Identification and stage-specific association with the translational apparatus of TbZFP3, a CCCH protein that promotes trypanosome life-cycle development

The post-transcriptional control of gene expression is becoming increasingly important in the understanding of regulated events in eukaryotic cells. The parasitic kinetoplastids have a unique reliance on such processes, because their genome is organized into polycistronic transcription units in which adjacent genes are not coordinately regulated. Indeed, the number of RNA-binding proteins predicted to be encoded in the genome of kinetoplastids is unusually large, invoking the presence of unique RNA regulators dedicated to gene expression in these evolutionarily ancient organisms. Here, we report that a small CCCH zinc finger protein, TbZFP3, enhances development between life-cycle stages in Trypanosoma brucei. Moreover, we demonstrate that this protein interacts both with the translational machinery and with other small CCCH proteins previously implicated in trypanosome developmental control. Antibodies to this protein also co-immunoprecipitate EP procyclin mRNA and encode the major surface antigen of insect forms of T. brucei. Strikingly, although TbZFP3 is constitutively expressed, it exhibits developmentally regulated association with polyribosomes, and mutational analysis demonstrates that this association is essential for the expression of phenotype. TbZFP3 is therefore a novel regulator of developmental events in kinetoplastids that acts at the level of the post-transcriptional control of gene expression

A influência do uso de espaços compartilhados sobre a prática da inovação aberta das empresas / The influence of the use of shared spaces on the practice of open business innovation

Com o crescimento de novas formas de locais de trabalho, abriu-se espaço para ambientes como o coworking e hub de inovação. Dentre seus benefícios, destacam-se a possibilidade de ampliar a rede de contatos e alavancar o processo de networking e posteriormente o processo de inovação das empresas. Ao mesmo tempo, o modelo de inovação aberta vem ganhando destaque nos últimos anos, apresentando processos menos burocráticos e facilidade na troca de informações. Com o objetivo de entender como o compartilhamento de espaços pode influenciar o modelo de inovação aberta nas empresas, estudou-se a caracterização, organização e funcionamento desses espaços. A partir da análise de conteúdo dos dados coletados, observou-se que o compartilhamento de espaços se mostra uma possibilidade de aumentar a rede de contatos empresarial, impactando de forma positiva a criação de modelos abertos de inovação.

Avaliação do estilo de vida e o impacto da pandemia da COVID-19 em estudantes de medicina e enfermagem

Introduction: Lifestyle directly affects an individual’s quality of life and subsequently their health. In 2020, society faced a pandemic which had financial and health impacts within a physical and emotional context. Objectives: To analyze such impacts on the lifestyle of medical and nursing students of the Pontifícia Universidade Católica de São Paulo, and to verify the outcome of the teaching-learning method by remote activities. Methods: This was a quantitative, exploratory and descriptive study. Data were collected using the Google Forms tool, the sample was composed of 45 students from the Nursing and Medicine courses at the aforementioned institution, and the questionnaire applied was titled “Fantastic lifestyle” and aimed at measuring the main elements that characterize an appropriate lifestyle for good health. The method used for the descriptive analysis of the variables was the measurement of absolute and relative frequency. Results: A large part of the students had a considerably good lifestyle according to the parameters of the instrument applied and the remote activities suffered interference from internet connection thus there were difficulties in using them. Regarding body mass index, the results showed a number of overweight and obese students. Conclusion: It is hoped that there will be awareness and reflection on lifestyle and the changes that have occurred during the period of social isolation, reinforcing that quality of life should be a priority for people who are training to care for other people.Introdução: O estilo de vida afeta diretamente a qualidade de vida de um indivíduo e, posteriormente, sua saúde. No ano de 2020 a sociedade enfrentou uma pandemia que causou impactos financeiros e de saúde em seu contexto físico e emocional. Objetivos: Analisar os impactos causados no estilo de vida de estudantes de Medicina e Enfermagem da Pontifícia Universidade Católica de São Paulo (PUC-SP) e verificar os efeitos do método de ensino-aprendizagem por atividades remotas. Métodos: Trata-se de estudo quantitativo, exploratório e descritivo. Os dados foram coletados por meio da ferramenta Google Forms, a amostra foi constituída de 45 estudantes dos cursos de Enfermagem e Medicina da instituição acima mencionada, e o questionário aplicado, intitulado “Estilo de vida fantástico”, tinha por objetivo de mensurar os principais elementos que caracterizam o estilo de vida adequado para a saúde. O método utilizado para a análise descritiva das variáveis foram as medidas de frequência absoluta e relativa. Resultados: Grande parte dos estudantes apresentou um estilo de vida consideravelmente bom de acordo com os parâmetros do instrumento aplicado, e as atividades remotas tiveram interferências de internet, de modo que houve dificuldades para o aproveitamento. Em relação ao índice de massa corpórea, os resultados mostraram que há um número considerável de estudantes em sobrepeso e obesidade. Conclusão: Espera-se que ocorram conscientização e reflexão acerca do estilo de vida e das alterações decorridas no período de isolamento social, reforçando que a qualidade de vida deve ser prioridade para pessoas que se preparam para cuidar de outras pessoas

A Polyadenylation Factor Subunit Implicated in Regulating Oxidative Signaling in Arabidopsis thaliana

