In dit advies onderzoekt het INBO de voor Vlaanderen relevante introductieroutes. We gaan ook na welke daarvan de belangrijkste zijn en prioritair maatregelen vereisen. Tot slot bekijken we ook welke informatie nodig is om een meer diepgaande analyse te kunnen uitvoeren

    Bilberries potentially alleviate stress-related retinal gene expression induced by a high-fat diet in mice

    PURPOSE: Obesity- and diabetes-associated visual impairment and vascular dysfunctions are increasing as causes of vision loss. The detailed mechanisms of how obesity and diabetes affect eye health are still largely unknown, but animal models have been useful in exploring the effects of potential protective compounds, i.e., compounds characterized by antioxidant and anti-inflammatory properties. These properties occur in anthocyanins, and bilberries (European wild blueberries, Vaccinium myrtillus) are a major source of dietary anthocyanins in Nordic diets. The main aim of the present work was to study the protective effects of dietary bilberries (BB) on the level of gene expression in retinas in mice that develop obesity when fed a high-fat diet (HFD). METHODS: Mice (n=6 per group, four groups) were fed ad libitum a normal control diet (NCD), a HFD, or a diet with 5% bilberries (NCD+BB, HFD+BB) for 12 weeks. Food consumption, weight gain, and blood pressure were measured during the feeding period and whole blood serum markers of obesity at sacrifice. Retinas were collected, and RNA extracted from all 24 mice and pooled samples from four mice per group were hybridized to Mouse-Ref8 V2 Expression BeadChips (Illumina platform) with 25,697 probes for genes and transcript variants. The expression profiles in the retinas were analyzed using R, PathVisio, and DAVID to screen for high fat–induced changes as well as for bilberry-induced changes in the HFD up- or downregulated transcripts. RESULTS: The HFD and HFD+BB groups gained weight from week 5 and final weight, blood glucose, serum free fatty acids, and systolic blood pressure as compared to mice fed the control diets (Mann–Whitney’s U-test, p<0.05). Bilberries had no significant effect on these parameters other than a trend to reduce systolic blood pressure in the HFD-fed mice (101±4 versus 113±9 mmHg, p=0.10). Gene ontology enrichment analysis of 810 differentially expressed genes (F-test, p<0.05) in the retina displayed differential regulation of genes in ontology groups, mainly pathways for apoptosis, inflammation, and oxidative stress, especially systemic lupus erythematosus, mitogen-activated protein kinase, and glutathione metabolism. Mice fed a HFD had increased retinal gene expression of several crystallins, while the HFD+BB mice showed potential downregulation of these crystallins when compared to the HFD mice. Bilberries also reduced the expression of genes in the mitogen-activated protein kinase (MAPK) pathway and increased those in the glutathione metabolism pathway. CONCLUSIONS: HFD feeding induces differential expression of several stress-related genes in the mouse retina. Despite minor effects in the phenotype, a diet rich in bilberries mitigates the upregulation of crystallins otherwise induced by HFD. Thus, the early stages of obesity-associated and stress-related gene expression changes in the retina may be prevented with bilberries in the diet

    Visualizing mineralization in deformed opercular bones of larval gilthead sea bream (Sparus aurata)

    During the rearing process of gilthead sea bream (Sparus aurata), abnormal development of the opercular bone is particularly common (Aquaculture 156, 1997, 165). In order to alleviate its occurrence in rearing facilities, it's crucial to identify the very first physical signs of deviation in normal skeletal development. Nano-CT-scanning was tested for its applicability to quantify deviations in bone mineralization levels. Seven opercles were dissected from larvi of 65 days post hatching, randomly sampled at the commercial sea bream hatchery Maricoltura di Rosignano Solvay (Livorno, Italy). The samples were nano-CT-scanned and computationally reconstructed. Mineralization intensity was colorcoded using Amira software, resulting in a detailed visualization of opercular morphology and mineralization patterns. In conclusion, nano-CT-scanning promises to be a good tool to both describe morphology and detect mineralization levels in the early onset of deformities

    Genome-Wide Identification of Expression Quantitative Trait Loci (eQTLs) in Human Heart.

    In recent years genome-wide association studies (GWAS) have uncovered numerous chromosomal loci associated with various electrocardiographic traits and cardiac arrhythmia predisposition. A considerable fraction of these loci lie within inter-genic regions. The underlying trait-associated variants likely reside in regulatory regions and exert their effect by modulating gene expression. Hence, the key to unraveling the molecular mechanisms underlying these cardiac traits is to interrogate variants for association with differential transcript abundance by expression quantitative trait locus (eQTL) analysis. In this study we conducted an eQTL analysis of human heart. For a total of 129 left ventricular samples that were collected from non-diseased human donor hearts, genome-wide transcript abundance and genotyping was determined using microarrays. Each of the 18,402 transcripts and 897,683 SNP genotypes that remained after pre-processing and stringent quality control were tested for eQTL effects. We identified 771 eQTLs, regulating 429 unique transcripts. Overlaying these eQTLs with cardiac GWAS loci identified novel candidates for studies aimed at elucidating the functional and transcriptional impact of these loci. Thus, this work provides for the first time a comprehensive eQTL map of human heart: a powerful and unique resource that enables systems genetics approaches for the study of cardiac traits

    A science-based approach to tackle invasive alien species in Belgium – the role of the ISEIA protocol and the Harmonia information system as decision support tools

    A coherent response to biological invasions involves science-based, up-to-date prioritization tools alongside information transfer to relevant authorities and stakeholders. Here, we describe how the collaboration between scientists and policy makers in Belgium has allowed the development of decision support tools regarding invasive alien species. We present the environmental impact assessment protocol ISEIA and comment on its applications. Furthermore, we describe and provide metadata for the information system Harmonia which was developed to disseminate this information to a diverse audience. Using several examples of initiatives addressing the threat of invasive alien species in Belgium, we show how these tools have been instrumental in strengthening capacity of the scientific community, authorities and stakeholders in Belgium on addressing the invasive alien species issue

    A 3D quantitative method for analyzing bone mineral densities: a case study on skeletal deformities in the gilthead sea bream, <i>Sparus aurata</i> (Linnaeus, 1758)

    Skeletal deformities, one of the major threats for aquaculture, have been studied extensively. These include opercular malformations in gilthead sea bream (Sparus aurata), a key fish species for Mediterranean aquaculture. What is causing it and at what morphogenetic level it arises, however, is still unclear. Here we focus on bone formation, at the level of bone mineralization. Several methods have been used to study bone mineralization density (BMD), however, these are frequently limited when targeting a high-resolution, three-dimensional mapping of BMD. We used micro-computed tomography (micro-CT) data to perform such a 3D quantification of BMD levels in gilthead sea bream that showed different levels of opercular bone deformations. This approach has the advantage of not having to rely on calibration phantoms, as long as relative BMD values are needed. The results show an increased BMD in deformed opercles compared to normal ones, especially in a bilaterally-deformed specimen. Furthermore, we show that opercular deformations are not necessarily associated with similar mineralization patterns in other mineralized cranial elements, except for the otoliths. Also, mineralization seems to occur left-right independently, matching earlier observations of such an independency of the opercular phenotype as a whole. This study confirms that a quantitative characterization of BMD patterns in 3D is feasible, even in smaller specimens, and that it has several advantages over other commonly used approaches
