    Using Temporal Self-Regulation Theory to understand healthy and unhealthy eating intentions and behaviour

    Objectives: The present research investigated whether Temporal Self-Regulation Theory (TST) can be used to help understand healthy and unhealthy eating intentions and behaviour. Design: A prospective design with two waves of data collection one week apart. Method: An online survey measured the key components of TST (i.e., connectedness, timing and valence beliefs, intentions, past behaviour, habit strength, perceived environmental cues, and self-control) with respect to eating fruit and vegetables (F&V; N = 133) or unhealthy snacks (N = 125). Eating behaviour was assessed one week later. Results: The components of TST explained significant amounts of the variance in intentions and behaviour for intake of F&Vs (22% and 64%, respectively) and unhealthy snacks (18% and 35%, respectively). Beliefs about positive and negative short-term outcomes significantly predicted intentions to perform both behaviours. Intentions and past behaviour significantly predicted consumption of F&Vs, and past behaviour moderated the relationship between intention and behaviour which became stronger as past behaviour increased. Past behaviour and habit strength significantly predicted unhealthy snacking. Conclusions: The findings suggest that TST may be a useful framework for understanding eating intentions and behaviour. However, research did not find support for all of the hypothesised relationships (e.g., self-regulatory capacity did not significantly predict eating behaviour and also failed to moderate the relationships between intentions and behaviour). Research using alternative measures of self-regulatory capacity, along with experimental manipulations of TST variables, may be needed to further understand eating intentions and behaviour

    Seminal fluid protein divergence among populations exhibiting postmating prezygotic reproductive isolation

    Despite holding a central role in fertilization, reproductive traits often show elevated rates of evolution and diversification. The rapid evolution of seminal fluid proteins (Sfps) within populations is predicted to cause mis‐signalling between the male ejaculate and the female during and after mating resulting in postmating prezygotic (PMPZ) isolation between populations. Crosses between Drosophila montana populations show PMPZ isolation in the form of reduced fertilization success in both noncompetitive and competitive contexts. Here we test whether male ejaculate proteins produced in the accessory glands or ejaculatory bulb differ between populations using liquid chromatography tandem mass spectrometry. We find more than 150 differentially abundant proteins between populations that may contribute to PMPZ isolation, including a number of proteases, peptidases and several orthologues of Drosophila melanogaster Sfps known to mediate fertilization success. Males from the population that elicit the stronger PMPZ isolation after mating with foreign females typically produced greater quantities of Sfps. The accessory glands and ejaculatory bulb show enrichment for different gene ontology (GO) terms and the ejaculatory bulb contributes more differentially abundant proteins. Proteins with a predicted secretory signal evolve faster than nonsecretory proteins. Finally, we take advantage of quantitative proteomics data for three Drosophila species to determine shared and unique GO enrichments of Sfps between taxa and which potentially mediate PMPZ isolation. Our study provides the first high‐throughput quantitative proteomic evidence showing divergence of reproductive proteins between populations that exhibit PMPZ isolation

    Final Report On Submerged Cylinder Wave Energy Device

    A first overall assessment of the submerged cylinder device has been prepared, from which at outline design for the principal components has been produced. A budget estimate for this design has been made, including allowances for annual operation and maintenance charges. The efficiency characteristics of the device have been determined from selected tank tests. With these efficiencies, the energy captured by each cylinder of an array has been estimated. Allowances have been made for the probable energy losses in each component through to the National network at Perth. On the basis of the preliminary specification for the device agreed in June 1979, at the end of Stage 1 of this Contract, the price of electricity delivered to the network is estimated to be 11. 4 p/kWh. This assumes a 5% discount rate and a 20 year operating life. A number of improvements to the reference design are already clear. Principally its tuned frequency should be increased so that the efficiency characteristics match the wave spectrum more closely. This single improvement in gross output appears sufficient to reduce the unit cost to about 8.5 p/kWh because of the significance of annual maintenance charges on the net annual revenue from sales of electricity. There is considerable scope to improve the design. Other power take off and energy transfer techniques deserve attention, and there is scope to rationalise the mooring and foundation arrangements. Because of the nature of its working environment, the same philosophy of using proven components or reasonable derivatives therefrom should be retained. The stable behaviour of the device in all sea conditions studied, and its high efficiency in the more persistent waves, confirm that this further step towards a comprehensive design of the basically simple, efficient and robust system is justified

    Illuminating the bacterial microbiome of Australian ticks with 16S and Rickettsia-specific next-generation sequencing

    Next-generation sequencing (NGS) studies show that mosquito and tick microbiomes influence the transmission of pathogens, opening new avenues for vector-borne pathogen control. Recent microbiological studies of Australian ticks highlight fundamental knowledge gaps of tick-borne agents. This investigation explored the composition, diversity and prevalence of bacteria in Australian ticks (n = 655) from companion animals (dogs, cats and horses). Bacterial 16S NGS was used to identify most bacterial taxa and a Rickettsia-specific NGS assay was developed to identify Rickettsia species that were indistinguishable at the V1-2 regions of 16S. Sanger sequencing of near full-length 16S was used to confirm whether species detected by 16S NGS were novel. The haemotropic bacterial pathogens Anaplasma platys, Bartonella clarridgeiae, “Candidatus Mycoplasma haematoparvum” and Coxiella burnetii were identified in Rhipicephalus sanguineus (s.l.) from Queensland (QLD), Western Australia, the Northern Territory (NT), and South Australia, Ixodes holocyclus from QLD, Rh. sanguineus (s.l.) from the NT, and I. holocyclus from QLD, respectively. Analysis of the control data showed that cross-talk compromises the detection of rare species as filtering thresholds for less abundant sequences had to be applied to mitigate false positives. A comparison of the taxonomic assignments made with 16S sequence databases revealed inconsistencies. The Rickettsia-specific citrate synthase gene NGS assay enabled the identification of Rickettsia co-infections with potentially novel species and genotypes most similar (97.9–99.1%) to Rickettsia raoultii and Rickettsia gravesii. “Candidatus Rickettsia jingxinensis” was identified for the first time in Australia. Phylogenetic analysis of near full-length 16S sequences confirmed a novel Coxiellaceae genus and species, two novel Francisella species, and two novel Francisella genotypes. Cross-talk raises concerns for the MiSeq platform as a diagnostic tool for clinical samples. This study provides recommendations for adjustments to Illuminaʼs 16S metagenomic sequencing protocol that help track and reduce cross-talk from cross-contamination during library preparation. The inconsistencies in taxonomic assignment emphasise the need for curated and quality-checked sequence databases

