    The effects of different egg yolk concentrations used with soy bean lecithin-based extender on semen quality to freeze bull semen

Eurasian J Vet Sci, 26, 1, 45-49 Amaç: Boğa spermasını dondurmak için soya lesitin temelli sulandırıcıya (Bioxcell ® ) %5 ve 10 konsantrasyonlarında santrifüj edilmiş yumurta sarısı (SYS) katıldı ve bunların çözüm sonu sperm motilite, morfolojik anormallikler ve membran bütünlüğü üzerindeki sinerjistik etkileri değerlendirildi. Gereç ve Yöntem: Her bir Simental boğadan alınan ejakulatlar (n=12) üç eşit miktara ayrıldı ve sırasıyla %5 (B5), %10 (B10) SYS eklenmiş ve hiç SYS (B0) içerme-yen soya lesitin temelli sulandırıcı ile sulandırıldı. Ardın-dan standart protokollere göre donduruldu ve çözdürül-dü. Spermatozoa kryocanlılığı, in vitro çözüm sonu motilite (CASA), akrozomal ve diğer anormallikler ve plasma membran bütünlüğü (HOST) yönünden değerlendirildi. Bulgular: Simental boğalarda, dondurma ve çözdürme sonrası B5 ile sulandırılan grupta, diğer sulandırıcılar-la sulandırılan gruplardan önemli derecede daha yüksek CASA motilite ve CASA progresif motilite oranı elde edilmiştir (p<0.001). Gruplar arasında, VAP, VCL ve ALH yö-nünden önemli bir farklılık bulunmamıştır (p>0.05). En yüksek VSL (p<0.01) ve LIN değeri (p<0.001) B10 grubundan elde edilmiştir. Membran bütünlüğü oranı, B5 grubunda, diğer gruplara göre önemli derecede yüksek bulunmuştur (p<0.001). Öneri: Soya lesitinle kombine olarak sulandırıcıya %5 SYS eklenmesi boğa spermasının dondurulabilirliğini önemli derecede artırmıştır. Eurasian J Vet Sci, 26, 1, 45-49 Aim: The centrifuged egg yolk (CEY) was added at concentrations of 0.5 or 10% to a defined soy lecithin-based extender (Bioxcell ® ) used to freeze bull semen and their synergistic effects on post-thaw sperm motility, morphological abnormalities and membrane integrity assessed. Materials and Methods: Ejaculates obtained from each Simmental bull (n:12) divided in three equal aliquots and diluted in CEY 5% (B5), 10% (B10) supplemented, and without any CEY (B0) in soy bean lecithin -based extender, respectively. Thereafter, they were frozen and thawed following a standart protocol. Sperm cryosurvival was evaluated in vitro by microscopic assessments of post-thaw sperm motility (by means of the CASA), acrosomal and other abnormalities (head, mid-pieces, and tail) and plasma membrane integrity (evaluated by HOST). Results: In Simmental bulls, semen extended with B5 had significantly higher CASA motility and CASA progressive motility than those extended with the rest of extenders after freezing and thawing (p<0.001). There was no significant difference in VAP, VCL, and ALH among the three groups (p>0.05). For VSL (p<0.01) and LIN (p< 0.001), the highest values were obtained from B10 group. The highest percentages of acrosomal and other abnormalities were found in semen diluted in B10 (p<0.001). In the group frozen B5, the percentage of membrane integrity was significantly higher than that of the other groups (p<0.001). Conclusion: The use of CEY 5% in combination with soy bean lecithin significantly improved bull semen freezability

