    The GPR55 agonist lysophosphatidylinositol acts as an intracellular messenger and bidirectionally modulates Ca2+-activated large-conductance K+ channels in endothelial cells

    Lysophospholipids are known to serve as intra- and extracellular messengers affecting many physiological processes. Lysophosphatidylinositol (LPI), which is produced in endothelial cells, acts as an endogenous agonist of the orphan receptor, G protein-coupled receptor 55 (GPR55). Stimulation of GPR55 by LPI evokes an intracellular Ca2+ rise in several cell types including endothelial cells. In this study, we investigated additional direct, receptor-independent effects of LPI on endothelial large-conductance Ca2+ and voltage-gated potassium (BKCa) channels. Electrophysiological experiments in the inside-out configuration revealed that LPI directly affects the BKCa channel gating properties. This effect of LPI strictly depended on the presence of Ca2+ and was concentration-dependent, reversible, and dual in nature. The modulating effects of LPI on endothelial BKCa channels correlated with their initial open probability (Po): stimulation at low Po (<0.3) and inhibition at high Po levels (>0.3). In the whole-cell configuration, LPI in the pipette facilitated membrane hyperpolarization in response to low (0.1–2 μM) histamine concentrations. In contrast, LPI counteracted membrane hyperpolarization in response to supramaximal cell stimulation with histamine. These results highlight a novel receptor-independent and direct bidirectional modulation of BKCa channels by LPI on endothelial cells. We conclude that LPI via this mechanism serves as an important modulator of endothelial electrical responses to cell stimulation

    Commitment of cultural minorities in organizations:Effects of leadership and pressure to conform

    PURPOSE: In this study, we investigated the commitment of cultural minorities and majorities in organizations. We examined how contextual factors, such as pressure to conform and leadership styles, affect the commitment of minority and majority members. DESIGN/METHODOLOGY/APPROACH: A field study was conducted on 107 employees in a large multinational corporation. FINDINGS: We hypothesize and found that cultural minorities felt more committed to the organization than majority members, thereby challenging the existing theoretical view that cultural minorities will feel less committed. We also found that organizational pressure to conform and effective leadership increased the commitment of minorities. IMPLICATIONS: Our findings indicate that organizational leaders and researchers should not only focus on increasing and maintaining the commitment of minority members, but should also consider how majority members react to cultural socialization and integration processes. The commitment of minority members can be further enhanced by effective leadership. ORIGINALITY/VALUE: In this study, we challenge the existing theoretical view based on similarity attraction theory and relational demography theory, that cultural minorities would feel less committed to the organization. Past research has mainly focused on minority groups, thereby ignoring the reaction of the majority to socialization processes. In this study, we show that cultural minorities can be more committed than majority members in organizations. Therefore, the perceptions of cultural majority members of socialization processes should also be considered in research on cultural diversity and acculturation

    Aim: Deficits in facial affect recognition are well documented in schizophrenia, and have been associated with reduced social functioning and interpersonal difficulties. The aim of the present study was to test the possibility that facial affect recognition deficits represent an endophenotypic marker of schizophrenia liability by testing this capacity in individuals with the predisposition to symptoms of schizophrenia. Methods: Eight hundred and fortythree psychologically healthy participants completed the Schizotypal Personality Questionnaire of which 28 scoring in the upper 15% (highschizotypy group) and 28 scoring in the lower 15% (low-schizotypy group) completed measures of facial affect identification, facial affect discrimination, facial identity recognition, and a measure of negative affect. Results: After controlling for group differences in negative affect and facial identity recognition, negative (but not positive or disorganized) aspects of schizotypy were found to be significantly associated with reduced facial affect discrimination and facial affect recognition accuracy, and in particular, difficulties with the identification of negative emotions. Conclusions: These results provide limited support for the potential trait status of facial affect recognition deficits in schizophrenia and schizophrenia spectrum disorders, and suggest that these deficits may be particularly associated with the predisposition to negative symptoms of schizophrenia
