    Fast polarizers installation for ECRH and ECE in TCV

    We report on the installation of fast polarizers for ECRH injection and ECE diagnostics, in the TCV tokamak. The main goal is to change the polarization during a plasma shot and react to changing conditions such as: plasma current and position, ECRH injection angles as well as ECE oblique observation angle and correlation ECE frequency. This will allow tracking of plasma variations, find optimum parameters to maximize ECRH absorption and improve the quality and consistency of ECE measurements. The polarization is varied using two fast polarizers, from General Atomics, positioned either in the evacuated transmission line for ECRH, or in a similar non-evacuated line near a real-time moveable directional antenna for ECE. A programmable driver is used to control a servo-motor and allows three operation modes during a plasma shot: fixed angle position, pre-programmed reference waveform and following of a real-time reference waveform. Preliminary tests of the motor controller achieved an 8 Hz bandwidth for a 4 +/-degrees amplitude motion and a 2.2 Hz bandwidth for a +/- 4 degrees amplitude motion.     Pharmacological screening using an FXN-EGFP cellular genomic reporter assay for the therapy of Friedreich ataxia

The presence of a GAA trinucleotide repeat expansion in the first intron of the FXN gene results in the inhibition of gene expression and an insufficiency of the mitochondrial protein frataxin. There is a correlation between expansion length, the amount of residual frataxin and the severity of disease. As the coding sequence is unaltered, pharmacological up-regulation of FXN expression may restore frataxin to therapeutic levels. To facilitate screening of compounds that modulate FXN expression in a physiologically relevant manner, we established a cellular genomic reporter assay consisting of a stable human cell line containing an FXN-EGFP fusion construct, in which the EGFP gene is fused in-frame with the entire normal human FXN gene present on a BAC clone. The cell line was used to establish a fluorometric cellular assay for use in high throughput screening (HTS) procedures. A small chemical library containing FDA-approved compounds and natural extracts was screened and analyzed. Compound hits identified by HTS were further evaluated by flow cytometry in the cellular genomic reporter assay. The effects on FXN mRNA and frataxin protein levels were measured in lymphoblast and fibroblast cell lines derived from individuals with FRDA and in a humanized GAA repeat expansion mouse model of FRDA.     Bending elasticity of a curved amphiphilic film decorated anchored copolymers: a small angle neutron scattering study

    Microemulsion droplets (oil in water stabilized by a surfactant film) are progressively decorated with increasing amounts of poly ethylene- oxide (PEO) chains anchored in the film by the short aliphatic chain grafted at one end of the PEO chain . The evolution of the bending elasticity of the surfactant film with increasing decoration is deduced from the evolution in size and polydispersity of the droplets as reflected by small angle neutron scattering. The optimum curvature radius decreases while the bending rigidity modulus remains practically constant. The experimental results compare well with the predictions of a model developed for the bending properties of a curved film decorated by non-adsorbing polymer chains, which takes into account, the finite curvature of the film and the free diffusion of the chains on the film.Comment: 30 June 200

    Electron Bernstein Wave Core Deposition via O-X-B Double Mode Conversion in TCV

    In the first results of electron Bernstein wave heating (EBWH) via O-X-B double mode conversion in a medium aspect ratio tokamak performed in TCV, the total power absorption efficiency was typically 60% and the deposition was made off-axis to avoid the central sawteeth perturbation that would prevent a clear detection of the deposition location. Central power deposition is however needed to maximize the global EBWH effects. Therefore, initial experiments have been started to optimize central deposition using an equatorial rather than poloidally oblique launch. A toroidal field scan provided promising central deposition results within a small database, in preparation for dedicated central high power deposition. The deposition location in presence of strong sawteeth could be determined from the slope break of soft X-ray traces at the EC power switch-ON times, in conditions where usual methods failed. Further analysis of these discharges will be presented. In particular, the difference between the experimental results and the non-relativistic calculations of deposition locations appears more significant than in the earlier off-axis deposition experiments, which provides a good opportunity for the simulation of relativistic ray propagation effects