    On the Two Species Asymmetric Exclusion Process with Semi-Permeable Boundaries

    We investigate the structure of the nonequilibrium stationary state (NESS) of a system of first and second class particles, as well as vacancies (holes), on L sites of a one-dimensional lattice in contact with first class particle reservoirs at the boundary sites; these particles can enter at site 1, when it is vacant, with rate alpha, and exit from site L with rate beta. Second class particles can neither enter nor leave the system, so the boundaries are semi-permeable. The internal dynamics are described by the usual totally asymmetric exclusion process (TASEP) with second class particles. An exact solution of the NESS was found by Arita. Here we describe two consequences of the fact that the flux of second class particles is zero. First, there exist (pinned and unpinned) fat shocks which determine the general structure of the phase diagram and of the local measures; the latter describe the microscopic structure of the system at different macroscopic points (in the limit L going to infinity in terms of superpositions of extremal measures of the infinite system. Second, the distribution of second class particles is given by an equilibrium ensemble in fixed volume, or equivalently but more simply by a pressure ensemble, in which the pair potential between neighboring particles grows logarithmically with distance. We also point out an unexpected feature in the microscopic structure of the NESS for finite L: if there are n second class particles in the system then the distribution of first class particles (respectively holes) on the first (respectively last) n sites is exchangeable.Comment: 28 pages, 4 figures.     Fast-ion redistribution and loss due to edge perturbations in the ASDEX Upgrade, DIII-D and KSTAR tokamaks

    The impact of edge localized modes (ELMs) and externally applied resonant and non-resonant magnetic perturbations (MPs) on fast-ion confinement/transport have been investigated in the ASDEX Upgrade (AUG), DIII-D and KSTAR tokamaks. Two phases with respect to the ELM cycle can be clearly distinguished in ELM-induced fast-ion losses. Inter-ELM losses are characterized by a coherent modulation of the plasma density around the separatrix while intra-ELM losses appear as well-defined bursts. In high collisionality plasmas with mitigated ELMs, externally applied MPs have little effect on kinetic profiles, including fast-ions, while a strong impact on kinetic profiles is observed in low-collisionality, low q 95 plasmas with resonant and non-resonant MPs. In low-collisionality H-mode plasmas, the large fast-ion filaments observed during ELMs are replaced by a loss of fast-ions with a broad-band frequency and an amplitude of up to an order of magnitude higher than the neutral beam injection prompt loss signal without MPs. A clear synergy in the overall fast-ion transport is observed between MPs and neoclassical tearing modes. Measured fast-ion losses are typically on banana orbits that explore the entire pedestal/scrape-off layer.     This article brings together Professor Donald Nicolson's intellectual interest in professional legal ethics and his long-standing involvement with law clinics both as an advisor at the University of Cape Town and Director of the University of Bristol Law Clinic and the University of Strathclyde Law Clinic. In this article he looks at how legal education may help start this process of character development, arguing that the best means is through student involvement in voluntary law clinics. And here he builds upon his recent article which argues for voluntary, community service oriented law clinics over those which emphasise the education of students

    Caste development and reproduction: a genome-wide analysis of hallmarks of insect eusociality

    The honey bee queen and worker castes are a model system for developmental plasticity. We used established expressed sequence tag information for a Gene Ontology based annotation of genes that are differentially expressed during caste development. Metabolic regulation emerged as a major theme, with a caste-specific difference in the expression of oxidoreductases vs. hydrolases. Motif searches in upstream regions revealed group-specific motifs, providing an entry point to cis-regulatory network studies on caste genes. For genes putatively involved in reproduction, meiosis-associated factors came out as highly conserved, whereas some determinants of embryonic axes either do not have clear orthologs (bag of marbles, gurken, torso), or appear to be lacking (trunk) in the bee genome. Our results are the outcome of a first genome-based initiative to provide an annotated framework for trends in gene regulation during female caste differentiation (representing developmental plasticity) and reproduction

    Tomato: a crop species amenable to improvement by cellular and molecular methods

    Tomato is a crop plant with a relatively small DNA content per haploid genome and a well developed genetics. Plant regeneration from explants and protoplasts is feasable which led to the development of efficient transformation procedures. In view of the current data, the isolation of useful mutants at the cellular level probably will be of limited value in the genetic improvement of tomato. Protoplast fusion may lead to novel combinations of organelle and nuclear DNA (cybrids), whereas this technique also provides a means of introducing genetic information from alien species into tomato. Important developments have come from molecular approaches. Following the construction of an RFLP map, these RFLP markers can be used in tomato to tag quantitative traits bred in from related species. Both RFLP's and transposons are in the process of being used to clone desired genes for which no gene products are known. Cloned genes can be introduced and potentially improve specific properties of tomato especially those controlled by single genes. Recent results suggest that, in principle, phenotypic mutants can be created for cloned and characterized genes and will prove their value in further improving the cultivated tomato.