    Effect of glutamine and sugars after bull spermatozoa cryopreservation

    The objective of this study was to evaluate the effects of the addition of different sugars (raffinose, sucrose, and trehalose) on bull spermatozoa cryopreserved in a commercial extender (Optidyl) supplemented with glutamine on semen parameters, fertilizing ability and superoxide dismutase (SOD) activity. Nine ejaculates for each bull were used in the study. Semen was frozen in five different extenders: raffinose 25 mM plus glutamine 3 mM (RGO), sucrose 25 mM plus glutamine 3 mM (SGO), trehalose 25 mM plus glutamine 3 mM (TGO), glutamine 3 mM (GO) and control (0). Insemination doses were processed so that each 0.25 mL straw contained 15 x 10(6)sperm. Groups of GO and RGO resulted in the higher rates of subjective (54.0 +/- 1.7% and 64.0 +/- 1.1%; P 0.05). For pregnancy rates, there were no significant differences among the groups. The supplementation of additives did not provide any significant difference on the level of SOD activity (P > 0.05). It can be also thought that these sugars might have worked with glutamine in a synergy. Thereby, sugars such as raffinose and sucrose with glutamine in freezing extender may be recommended to facilitate bull semen freezability. (C) 2011 Elsevier Inc. All rights reserved

    The effect of green tea extract supplementation in bull semen cryopreservation

    WOS: 000471814000017This study aims to investigate the effect of catechin (CT), green tea extract, as a supplement to Tris extender on semen quality parameters in frozen-thawed of bull sperm. Ejaculates were taken with artificial vagina from Holstein bulls and divided equal five aliquots, diluted to containing different amounts of CT (5, 10, 25 and 50 mu g/mL) and no-additive (control). All samples were equilibrated at 4 degrees C for 4 h and were frozen using a digital freezing machine. Post-thawed sperm motility and kinetic parameters were determined using the sperm analyser system. Spermatozoa DNA integrity was evaluated with the single cell gel electrophoresis, abnormal spermatozoa rate was evaluated by fluid fixation test and lipid peroxidation status was evaluated calorimetrically. CT supplementation did not improve motility and kinetic parameters. However, the higher morphological integrity was detected in CT10, 25 and 50 groups compared to control (P<0.05). Regarding chromatin integrity, positive effects of catechin were observed in the treatment groups while in CT 50 group adverse effects were found (P<0.05). Although there was no improvement in malondialdehyde levels, the highest total antioxidant activity was seen in the CT50 group (P<0.05). In conclusion, CT supplementation could be used the protection of morphological and DNA integrity from cryodamage and it has increased the total antioxidant activity depending of the dose in bull semen.General Directorate of Agribusiness, Ankara, TurkeyThis study was supported by General Directorate of Agribusiness, Ankara, Turkey

    Effect of cholesterol loaded cyclodextrin on semen cryopreservation of Aksaray Malakli shepherd dogs of different ages

    WOS: 000433999000021PubMed: 29699920The objective of the study was to determine the effect of cholesterol-loaded cyclodextrin (CLC) on the quality parameters of semen from Aksaray Malakli Shepherd dogs of different age groups. Forty-eight male dogs were divided into 3 groupings according to their ages (young age (Y): = 7 years; n: 8). The sperm-rich portion of the ejaculate from each dog was divided into four aliquots and extended with either tris as a control (C) or tris loaded with 0.5, 1.0, and 1.5 mg/120 x 10(6) CLC as low (L), intermediate (I), and high (H) doses, respectively. Following equilibration for at least half an hour, the straws were frozen in nitrogen vapor and then stored in liquid nitrogen at least for 48 h. Later, the frozen straws were thawed in a water bath for spermatological evaluation. Significant differences were observed between different age groups in terms of the spermatological parameters (p < 0.05). The evidence suggests that increasing age is associated with poor in-vitro spermatological parameters and CLC was able to protect the acrosome integrity from cryo-damage during the freeze-thawing process.     Cryoprotective effect of vaccenic acid supplemented in bull semen extender

