    A global lake and reservoir volume analysis using a surface water dataset and satellite altimetry

    Lakes and reservoirs are crucial elements of the hydrological and biochemical cycle and are a valuable resource for hydropower, domestic and industrial water use, and irrigation. Although their monitoring is crucial in times of increased pressure on water resources by both climate change and human interventions, publically available datasets of lake and reservoir levels and volumes are scarce. Within this study, a time series of variation in lake and reservoir volume between 1984 and 2015 were analysed for 137 lakes over all continents by combining the JRC Global Surface Water (GSW) dataset and the satellite altimetry database DAHITI. The GSW dataset is a highly accurate surface water dataset at 30&thinsp;m resolution compromising the whole L1T Landsat 5, 7 and 8 archive, which allowed for detailed lake area calculations globally over a very long time period using Google Earth Engine. Therefore, the estimates in water volume fluctuations using the GSW dataset are expected to improve compared to current techniques as they are not constrained by complex and computationally intensive classification procedures. Lake areas and water levels were combined in a regression to derive the hypsometry relationship (dh&thinsp;∕&thinsp;dA) for all lakes. Nearly all lakes showed a linear regression, and 42&thinsp;% of the lakes showed a strong linear relationship with a R2&thinsp;&gt;&thinsp;0.8, an average R2 of 0.91 and a standard deviation of 0.05. For these lakes and for lakes with a nearly constant lake area (coefficient of variation &lt;&thinsp;0.008), volume variations were calculated. Lakes with a poor linear relationship were not considered. Reasons for low R2 values were found to be (1) a nearly constant lake area, (2) winter ice coverage and (3) a predominant lack of data within the GSW dataset for those lakes. Lake volume estimates were validated for 18 lakes in the US, Spain, Australia and Africa using in situ volume time series, and gave an excellent Pearson correlation coefficient of on average 0.97 with a standard deviation of 0.041, and a normalized RMSE of 7.42&thinsp;%. These results show a high potential for measuring lake volume dynamics using a pre-classified GSW dataset, which easily allows the method to be scaled up to an extensive global volumetric dataset. This dataset will not only provide a historical lake and reservoir volume variation record, but will also help to improve our understanding of the behaviour of lakes and reservoirs and their representation in (large-scale) hydrological models.</p

    STAT3 differential scanning fluorimetry and differential scanning light scattering assays: Addressing a missing link in the characterization of STAT3 inhibitor interactions

    STAT3 protein is an established target for the development of new cancer therapeutic agents. Despite lacking a traditional binding site for small molecule inhibitors, many STAT3 inhibitors have been identified and explored for their anti-cancer activity. Because STAT3 signaling is mediated by protein-protein interactions, indirect methods are often employed to determine if proposed STAT3 inhibitors bind to STAT3 protein. While established STAT3 inhibition assays (such as the fluorescence polarization assay, electrophoretic mobility shift assay and ELISAs) have been used to identify novel inhibitors of STAT3 signaling, methods that directly assess STAT3 protein-inhibitor interactions could facilitate the development of novel inhibitors. In this context, we herein report new STAT3 binding assays based on differential scanning fluorimetry (DSF) and differential scanning light scattering (DSLS) to characterize interactions between STAT3 protein and inhibitors. Several peptide and small molecule STAT3 inhibitors have been evaluated, and new insight into how these compounds may interact with STAT3 is provided

    Heme Oxygenase Isoforms Differ in Their Subcellular Trafficking during Hypoxia and Are Differentially Modulated by Cytochrome P450 Reductase

