Influence of potassium nutrition and the nitrate ammonium ratio on the putrescine and spermidine contents in banana vitroplants

Potassium (K) is required in high doses by the banana (Musa sp.) plant and interacts with other nutrient elements in which banana tissues are maintained under in vitro condition as a consequence modifications in the plant metabolism take place mainly in nitrogen (N) compounds, such as proteins, amino acids, and secondary compounds. When K is present in concentrations lower than that required, diamines such as putrescine and poliamines are formed. This metabolic disorder can also be correlated with the presence of different inorganic N forms, such as nitrate (NO3) and ammonium (NH4), and the ratios between both ions as well. In order to follow the physiological performance of the interrelationships, K/putrescine and of the NO3/NH4 ratio in the tissue of banana vitroplantlets, shoot apex of two banana cvs. Nanica and Prata Ana were maintained in modified MS medium in the presence of six different doses of K: 5, 10, 15, 20, 25, and 30 mM. After the period of tissue proliferation the cultures were transferred to rooting media containing the same different K doses. Dry matter, K, putrescine, and spermidine contents and their accumulation were determined in the shoots and roots of the vitroplantlets and in the shoot apex of the explant donor cultivar as well as the corresponding values for the whole vitroplantlets calculated. The data were statistically analyzed. The contents and accumulations of putrescine and spermidine in banana tissues were enhanced as K concentration decreased in the medium: four times (0.19% of the dry matter) for cv. Nanica and eight times (0.25% of the dry matter) for cv. Prata Ana. This behavior was not only related to the K depletion but to the NO3/NH4 ratio as well

Carbon isotope composition as a tool to control the quality of herbs and medicinal plants

Isotope screening is a simple test for determining the photosynthetic pathway used by plants. The scope of this work was to classify the photosynthetic type of some herbs and medicinal plants through studies of the carbon isotope composition (δ13C). Also, we propose the use of carbon isotope composition as a tool to control the quality of herbs and medicinal plants. For studies of δ13C, δ 13C‰ = [R (sample)/R (standard) - 1] × 10-3, dry leaves powdered in cryogenic mill were analyzed in a mass spectrometer coupled with an elemental analyzer for determining the ratio R = 13CO2/12CO2. In investigation of δ13C of 55 species, 23 botanical families, and 44 species possessed a C3 photosynthetic type. Six species found among the botanical families Euphorbiaceae and Poaceae were C4 plants, and 5 species found among the botanical families Agavaceae, Euphorbiaceae, and Liliaceae possessed CAM-type photosynthesis. Carbon isotope composition of plants can be used as quality control of herbs and medicinal plants, allowing the identification of frauds or contaminations. Also, the information about the photosynthetic type found for these plants can help in introducing and cultivating exotic and wild herbs and medicinal plants

Tolerância à salinidade em feijão (Phaseolus vulgaris L) Salt tolerance in bean (Paseolus vulgaris) cell culture

Paralelamente, observou-se aumento significativo no conteúdo de prolina e atividade da enzima peroxidase.<br>One of the applications of the tissue culture technique in plant improvement is the identification of cell lines which show salinity tolerance. Several authors were able to obtain saline stress-tolerant cell lines and show that mechanisms of tolerance to salts have a strong correlation between this phenomenon and a high macromolecule concentration in plant tissues. Callus obtained from embrionic axis of Phaseolus vulgarís cv. IAC carioca in solid medium, supplemented with 0 to 60 mM NaCl, as the salt treatment, were used. Callus harvesting was done on the 13th day, when they were processed for relative growth, protein, proline content and peroxidase acivity. The results show both, a decrease of the relative growth and of protein content in response to the NaCl treatment, as compared to controls. However, there was a significant increase on the proline content and on the peroxidase activity