    The aim of this study was to investigate whether vaccenic acid (VA) added to TRIS semen extender has an effect on freezing bull semen. The semen was collected at intervals of three days through an artificial vagina then pooled. The pooled ejaculates were divided into five components as the control (C; no supplement) and VA (25, 50, 100, and 200 mu g / mL) groups then extended with the TRIS-based medium. The samples were left in a cold air cabinet for cooling process (4 degrees C), then they were equilibrated for 4 h. Afterward, the samples in the groups were frozen by using a programmable freezer. The greatest progressive and total motility results were obtained in the VA25 group (p < 0.001). Besides, the total abnormality and chromatin damage was determined to be on the lowest level in the VA25 group (p < 0.05). Except for VA200, the glutathione level was greater in the treatment groups in comparison to the control group (p < 0.05), while the lowest malondialdehyde was determined in VA25. In conclusion, VA supplemented with TRIS-based extender at the concentration of 25 mu g / mL improves the progressive and total motility properties of spermatozoa, reduces the abnormal spermatozoa rate, and has a positive effect on chromatin integrity

    The effects of different sugars on motility, morphology and DNA damage during the liquid storage of rat epididymal sperm at 4 degrees C

    This study evaluated the protective effects of supplementation with three different sugars on the motility, morphology and DNA integrity of rat epididymal sperm chilled and stored at 4 degrees C Epididymides were obtained from each donor. Rat epididymal sperm was diluted in Ham's F10 plus raffinose, Ham's F10 plus trehalose, Ham's F10 plus fructose, and Ham's F10 medium for control purposes. Thereafter, the extended sperm were chilled and stored in liquid form at 4 degrees C. Sperm motility, morphological abnormalities and DNA damage were determined at 0 and 12 h after chilling. No significant difference was observed in any of the parameters evaluated at 0 h, before storage (P > 0.05). After 12 h of storage, all sugar additives led to statistically higher motility, normal sperm morphology and DNA integrity in comparison to the control group. Raffinose gave the best motility percentages (32.86 +/- 1.84%) after 12 h of storage at 4 degrees C, compared to the other groups (P < 0.001). In conclusion, Raffinose, trehalose and fructose provided a better protection of sperm functional parameters against chilling injury, in comparison to the control group.     The effect of cysteine and glutathione on sperm and oxidative stress parameters of post-thawed bull semen

    The aim of this study was to determine the effects of antioxidants such as reduced glutathione (GSH) and cysteine in Laiciphose (R) extender on semen parameters, fertilizing ability, lipid peroxidation (LPO) level and glutathione peroxidise (GPx) activity of post-thawed bull semen. Totally 54 ejaculates of three bulls were used in the study. Five groups, namely; GSH (0.5 and 2 mM), cysteine (5 and 10 mM) and control group, were conducted to test the antioxidants in Laiciphose (R). Insemination doses were processed that each 0.25-mL straw contained 15 x 10(6) sperm. The addition of antioxidants did not present any significant effect on the percentages of post-thaw sperm morphology (acrosome and total abnormalities), subjective, CASA and progressive motilities, as well as sperm motility characteristics (VAP, VSL, VCL, LIN and ALH), compared to the control groups (P > 0.05). GSH 0.5 mM (55.5 +/- 7.38%) and cysteine 10 mM (48 +/- 5.65%) led to lower rates of DNA damage, compared to control (P 0.05). The results of this study may provide an useful information for the future studies in this area. So, further studies could be suggested to achieve better information in terms of the DNA damage and fertilizing capacity of bull sperm frozen with effective antioxidants.     This study was conducted to determine the effects of methionine, inositol and carnitine on sperm (motility, abnormality, DNA integrity and in vivo fertility) and oxidative stress parameters (lipid peroxidation, total glutathione and antioxidant potential levels) of bovine semen after the freeze-thawing process. Nine ejaculates, collected with the aid of an artificial vagina twice a week from each Simmental bovine, were included in the study. Each ejaculate, splitted into seven equal groups and diluted in Tris-based extender containing methionine (2.5 and 7.5 mM), carnitine (2.5 and 7.5 mM), inositol (2.5 and 7.5 mM) and no additive (control), was cooled to 5 degrees C and then frozen in 0.25 ml straws. Frozen straws were then thawed individually at 37 degrees C for 20 s in a water bath for the evaluation
