briefly summarize characteristics of the nephritogenic IgA/IgA IC, responsible B cells, and underlying mechanisms. This clinical and experimental information may provide important clues for a therapeutic rationale

Role of treatment for solitary pulmonary nodule in breast cancer patients
<p>Abstract</p> <p>Background</p> <p>Metastatic pulmonary tumors secondary to breast cancer detected either before or after surgery are predominantly multiple and bilateral. However, in cases detected to have a solitary pulmonary nodule (SPN), determining whether the lesion represents a primary cancer, metastasis, or a benign pulmonary lesion can be difficult.</p> <p>Materials and methods</p> <p>Between January 2000 and December 2009, we performed breast cancer surgery on 1,226 patients, of which 49 cases (3.9%) were detected to have pulmonary lesions before or after the surgery. In 14 of these patients, video-assisted thoracoscopic surgery was performed to remove a SPN.</p> <p>Result</p> <p>Pathological examination of the resected specimens in these 14 cases revealed metastatic pulmonary tumor in 8 cases, primary lung cancer in 3 cases, and benign disease in 3 cases. While lobectomy was performed in one of these patients with metastatic pulmonary tumor, the remaining 7 underwent partial resection of the lung. The primary lung cancer was an adenocarcinoma in all 3 patients, and lobectomy plus mediastinal lymph node dissection was performed in these patients. The tumor grading based on pathological diagnosis was T1N0M0, p-Stage 1A in all 3 patients. The prognosis was good in the breast cancer patients in whom the metastatic lung tumor was a SPN.</p> <p>Conclusion</p> <p>Evaluating the immunohistochemical cytokeratin profile and levels of the TTF-1 and GCDFP-15 of the lesion was useful when distinguishing between pulmonary cancer and metastatic pulmonary tumor. In addition, some patients exhibited changes in the biological properties of the metastatic tumor, and delete tumor resection by video-assisted thoracoscopic surgery can be useful for deciding the drug treatment strategy in some cases</p
A Geophysical Interpretation of the Secular Displacement and Gravity Rates Observed at Ny-Ålesund, Svalbard in the Arctic— Effects of Post-Glacial Rebound and Present-Day Ice Melting
We have analysed the Ny-Ålesund very long baseline interferometry (VLBI) data over the period 1994 August to 2004 May, and we obtain secular displacement rates relative to a NNR-NUVEL-1A reference frame of and for the north, east and vertical directions, respectively. The corresponding global positioning system (GPS) station displacement rates relative to the same reference frame for the north, east, and vertical directions are at NYA1 and at NALL, where these GPS rates were derived from the ITRF2000 velocity solution of Heflin. From the comparison at 25 globally distributed collocated sites, we found that the difference in uplift rate between VLBI and GPS at Ny-Ålesund is mainly due to a GPS reference frame scale rate error corresponding to 1.6 mm yr−1 in the GPS vertical rates. The uplift rate was estimated to be 5.2 ± 0.3 mm yr−1 from the analysis of the tide gauge data at Ny-Ålesund. Hence the uplift rates obtained from three different kinds of data are very consistent each other. The absolute gravity (AG) measurements at Ny-Ålesund, which were carried out four times (period: 1998–2002) by three different FG5 absolute gravimeters, lead to a decreasing secular rate of . In this analysis, the actual data obtained from a superconducting gravimeter at Ny-Ålesund were used in the corrections for the gravity tide (including the ocean tide effect) and for the air pressure effect. We have estimated three geophysical contributions to examine the observed rates: (1) the effect of the sea-level (SL) change on a timescale of a few decades, (2) the effect of the present-day ice melting (PDIM) in Svalbard and (3) the sensitivity of the computed post-glacial rebound (PGR) effects to different choices of the models of past ice history and Earth\u27s viscosity parameters. Our analysis indicates that the effect of SL change can be neglected as the main source of the discrepancy. On the other hand, the effect of PDIM cannot be ignored in explaining the mutual relation between the observed horizontal and vertical rates and the predicted ones. A large melting rate of the order of −75 cm yr11 (i.e. roughly 1.6 times larger than the mean rate derived from glaciology over Svalbard) would explain the observed uplift but only half of the gravity changes. Our comparison results clearly point out the importance of both the estimation accuracy of the elastic deformations and better observation accuracy to constrain the size of PGR effects in the northwestern Svalbard more tightly
Broccoli consumption and chronic atrophic gastritis among Japanese males: an epidemiological investigation.
