    International audienceWe have fabricated rhenium microwave resonators from epitaxial films. We have used thin films of different structural quality depending on their growth conditions. The resonators were coupled to a microwave transmission line which allows the measurement of their resonance frequencies and internal quality factors. From the resonance frequency at low temperature , the effective penetration depth and the London penetration depth of the rhenium film are extracted

    Variability and connectivity of plaice populations from the Eastern North Sea to the Western Baltic Sea, and implications for assessment and management

    An essential prerequisite of sustainable fisheries is the match between biologically relevant processes and management action. Various populations may however co-occur on fishing grounds, although they might not belong to the same stock, leading to poor performance of stock assessment and management. Plaice in Kattegat and Skagerrak have traditionally been considered as one stock unit. Current understanding indicates that several plaice components may exist in the transition area between the North Sea and the Baltic Sea. A comprehensive review of all available biological knowledge on plaice in this area is performed, including published and unpublished literature together with the analyses of commercial and survey data and historical tagging data. The results suggest that plaice in Skagerrak is closely associated with plaice in the North Sea, although local populations are present in the area. Plaice in Kattegat, the Belts Sea and the Sound can be considered a stock unit, as is plaice in the Baltic Sea. The analyses revealed great heterogeneity in the dynamics and productivity of the various local components, and suggested for specific action to maintain biodiversity

    Supernatant from Bifidobacterium Differentially Modulates Transduction Signaling Pathways for Biological Functions of Human Dendritic Cells

    International audienceBACKGROUND:Probiotic bacteria have been shown to modulate immune responses and could have therapeutic effects in allergic and inflammatory disorders. However, the signaling pathways engaged by probiotics are poorly understood. We have previously reported that a fermentation product from Bifidobacterium breve C50 (BbC50sn) could induce maturation, high IL-10 production and prolonged survival of DCs via a TLR2 pathway. We therefore studied the roles of mitogen-activated protein kinases (MAPK), glycogen synthase kinase-3 (GSK3) and phosphatidylinositol 3-kinase (PI3K) pathways on biological functions of human monocyte-derived DCs treated with BbC50sn.METHODOLOGY/PRINCIPAL FINDINGS:DCs were differentiated from human monocytes with IL-4 and GM-CSF for 5 days and cultured with BbC50sn, lipopolysaccharide (LPS) or Zymosan, with or without specific inhibitors of p38MAPK (SB203580), ERK (PD98059), PI3K (LY294002) and GSK3 (SB216763). We found that 1) the PI3K pathway was positively involved in the prolonged DC survival induced by BbC50sn, LPS and Zymosan in contrast to p38MAPK and GSK3 which negatively regulated DC survival; 2) p38MAPK and PI3K were positively involved in DC maturation, in contrast to ERK and GSK3 which negatively regulated DC maturation; 3) ERK and PI3K were positively involved in DC-IL-10 production, in contrast to GSK3 that was positively involved in DC-IL-12 production whereas p38MAPK was positively involved in both; 4) BbC50sn induced a PI3K/Akt phosphorylation similar to Zymosan and a p38MAPK phosphorylation similar to LPS.CONCLUSION/SIGNIFICANCE:We report for the first time that a fermentation product of a bifidobacteria can differentially activate MAPK, GSK3 and PI3K in order to modulate DC biological functions. These results give new insights on the fine-tuned balance between the maintenance of normal mucosal homeostasis to commensal and probiotic bacteria and the specific inflammatory immune responses to pathogen bacteria

    Chronic joint disease caused by persistent Chikungunya virus infection is controlled by the adaptive immune response

    Chikungunya virus (CHIKV) is a reemerging mosquito-borne pathogen that causes incapacitating disease in humans characterized by intense joint pain that can persist for weeks, months, or even years. Although there is some evidence of persistent CHIKV infection in humans suffering from chronic rheumatologic disease symptoms, little is known about chronic disease pathogenesis, and no specific therapies exist for acute or chronic CHIKV disease. To investigate mechanisms of chronic CHIKV-induced disease, we utilized a mouse model and defined the duration of CHIKV infection in tissues and the associated histopathological changes. Although CHIKV RNA was readily detectable in a variety of tissues very early after infection, CHIKV RNA persisted specifically in joint-associated tissues for at least 16 weeks. Inoculation of Rag1(−/−) mice, which lack T and B cells, resulted in higher viral levels in a variety of tissues, suggesting that adaptive immunity controls the tissue specificity and persistence of CHIKV infection. The presence of CHIKV RNA in tissues of wild-type and Rag1(−/−) mice was associated with histopathological evidence of synovitis, arthritis, and tendonitis; thus, CHIKV-induced persistent arthritis is not mediated primarily by adaptive immune responses. Finally, we show that prophylactic administration of CHIKV-specific monoclonal antibodies prevented the establishment of CHIKV persistence, whereas therapeutic administration had tissue-specific efficacy. These findings suggest that chronic musculoskeletal tissue pathology is caused by persistent CHIKV infection and controlled by adaptive immune responses. Our results have significant implications for the development of strategies to mitigate the disease burden associated with CHIKV infection in humans

