    Vertical flows and mass flux balance of sunspot umbral dots

    A new Stokes inversion technique that greatly reduces the effect of the spatial point spread function of the telescope is used to constrain the physical properties of umbral dots (UDs). The depth-dependent inversion of the Stokes parameters from a sunspot umbra recorded with Hinode SOT/SP revealed significant temperature enhancements and magnetic field weakenings in the core of the UDs in deep photospheric layers. Additionally, we found upflows of around 960 m/s in peripheral UDs (i.e., UDs close to the penumbra) and \approx 600 m/s in central UDs. For the first time, we also detected systematic downflows for distances larger than 200 km from the UD center that balance the upflowing mass flux. In the upper photosphere, we found almost no difference between the UDs and their diffuse umbral background.Comment: Accepted for publication in Astronomy and Astrophysic

    spFRET Using Alternating Excitation and FCS Reveals Progressive DNA Unwrapping in Nucleosomes

    AbstractAccessibility to DNA wrapped in nucleosomes is essential for nuclear processes such as DNA transcription. Large conformational changes in nucleosome structure are required to facilitate protein binding to target sites within nucleosomal DNA. Transient unwrapping of DNA from nucleosome ends can provide an intrinsic exposure of wrapped DNA, allowing proteins to bind DNA that would otherwise be occluded in the nucleosome. The molecular details underlying these mechanisms remain to be resolved. Here we show how DNA unwrapping occurs progressively from both nucleosome ends. We performed single-pair fluorescence resonance energy transfer (spFRET) spectroscopy with alternating laser excitation (ALEX) on nucleosomes either in free solution or confined in a gel after PAGE separation. We combined ALEX-spFRET with a correlation analysis on selected bursts of fluorescence, to resolve a variety of unwrapped nucleosome conformations. The experiments reveal that nucleosomes are unwrapped with an equilibrium constant of ∼0.2–0.6 at nucleosome ends and ∼0.1 at a location 27 basepairs inside the nucleosome, but still remain stably associated. Our findings, obtained using a powerful combination of single-molecule fluorescence techniques and gel electrophoresis, emphasize the delicate interplay between DNA accessibility and condensation in chromatin

    Online users are increasingly exposed to chatbots as one form of AI-enabled media technologies, employed for persuasive purposes, e.g., making product/service recommendations. However, the persuasive potential of chatbots has not yet been fully explored. Using an online experiment (N = 242), we investigate the extent to which communicating with a stand-alone chatbot influences affective and behavioral responses compared to interactive Web sites. Several underlying mechanisms are studied, showing that enjoyment is the key mechanism explaining the positive effect of chatbots (vs. Web sites) on recommendation adherence and attitudes. Contrary to expectations, perceived anthropomorphism seems not to be particularly relevant in this comparison

    Elucidation of in Vitro Chlorinated Tyrosine Adducts in Blood Plasma as Selective Biomarkers of Chlorine Exposure

    [Image: see text] Chlorine is a widely available industrial chemical and involved in a substantial number of cases of poisoning. It has also been used as a chemical warfare agent in military conflicts. To enable forensic verification, the persistent biomarkers 3-chlorotyrosine and 3,5-dichlorotyrosine in biomedical samples could be detected. An important shortfall of these biomarkers, however, is the relatively high incidence of elevated levels of chlorinated tyrosine residues in individuals with inflammatory diseases who have not been exposed to chlorine. Therefore, more reliable biomarkers are necessary to distinguish between endogenous formation and exogeneous exposure. The present study aims to develop a novel diagnostic tool for identifying site-specific chlorinated peptides as a more unambiguous indicator of exogeneous chlorine exposure. Human blood plasma was exposed in vitro to various chlorine concentrations, and the plasma proteins were subsequently digested by pronase, trypsin, or pepsin. After sample preparation, the digests were analyzed by liquid chromatography tandem mass spectrometry (LC–MS/MS) and liquid chromatography high-resolution tandem mass spectrometry (LC–HRMS/MS). In line with other studies, low levels of 3-chlorotyrosine and 3,5-dichlorotyrosine were found in blank plasma samples in this study. Therefore, 50 site-specific biomarkers were identified, which could be used as more unambiguous biomarkers for chlorine exposure. Chlorination of the peptides TY*ETTLEK, Y*KPGQTVK, Y*QQKPGQAPR, HY*EGSTVPEK, and Y*LY*EIAR could already be detected at moderate in vitro chlorine exposure levels. In addition, the latter two peptides were found to have dichlorinated fragments. Especially, Y*LY*EIAR, with a distinct chlorination pattern in the MS spectra, could potentially be used to differentiate exogeneous exposure from endogenous causes as other studies reported that this part of human serum albumin is nitrated rather than chlorinated under physiological conditions. In conclusion, trypsin digestion combined with high-resolution MS analysis of chlorinated peptides could constitute a valuable technique for the forensic verification of exposure to chlorine

    High-order aberration compensation with Multi-frame Blind Deconvolution and Phase Diversity image restoration techniques

    Context. For accurately measuring intensities and determining magnetic field strengths of small-scale solar (magnetic) structure, knowledge of and compensation for the point spread function is crucial. For images recorded with the Swedish 1-meter Solar Telescope, restoration with Multi-Frame Blind Deconvolution and Joint Phase Diverse Speckle methods lead to remarkable improvements in image quality but granulation contrasts that are too low, indicating additional stray light. Aims. We propose a method to compensate for stray light from high-order atmospheric aberrations not included in MFBD and JPDS processing. Methods. To compensate for uncorrected aberrations, a reformulation of the image restoration process is proposed that allows the average effect of hundreds of high-order modes to be compensated for by relying on Kolmogorov statistics for these modes. The applicability of the method requires simultaneous measurements of Fried's parameter r0. The method is tested with simulations as well as real data and extended to include compensation for conventional stray light. Results. We find that only part of the reduction of granulation contrast in SST images is due to uncompensated high-order aberrations. The remainder is still unaccounted for and attributed to stray light from the atmosphere, the telescope with its re-imaging system and to various high-altitude seeing effects. Conclusions. We conclude that statistical compensation of high-order modes is a viable method to reduce the loss of contrast occurring when a limited number of aberrations is explicitly compensated for with MFBD and JPDS processing. We show that good such compensation is possible with only 10 recorded frames. The main limitation of the method is that already MFBD and JPDS processing introduces high-order compensation that, if not taken into account, can lead to over-compensation.Comment: in press in Astronomy & Astrophysic

    Emission Lines in the Spectrum of the 3He Star 3 Cen A

    Emission in the 4d - 4f transitions of MnII (multiplet 13, 6122-6132 Ang), in the 4f - 6g transitions of PII, and in 6149.5 Ang of HgII has been detected in the spectrum of the helium weak star 3 Centauri A (B5 III-IVp). Weaker emission from the same MnII multiplet is also seen in the hot, mild HgMn star 46 Aquila (B9 III).It is suggested that the emission is of photospheric origin and may be evidence for the stratification of manganese, phosphorus and mercury in the photosphere of 3 CenA, and of manganese in 46Aql.Comment: 16 pages, 3 figure