Previous in vitro and animal experiments have shown that sulforaphane, which is abundant in broccoli, inhibits Helicobacter pylori (H. pylori) infection and blocks gastric tumor formation. This suggests that broccoli consumption prevents chronic atrophic gastritis (CAG) introduced by H. pylori infection and, therefore, gastric cancer. For an epidemiological investigation of the relationship between the broccoli consumption and CAG, a cross-sectional study of 438 male employees, aged 39 to 60 years, of a Japanese steel company was conducted. CAG was serologically determined with serum cut-off values set at pepsinogen I < or = 70 ng/ml and a ratio of serum pepsinogen I/pepsinogen II < or = 3.0. Broccoli consumption (weekly frequency) and diet were monitored by using a 31-item food frequency questionnaire. The prevalence of CAG among men who ate broccoli once or more weekly was twice as high as that among men who consumed a negligible amount (P < 0.05). Multiple logistic regression analysis indicated that broccoli consumption once or more weekly significantly increased the risk for CAG (odds ratio, 3.06; 95% confidence interval, 1.12-8.38; P < 0.05), after controlling for age, education, cigarette smoking, and alcohol consumption. The present study failed to show an expected association between frequent broccoli consumption and a low prevalence of CAG.</p
A validation of the Japanese adaptation of the Big Five Inventory-2
The purpose of this study was to adapt a Japanese version of the Big Five Inventory-2 (BFI-2-J) to examine its factor structure, reliability, validity, and measurement invariance. The BFI-2-J assesses five domains and 15 facets of the Big Five personality traits. We analyzed two datasets: 487 Japanese undergraduates and 500 Japanese adults. The results of the principal component analysis and confirmatory factor analysis revealed that the domain-facet structure of the BFI-2-J was similar to that of other language versions. The reliability of the BFI-2-J is sufficient. The correlation coefficients between the BFI-2-J and the other Big Five and self-esteem measures supported convergent and discriminant validity. Moreover, we confirmed measurement invariance across age and sex groups in domain-level and facet-level models. The results suggest that the BFI-2-J is a good instrument for measuring the Big Five personality traits and their facets in Japan. The BFI-2-J is expected to be useful in Japanese personality research and international comparative research
Delivery of pDNA to the Lung by Lipopolyplexes Using N-Lauroylsarcosine and Effect on the Pulmonary Fibrosis
In a previous study, we constructed a lung-targeting lipopolyplex containing polyethyleneimine (PEI), 1,2-di-O-octadecenyl-3-trimethylammonium propane (DOTMA), and N-lauroylsarcosine (LS). The lipopolyplex exhibited an extremely high gene expression in the lung after intravenous administration. Here, we optimized the lipopolyplex and used it to deliver a TGF-β1 shRNA to treat refractory pulmonary fibrosis. We constructed several lipopolyplexes with pDNA, various cationic polymers, cationic lipids, and LS to select the most effective formulation. Then, the pDNA encoding shRNA against mouse TGF-β1 was encapsulated in the lipopolyplex and injected into mice with bleomycin-induced pulmonary fibrosis. After optimizing the lipopolyplex, dendrigraft poly-L-lysine (DGL) and DOTMA were selected as the appropriate cationic polymer and lipid, respectively. The lipopolyplex was constructed with a pDNA, DGL, DOTMA, and LS charge ratio of 1:2:2:4 showed the highest gene expression. After intravenous administration of the lipopolyplex, the highest gene expression was observed in the lung. In the in vitro experiment, the lipopolyplex delivered pDNA into the cells via endocytosis. As a result, the lipopolyplex containing pDNA encoding TGF-β1 shRNA significantly decreased hydroxyproline in the pulmonary fibrosis model mice.