BACKGROUND: Plants respond to many unfavorable environmental conditions via signaling mediated by altered levels of various reactive oxygen species (ROS). To gain additional insight into oxidative signaling responses, Arabidopsis mutants that exhibited tolerance to oxidative stress were isolated. We describe herein the isolation and characterization of one such mutant, oxt6. METHODOLOGY/PRINCIPAL FINDINGS: The oxt6 mutation is due to the disruption of a complex gene (At1g30460) that encodes the Arabidopsis ortholog of the 30-kD subunit of the cleavage and polyadenylation specificity factor (CPSF30) as well as a larger, related 65-kD protein. Expression of mRNAs encoding Arabidopsis CPSF30 alone was able to restore wild-type growth and stress susceptibility to the oxt6 mutant. Transcriptional profiling and single gene expression studies show elevated constitutive expression of a subset of genes that encode proteins containing thioredoxin- and glutaredoxin-related domains in the oxt6 mutant, suggesting that stress can be ameliorated by these gene classes. Bulk poly(A) tail length was not seemingly affected in the oxt6 mutant, but poly(A) site selection was different, indicating a subtle effect on polyadenylation in the mutant. CONCLUSIONS/SIGNIFICANCE: These results implicate the Arabidopsis CPSF30 protein in the posttranscriptional control of the responses of plants to stress, and in particular to the expression of a set of genes that suffices to confer tolerance to oxidative stress

The connections thar weave the future nurse's competences

Este estudo apóia-se na premissa de que o ato pedagógico é uma prática social; desse fato, o cenário é indissociável, complexo e contraditório criando-se uma série de conexões que constitui as competências de futuros enfermeiros. A questão que orientou este estudo assim se coloca: A realidade na qual tanto os fenômenos visíveis imprevistos, fracassos, conquistas e celebrações bem como os fenômenos invisíveis - as emoções, motivos necessidades, interesses de que modo integram e condicionam a formação do indivíduo? O procedimento da pesquisa ação participante foi construído através de uma conversa reflexiva com a situação. A compreensão da gênese da subjetividade social que produz os sistemas de sentidos na ação educativa levou-nos a três aspectos que constituíram os espaços teóricos hipotéticos construtivo interpretativo:1) Partimos da história dos currículos de enfermagem e a confrontamos com fatos históricos do Curso de Enfermagem da PUC-SP. 2) As variáveis: o desencontro entre a teoria e a prática e as configurações subjetivas do espaço social em que ocorre a aprendizagem, 3) Ancoramos no presente e focalizamos no que nos move para uma mudança na construção do novo currículo. É aqui que o passado e o presente se fundem, e o presente é visto à luz da História. Ao tentar decifrar o emaranhado de fatos que ocorrem no tecer das competências de futuros enfermeiros, pedimos emprestado olhares e diálogos de outras pessoas (alunos, colegas de trabalho e estudiosos no assunto) que apesar da distância e as lembranças do passado, com suas dúvidas e proposições, contribuíram para elaborar argumentos que possam diminuir a insatisfação desse cenário que ora se apresenta e oferecer subsídios para decifrar as preocupações, que neste momento estamos vivenciando, de reforma curricular do Curso de Enfermagem da PUC-SP para adequar às novas Diretrizes CurricularesThis study lies on the premise that the pedagogic action is a social practice of such fact: the scenery is undissociable, complex and contradictory, creating a series of connections that constitutes the future nurse's competences. The subject that guided this study is thus put up as: The reality in the which both the visible phenomena - unexpectation, failures, conquests and celebrations - and the invisible phenomena - the emotions, motives, necessities, interests - do they integrate condition the individual's formation? The procedure of the research participant action was built through a reflexive conversation with the situation. To understand the genesis of the social subjectivity that it produces the systems of senses in the educational action took us to three aspects that constituted the theoretical-hypothetical-constructiveinterpretative spaces: 1) we appealed starting from the history of the nursing currícula, and we confronted with historical facts of the Course of Nursing of PUCSP. 2) the variables: the difference between the theory and the practice and the subjective configurations of the social space in which the learning takes place, 3) Finally, we have anchored in the present time and focused in whatever moves us for a change in the construction of the new currículum, it is right here that the past and the present are founded and the present is seen to the light of the History. When deciphering the entanglements of the facts that happen while weaving the future nurse's competences we borrow glances and other people's dialogues (students, work mates and experts in the subject) that in spite of the distance and the memories of the past, with their doubts and propositions, they contributed to elaborate arguments to reduce the dissatisfaction of that scenery that now we are presented to and to offer subsidies to decipher the concerns, that at this very moment time are living, of the curricular reform of the Course of Nursing of PUC-SP to adapt to the new Curricular Directive

Assistência de enfermagem pré-hospitalar às emergências: um novo desafio para enfermagem

PROJETO RESGATE nasceu de uma ação conjunta, entre a Secretaria Estadual da Saúde e a Secretaria da Segurança Pública de São Paulo, que reuniram seus recursos materiais e humanos para tratar do problema das emergências traumáticas, que acometem a população do Estado de São Paulo. 0 presente trabalho traz o relato da experiência de uma Enfermeira, que está inserida no PROJETO RESGATE, dando atendimento de enfermagem pré-hospitalar às emergências, utilizando-se como meio de transporte um helicóptero pertencente a Polícia Militar de São Paulo. Trata-se de um serviço novo, pioneiro no Brasil, onde os profissionais Médico e Enfermeiro saem dos seus ambientes habituais de trabalho para irem até o local onde se encontro paciente e dar os primeiros cuidados