    Heme oxygenase (HO) degrades heme in concert with NADPH cytochrome P450 reductase (CPR) which donates electrons to the reaction. Earlier studies reveal the importance of the hydrophobic carboxy-terminus of HO-1 for anchorage to the endoplasmic reticulum (ER) which facilitates the interaction with CPR. In addition, HO-1 has been shown to undergo regulated intramembrane proteolysis of the carboxy-terminus during hypoxia and subsequent translocation to the nucleus. Translocated nuclear HO-1 was demonstrated to alter binding of transcription factors and to alter gene expression. Little is known about the homologous membrane anchor of the HO-2 isoform. The current work is the first systematic analysis in a eukaryotic system that demonstrates the crucial role of the membrane anchor of HO-2 for localization at the endoplasmic reticulum, oligomerization and interaction with CPR. We show that although the carboxy-terminal deletion mutant of HO-2 is found in the nucleus, translocation of HO-2 to the nucleus does not occur under conditions of hypoxia. Thus, we demonstrate that proteolytic regulation and nuclear translocation under hypoxic conditions is specific for HO-1. In addition we show for the first time that CPR prevents this translocation and promotes oligomerization of HO-1. Based on these findings, CPR may modulate gene expression via the amount of nuclear HO-1. This is of particular relevance as CPR is a highly polymorphic gene and deficiency syndromes of CPR have been described in humans

    The Amino-Terminus of Nitric Oxide Sensitive Guanylyl Cyclase α1 Does Not Affect Dimerization but Influences Subcellular Localization

    BACKGROUND: Nitric oxide sensitive guanylyl cyclase (NOsGC) is a heterodimeric enzyme formed by an α- and a β₁-subunit. A splice variant (C-α₁) of the α₁-subunit, lacking at least the first 236 amino acids has been described by Sharina et al. 2008 and has been shown to be expressed in differentiating human embryonic cells. Wagner et al. 2005 have shown that the amino acids 61-128 of the α₁-subunit are mandatory for quantitative heterodimerization implying that the C-α₁-splice variant should lose its capacity to dimerize quantitatively. METHODOLOGY/PRINCIPAL FINDINGS: In the current study we demonstrate preserved quantitative dimerization of the C-α₁-splice by co-purification with the β₁-subunit. In addition we used fluorescence resonance energy transfer (FRET) based on fluorescence lifetime imaging (FLIM) using fusion proteins of the β₁-subunit and the α₁-subunit or the C-α₁ variant with ECFP or EYFP. Analysis of the respective combinations in HEK-293 cells showed that the fluorescence lifetime was significantly shorter (≈0.3 ns) for α₁/β₁ and C-α₁/β₁ than the negative control. In addition we show that lack of the amino-terminus in the α₁ splice variant directs it to a more oxidized subcellular compartment. CONCLUSIONS/SIGNIFICANCE: We conclude that the amino-terminus of the α₁-subunit is dispensable for dimerization in-vivo and ex-vivo, but influences the subcellular trafficking

    Un nouveau chinchilloïde (Rodentia, Hystricognathi) du Miocène inférieur des localités de Bryn Gwyn et Gran Barranca (Patagonie, Argentine)

    A new genus and species of chinchilloid rodent is described here. It was found in Colhuehuapian levels (early Miocene) of the localities of Bryn Gwyn and Gran Barranca, Chubut Province, Argentina. The new taxon shows a unique combination of characters (e.g., protohypsodont teeth, upper molars trilophodont, m1 and m2 tri- or tetralophodont with the second crest in position of variable development, and m3 trilophodont, cement absent) that make it different from any other known chinchilloids. We performed a phylogenetic analysis to corroborate the relationships of the new taxon within the Chinchilloidea. Our results indicate that the new taxon is best classified as Chinchilloidea incertae sedis, together with Incamys, Garridomys, and Scotamys. These species form the basal stock that leads to the modern lineage Chinchillidae (chinchillas and viscachas). The presence of the new taxon in these localities increases the diversity of chinchilloids during the early Miocene and reduces the dissimilarity between the faunas found in Bryn Gwyn and Gran Barranca.Un nouveau genre et une nouvelle espèce de rongeur chinchilloïde sont ici décrits. Celui-ci a été trouvé dans les niveaux colhuéhuapiens (Miocène inférieur des localités de Bryn Gwyn et de Gran Baranca, province de Chubut, Argentine). Le nouveau taxon montre une combinaison unique de caractères (à savoir, dents protohypsodontes, molaires supérieures trilophodontes, m1 et m2 tri- ou tétralophodontes avec la seconde crête en position de développement variable et m3 trilophodonte, ciment absent) qui le rend différent de tous les autres chinchilloïdes connus. Une analyse phylogénétique a été réalisée pour confirmer les relations du nouveau taxon au sein des Chinchilloidea. Nos résultats montrent que le nouveau taxon se classe au mieux en tant que Chinchilloida incertae sedis, avec Incamys, Garridomys et Scotamys. Ces espèces forment le stock basal qui conduit à la lignée moderne des Chinchillidae (chinchillas et viscachas).     Assessing sponge cities performance at city scale using remotely sensed LULC changes: Case study Nanjing