    Phase Separation of Rigid-Rod Suspensions in Shear Flow

    We analyze the behavior of a suspension of rigid rod-like particles in shear flow using a modified version of the Doi model, and construct diagrams for phase coexistence under conditions of constant imposed stress and constant imposed strain rate, among paranematic, flow-aligning nematic, and log-rolling nematic states. We calculate the effective constitutive relations that would be measured through the regime of phase separation into shear bands. We calculate phase coexistence by examining the stability of interfacial steady states and find a wide range of possible ``phase'' behaviors.Comment: 23 pages 19 figures, revised version to be published in Physical Review

    Comparison of detachment in Ohmic plasmas with positive and negative triangularity

    Detachment is investigated using core density ramps for lower single null Ohmic L-mode plasmas across a wide range of upper, lower, and total triangularity (δ\delta) in the TCV tokamak. It is universally found that detachment is more difficult to access with negative triangularity (NT) shaping. The outer divertor leg of discharges with δ≈−0.3\delta\approx -0.3 could not be cooled below 5 eV using core density ramps alone. The behavior of the upstream plasma and geometrical divertor effects (e.g. a reduced connection length at negative lower triangularity) do not fully explain the challenges of detaching NT plasmas. Langmuir probe measurements of the target heat flux widths (λq\lambda_q) remained constant within 30% across an upper triangularity scan, while the spreading factor SS was found to be lower by up to 50% in NT, indicating a generally lower integral SOL width. An interesting pattern has been observed in the particle balance where the line-averaged core density was typically higher in NT discharges for a given fuelling rate. Conversely, the divertor neutral pressure and integrated particle content were typically lower for the same line-averaged density. This indicates that NT plasmas may be closer to the sheath-limited regime than their PT counterparts, which could explain why NT is more challenging to detach

    Dual hypothermic oxygenated machine perfusion in liver transplants donated after circulatory death

    Background: Experimental studies have suggested that end-ischaemic dual hypothermic oxygenated machine perfusion (DHOPE) may restore hepatocellular energy status and reduce reperfusion injury in donation after circulatory death (DCD) liver grafts. The aim of this prospective case-control study was to assess the safety and feasibility of DHOPE in DCD liver transplantation. Methods: In consecutive DCD liver transplantations, liver grafts were treated with end-ischaemic DHOPE. Outcome was compared with that in a control group of DCD liver transplantations without DHOPE, matched for donor age, donor warm ischaemia time, and recipient Model for End-stage Liver Disease (MELD) score. All patients were followed for 1 year. Results: Ten transplantations involving liver grafts treated with DHOPE were compared with 20 control procedures. There were no technical problems. All 6-month and 1-year graft and patient survival rates were 100 per cent in the DHOPE group. Six-month graft survival and 1-year graft and patient survival rates in the control group were 80, 67 and 85 per cent respectively. During DHOPE, median (i.q.r.) hepatic adenosine 5'-triphosphate (ATP) content increased 11-fold, from 6 (3-10) to 66 (42-87) mu mol per g protein (P = 0.005). All DHOPE-preserved livers showed excellent early function. At 1 week after transplantation peak serum alanine aminotransferase (ALT) and bilirubin levels were twofold lower in the DHOPE group than in the control group (ALT: median 966 versus 1858 units/l respectively, P = 0.006; bilirubin: median 1.0 (i.q.r. 0.7-1.4) versus 2.6 (0.9-5.1) mg/dl, P=0.044). None of the ten DHOPE-preserved livers required retransplantation for non-anastomotic biliary stricture, compared with five of 20 in the control group (P = 0.140). Conclusion: This clinical study of end-ischaemic DHOPE in DCD liver transplantation suggests that the technique restores hepatic ATP, reduces reperfusion injury, and is safe and feasible. RCTs with larger numbers of patients are warranted to assess the efficacy in reducing post-transplant biliary complications
