    Salvinorin A 1, a psychoactive neoclerodane diterpenoid from the Mexican sage S. divinorum, has gained interest as a selective kappa-opioid receptor agonist. Non-racemic 3-furylamines 9a and 9b have been prepared from (+)-pseudoephedrine and (-)-ephedrine for application in the stereoselective synthesis of the ketone ring of 1. Diels-Alder reaction of 9b with methyl acrylate in aqueous media, followed by selective ether bridge cleavage, has allowed access to the cyclohexenone 17 with preservation of stereochemistry at C2. A model route to the lactone ring has also been achieved through a one-pot deconjugation/esterification procedure of 2-bromocrotonyl chloride 20 to the furyl alcohol 19 followed by Reformatski-mediated ring closure

    There is an increasing body of evidence which suggests that IL-4 plays a role in the pathogenesis of TB, but a general consensus on its role remains elusive. We have previously published data from a cohort of Ethiopian TB patients, their contacts, and community controls suggesting that enhanced IL-4 production is associated with infection with M. tuberculosis, rather than overt disease and that long-term protection in infected community controls is associated with co-production of the IL-4 antagonist IL-4d2, alongside elevated IL-4. Here, for the first time, we compare data on expression of IFN-gamma, IL-4 and IL-4delta2 over time in TB patients and their household contacts. During the follow-up period, the TB patients completed therapy and ceased to display TB-like symptoms. This correlated with a decrease in the relative amount of IL-4 expressed. Over the same period, the clinical status of some of their contacts also changed, with a number developing TB-like symptoms or clinically apparent TB. IL-4 expression was disproportionately increased in this group. The findings support the hypothesis that elevated IL-4 production is generally associated with infection, but that TB disease is associated with a relatively increased expression of IL-4 compared to IFN-gamma and IL-4delta2. However, the data also suggest that there are no clear-cut differences between groups: the immune response over time appears to include changes in the expression of IFN-gamma, IL-4 and IL-4delta2, and it is the relative, not absolute levels of cytokine expression that are characteristic of clinical status

    Forward and hybrid path-integral methods in photoelectron holography: Sub-barrier corrections, initial sampling, and momentum mapping

    We construct two strong-field path integral methods with full Coulomb distortion, in which the quantum pathways are mimicked by interfering electron orbits: the rate-based CQSFA (R-CQSFA) and the hybrid forward-boundary CQSFA (H-CQSFA). The methods have the same starting point as the standard Coulomb quantum-orbit strong-field approximation (CQSFA), but their implementation does not require preknowledge of the orbits' dynamics. These methods are applied to ultrafast photoelectron holography. In the rate-based method, electron orbits are forward propagated and we derive a nonadiabatic ionization rate from the CQSFA, which includes sub-barrier Coulomb corrections and is used to weight the initial orbit ensemble. In the H-CQSFA, the initial ensemble provides initial guesses for a subsequent boundary problem and serves to include or exclude specific momentum regions, but the ionization probabilities associated with individual trajectories are computed from sub-barrier complex integrals. We perform comparisons with the standard CQSFA and ab initio methods, which show that the standard, purely boundary-type implementation of the CQSFA leaves out whole sets of trajectories. We show that the sub-barrier Coulomb corrections broaden the resulting photoelectron momentum distributions (PMDs) and improve the agreement of the R-CQSFA with the H-CQSFA and other approaches. We probe different initial sampling distributions, uniform and otherwise, and their influence on the PMDs. We find that initial biased sampling emphasizes rescattering ridges and interference patterns in high-energy ranges, while an initial uniform sampling guarantees accurate modeling of the holographic patterns near the ionization threshold or polarization axis. Our results are explained using the initial to final momentum mapping for different types of interfering trajectories

    SAHABI EURYGNATHOHIPPUS FEIBELI: ITS SYSTEMATIC, STRATIGRAPHIC, CHRONOLOGIC AND BIOGEOGRAPHIC CONTEXTS

    Sahabi, Libya is an important latest Miocene locality having yielded an extensive paleobotanical and vertebrate fauna. Amongst the fossil mammals there occurs an extensive, species diverse record of hipparionine horses. We develop here a complete record of Sahabi Eurygnathohippus feibeli now based on dental and postcranial material, in comparison to other equids from late Miocene equids from Europe, West Asia and Africa. We find that E. feibeli is the earliest recognized species of the predominantly African clade Eurygnathohippus, that its biogeographic range was Kenya, Ethiopia, Libya and Morocco, it ranged between 7.0 and 5.7 Ma and that it had deep-time evolutionary roots extending back to first occurring Old World hipparions. We further find that Eurygnathohippus was restricted to Africa until a more advanced member of the clade extended its range into the Indian Subcontinent during the late Pliocene, ca. 3.6-2.6 Ma