    As a result of high-density urbanization and climate change, both the frequency and intensity of extreme urban rainfall are increasing. Drainage systems are not designed to cope with this increase, and as a result, floods are becoming more common in cities, particularly in the rapidly growing cities of China. To better cope with more frequent and severe urban flooding and to improve the water quality of stormwater runoff, the Chinese government launched the national Sponge City Construction (SCC) program in 2014. The current SCC design standards and guidelines are based on static values (e.g., return periods, rainfall intensities, and volume capture ratio (VCR)). They do not fully acknowledge the large differences in climate conditions across the country and assume that the hydraulic conditions will not change over time. This stationary approach stems from the traditional engineering approach designed for grey infrastructure (following a “one size fits all” approach). The purpose of this study was to develop a methodology to assess the VCR baseline (before construction in the pre-development stage) and changes in VCR (difference between the VCR of the pre- and post-development stage). The VCR of the post-development stage is one of the required indicators of the Assessment Standard for Sponge Cities Effects to evaluate SCC projects. In this study, the VCR was derived from remote-sensing-based land use land cover (LULC) change analysis, applying an unsupervised classification algorithm on different Landsat images from 1985 to 2015. A visualization method (based upon Sankey chart, which depicts the flows and their proportions of components) and a novel and practical partitioning method for built-up regions were developed to visualize and quantify the states and change flows of LULC. On the basis of these findings, we proposed a new indicator, referred to as VCRa−L, in order to assess the changes in urban hydrology after SCC construction. This study employed the city of Nanjing as a case study and analyzed detailed information on how LULC changes over time of built-up areas. The surface area of the urban and built-up areas of Nanjing quadrupled from 11% in 1985 to 44% in 2015. In the same period, neither the entire city nor its subregions reached the VCR target of 80%. The proposed new methodology aims to support national, regional, and city governments to identify and prioritize where to invest and implement SCC measures more effectively in cities across China

    Hepatic denervation and dyslipidemia in obese Zucker (fa/fa) rats

    Human and animal studies increasingly point toward a neural pathogenesis of the metabolic syndrome, involving hypothalamic and autonomic nervous system dysfunction. We hypothesized that increased very-low-density lipoprotein-triglyceride (VLDL-TG) secretion by the liver in a rat model for dyslipidemia, that is, the obese Zucker (fa/fa) rat, is due to relative hyperactivity of sympathetic, and/or hypoactivity of parasympathetic hepatic innervation. To test the involvement of the autonomic nervous system, we surgically denervated the sympathetic or parasympathetic hepatic nerve in obese Zucker rats. Our results show that cutting the sympathetic hepatic nerve lowers VLDL-TG secretion in obese rats, finally resulting in lower plasma TG concentrations after 6 weeks. In contrast, a parasympathetic denervation results in increased plasma total cholesterol concentrations. The effect of a sympathetic or parasympathetic denervation of the liver was independent of changes in humoral factors or changes in body weight or food intake. In conclusion, a sympathetic denervation improves the lipid profile in obese Zucker rats, whereas a parasympathetic denervation increases total cholesterol levels. We believe this is a novel treatment target, which should be further investigated
