    Significance of the presence of antibiotics on the microbial consortium in wastewater - The case of nitrofurantoin and furazolidone.

    Antibiotics in wastewater leads to migration of pollutants and disrupts natural processes of mineralization of organic matter. In order to understand the mechanism of this, research was undertaken on the influence of nitrofurantoin (NFT) and furazolidone (FZD), on the behaviour of a consortium of microorganisms present in a model wastewater in a bioreactor. Our study confirmed biodegradation of the antibiotics by the microbial consortium, with the degradation efficiency within 10 days of 65% for FZD, but only 20% for NFT. The kinetic study proved that the presence of analysed antibiotics had no adverse effect on the microbes, but the consortium behaviour differ significantly with the NFT reducing the consumption of organic carbon in wastewater and increasing the production of extracellular biopolymeric and volatile organic compounds, and the FZD reducing assimilation of other carbon sources to a less extent, at the expense of cellular focus on biodegradation of this antibiotic

    The influence of the time-of-day administration of the drug on the pharmacokinetics of sunitinib in rabbits

    OBJECTIVES: At present it is known that the adjustment of the anticancer therapy to the circadian rhythms in tissues reduces the toxicity of the treatment. Chronotherapy also increases the efficacy of the anticancer treatment, which has been proved for many drugs. Sunitinib is a tyrosine kinase inhibitor, which is broadly used for the treatment of numerous cancers. The aim of the study was a comparison of the concentrations and pharmacokinetics of sunitinib after a single administration to rabbits at 08:00 (control group) and 20:00. Additionally, the effect of sunitinib on glucose levels was investigated. MATERIALS AND METHODS: The research was carried out on two groups of rabbits: I08:00, a group with the drug administered at 08:00 (n=8) and II20:00, a group with the drug administered at 20:00 (n=8). The rabbits were treated with sunitinib at an oral dose of 25 mg. Plasma concentrations of sunitinib and its metabolite (SU12662) were measured with a validated HPLC method with UV detection. RESULTS: The comparison of the sunitinib Cmax and AUC0-t in the group with sunitinib administered at 20:00 with the control group gave the ratios of 2.20 (90% confidence interval (CI) (2.17; 2.22) and 1.64 (1.61; 1.68), respectively. Statistically significant differences between the groups under analysis were revealed for Cmax (p \u3c 0.0001), AUC0-t (p = 0.0079), AUC0-∞ (p = 0.0149), and tmax (p = 0.0085). The mean glycemia drop was higher in group I08:00. than in group II20:00 (22.7% vs. 14.3%; p = 0.0622). The glycemia values returned to the initial values in 24 h after the administration of the drug in both groups. CONCLUSIONS: The research proved a significant influence of the time-of-day administration on the pharmacokinetics of sunitinib

    Assessing circadian rhythms in propofol PK and PD during prolonged infusion in ICU patients

    This study evaluates possible circadian rhythms during prolonged propofol infusion in patients in the intensive care unit. Eleven patients were sedated with a constant propofol infusion. The blood samples for the propofol assay were collected every hour during the second day, the third day, and after the termination of the propofol infusion. Values of electroencephalographic bispectral index (BIS), arterial blood pressure, heart rate, blood oxygen saturation and body temperature were recorded every hour at the blood collection time points. A two-compartment model was used to describe propofol pharmacokinetics. Typical values of the central and peripheral volume of distribution and inter-compartmental clearance were VC = 27.7 l, VT = 801 l, and CLD = 2.73 l/min. The systolic blood pressure (SBP) was found to influence the propofol metabolic clearance according to Cl (l/min) = 2.65·(1 − 0.00714·(SBP − 135)). There was no significant circadian rhythm detected with respect to propofol pharmacokinetics. The BIS score was assessed as a direct effect model with EC50 equal 1.98 mg/l. There was no significant circadian rhythm detected within the BIS scores. We concluded that the light–dark cycle did not influence propofol pharmacokinetics and pharmacodynamics in intensive care units patients. The lack of night–day differences was also noted for systolic blood pressure, diastolic blood pressure and blood oxygenation. Circadian rhythms were detected for heart rate and body temperature, however they were severely disturbed from the pattern of healthy patients

    BACKGROUND: The aim of this open-label, randomized controlled trial conducted in four African countries (Madagascar, Niger, Central African Republic, and Senegal) is to compare three strategies of renutrition for moderate acute malnutrition (MAM) in children based on modulation of the gut microbiota with enriched flours alone, enriched flours with prebiotics or enriched flours coupled with antibiotic treatment. METHODS: To be included, children aged between 6 months and 2 years are preselected based on mid-upper-arm circumference (MUAC) and are included based on a weight-for-height Z-score (WHZ) between - 3 and - 2 standard deviations (SD). As per current protocols, children receive renutrition treatment for 12 weeks and are assessed weekly to determine improvement. The primary endpoint is recovery, defined by a WHZ >/= - 1.5 SD after 12 weeks of treatment. Data collected include clinical and socioeconomic characteristics, side effects, compliance and tolerance to interventions. Metagenomic analysis of gut microbiota is conducted at inclusion, 3 months, and 6 months. The cognitive development of children is evaluated in Senegal using only the Developmental Milestones Checklist II (DMC II) questionnaire at inclusion and at 3, 6, and 9 months. The data will be correlated with renutrition efficacy and metagenomic data. DISCUSSION: This study will provide new insights for the treatment of MAM, as well as original data on the modulation of gut microbiota during the renutrition process to support (or not) the microbiota hypothesis of malnutrition.     Inverse Gas Chromatography in the Examination of Modified Highly Dispersed Silicas

    Silicas having a highly extended outer surface, i.e. silicas precipitated from sodium metasilicate solutions, were used as the raw materials in the present study. Highly dispersed silicas were modified with silane coupling agents to decrease their surface hydrophilicity. The modification procedure was optimized, e.g. by the change of the modifier amounts added. Modified silicas were characterized by means of inverse gas chromatography at infinite dilution through the estimation of their dispersive and acid–base characteristics. The dispersive component of the surface free energy γ d s , the specific increment of the free energy of adsorption ΔG S , and the parameters K D and K A characterizing the ability of the surface to act as an electron donor and an electron acceptor, respectively, as well as S C = K D /K A , were used in this study. The physicochemical status of the modifier monolayer was described by a solubility parameter and its components. The influence of the surface character of the raw material, the modifier structure and its amount on the surface properties of the modified material is presented and discussed