    Multi-wave coherent control of a solid-state single emitter

An impressive level of control is now achieved using nuclear, vacancies and charge spins. Manipulation of bright exciton transitions in semiconductor quantum dots (QDs) is less advanced, principally due to the sub-nanosecond dephasing. Conversely, owing to their robust coupling to light, one can apply tools of nonlinear spectroscopy to achieve all-optical command. Here, we report on the coherent manipulation of an exciton via multi-wave mixing. Specifically, we employ three resonant pulses driving a single InAs QD. The first two induce a four-wave mixing (FWM) transient, which is projected onto a six-wave mixing (SWM) depending on the delay and area of the third pulse, in agreement with analytical predictions. Such a switch enables to demonstrate the generation of SWM on a single emitter and to engineer the spectro-temporal shape of the coherent response originating from a TLS. These results pave the way toward multi-pulse manipulations of solid state qubits via implementing the NMR-like control schemes in the optical domain.PostprintPeer reviewe

    CONCERTO: Simulating the CO, [CII], and [CI] line emission of galaxies in a 117 deg2 field and the impact of field-to-field variance

    In the submillimeter regime, spectral line scans and line intensity mapping (LIM) are new promising probes for the cold gas content and star formation rate of galaxies across cosmic time. However, both of these two measurements suffer from field-to-field variance. We study the effect of field-to-field variance on the predicted CO and [CII] power spectra from future LIM experiments such as CONCERTO, as well as on the line luminosity functions (LFs) and the cosmic molecular gas mass density that are currently derived from spectral line scans. We combined a 117 deg2 dark matter lightcone from the Uchuu cosmological simulation with the simulated infrared dusty extragalactic sky (SIDES) approach. The clustering of the dusty galaxies in the SIDES-Uchuu product is validated by reproducing the cosmic infrared background anisotropies measured by Herschel and Planck. We find that in order to constrain the CO LF with an uncertainty below 20%, we need survey sizes of at least 0.1 deg2. Furthermore, accounting for the field-to-field variance using only the Poisson variance can underestimate the total variance by up to 80%. The lower the luminosity is and the larger the survey size is, the higher the level of underestimate. At z < 3, the impact of field-to-field variance on the cosmic molecular gas density can be as high as 40% for the 4.6 arcmin2 field, but drops below 10% for areas larger than 0.2 deg2. However, at z > 3 the variance decreases more slowly with survey size and for example drops below 10% for 1 deg2 fields. Finally, we find that the CO and [CII] LIM power spectra can vary by up to 50% in 1 deg2 fields. This limits the accuracy of the constraints provided by the first 1 deg2 surveys. In addition the level of the shot noise power is always dominated by the sources that are just below the detection thresholds, which limits its potential for deriving number densities of faint [CII] emitters. We provide an analytical formula to estimate the field-to-field variance of current or future LIM experiments given their observed frequency and survey size.     Chikungunya Virus Infection

    Chikungunya virus (CHIKV) is an alphavirus transmitted by mosquitoes, mostly Aedes aegypti and Aedes albopictus. After half a century of focal outbreaks of acute febrile polyarthralgia in Africa and Asia, the disease unexpectedly spread in the past decade with large outbreaks in Africa and around the Indian Ocean and rare autochthonous transmission in temperate areas. This emergence brought new insights on its pathogenesis, notably the role of the A226V mutation that improved CHIKV fitness in Ae. albopictus and the possible CHIKV persistence in deep tissue sanctuaries for months after infection. Massive outbreaks also revealed new aspects of the acute stage: the high number of symptomatic cases, unexpected complications, mother-to-child transmission, and low lethality in debilitated patients. The follow-up of patients in epidemic areas has identified frequent, long-lasting, rheumatic disorders, including rare inflammatory joint destruction, and common chronic mood changes associated with quality-of-life impairment. Thus, the globalization of CHIKV exposes countries with Aedes mosquitoes both to brutal outbreaks of acute incapacitating episodes and endemic long-lasting disorders
