    Functional Eubacteria Species Along with Trans-domain Gut Inhabitants Favour Dysgenic Diversity in Oxalate Stone Disease

    Analyses across all three domains of life are necessary to advance our understanding of taxonomic dysbiosis in human diseases. In the present study, we assessed gut microbiota (eubacteria, archaea, and eukaryotes) of recurrent oxalate kidney stone suffers to explore the extent of trans-domain and functional species dysbiosis inside the gut. Trans-domain taxonomic composition, active oxalate metabolizer and butyrate-producing diversity were explored by utilizing frc-, but-, and buk- functional gene amplicon analysis. Operational taxonomic units (OTUs) level analyses confound with the observation that dysbiosis in gut microbiota is not just limited to eubacteria species, but also to other domains like archaea and eukaryotes. We found that some of healthy eubacterial population retained together with Oxalobacter formigenes and Lactobacillus plantarum colonization in disease condition (p \u3c 0.001 & FDR = 0.05). Interestingly, trans-domain species diversity has been less shared and dysgenic taxa augmentation was found to be higher. Oxalate metabolizing bacterial species (OMBS) and butyrate-producing eubacteria species were found to be decreased in Oxalobacter non-colonizers; and Prevotella and Ruminococcus species which may contribute to oxalate metabolism and butyrate synthesis as well. Our study underscores fact that microbial dysbiosis is not limited to eubacteria only hence suggest the necessity of the trans-domain surveillance in metabolic diseases for intervention studies

Panchakarma (5 purification therapies) practices are carried out abundantly by physicians considering its effective results in shorter time duration. Many people also practice these at home without proper consultation from expert physician just by reliying on the information from social media. Administration of purification therapy at inappropriate stage or with faulty manners leads to vitiation of Dosha, which ultimately leads to occurrence of disease. Bruhatrayee (3 main classical texts) of Ayurveda has mentioned Shodhana Mithyayoga (improper administration of purification therapy) as causative factor for various diseases quoted in Nidanasthana and Chikitsasthana.     HPLC ANALYSIS OF HUMAN URINE FOR OXALATE CONTENT

    Objective: In the present communication, development and validation of reverse phase-high performance liquid chromatography method have been carried out for estimation of oxalate content in the urine of human volunteers with recurrent kidney stone disease and healthy status.Methods: The analysis of oxalic acid has been carried out on KYA TECH HiQ Sil C18HS column using a mobile phase of methanol: 0.001 N acetic acid in water (50:50, v/v) with a flow rate of 1 ml/min and detection wavelength, 237 nm.Results: Analysis of oxalate content was carried out using single point calibration method with retention at 2.705 min with good resolution parameters. Urine sample collected from kidney stone patients and healthy volunteers over the period of 24 h were analyzed and it has been found that concentration of oxalate in healthy volunteers is less than 12 µg/ml whereas that in case of kidney stone patients is in the range of 39-151 µg/ml and this data can be utilized for further interpretations about oxalate content in healthy and kidney stone diseased volunteers. This method was validated as per united states food and drug administration (USFDA) guidelines by the study of accuracy, precision, linearity, range, selectivity, the lower limit of quantitation, extraction recovery studies and stability studies for determining oxalate content in the urine of human volunteers. As relative standard deviations of oxalate content estimated are less than 5 percent, the method can be claimed accurate, precise, sensitive and selective for determining oxalate content in the urine of human volunteers.Conclusion: The results are satisfactory, proving the effectiveness of the method for analysis of oxalate content from other biological fluids with few optimizations

    Evaluation of harvesting time and standardization of distillation duration for higher essential oil content and quality in German chamomile (Chamomilla recutita L.)

    Essential oil yield and composition in aromatic crops might be affected by genetic, agronomical and environmental factors but till date there is no clear information about the harvesting time and distillation for higher essential oil content without affecting quality. The current study was carried out to evaluate harvesting of chamomile flowers without herb and with herb part at three different times (6 A.M., 12 P.M. and 6 P.M.) and four distillation treatments (3 h, 4 h, 5 h & 6 h) for dried chamomile flowers. Results indicated that essential oil content was more in chamomile flowers without herb (0.15-0.18%) as compared to flowers with herb (0.06-0.09%). Essential oil content in chamomile flowers without herb was found statistically at par at harvest time of 12 P.M. (0.18%) and 6 P.M. (0.18%) and significantly higher than harvesting time of 6 A.M. (0.15%). Essential oil of chamomile flowers without herb contained maximum α-bisabolol oxide-B, (Z)-spiroether, and chamazulene at 12 P.M. and 6 P.M. while, α-bisabolone oxide-A and α-bisabolol oxide-A were maximum at 6 A.M. and (E)-β-farnesene was more at 12 P.M. Similarly, in distillation experiment, higher oil content was observed in chamomile dried flowers which were hydro-distilled for 6 h (1.20%) compared to other hydro-distillation durations. Marker compounds i.e. α-bisabolol oxide-A, α-bisabolone oxide-A, α-bisabolol oxide-B, (E)-β-farnesene and chamazulene were more at 5 h and 6 h distillation duration while (Z)-spiroether was more at 3 h distillation duration. The present study showed that in order to obtain higher essential oil, flowers without herb harvested at 12 P.M or 6 P.M. should be subjected to 5-6 h hydro-distillation

In analytical solution Simulation and analysis is carried out with the help of mold flow software. In this work we use Autodesk mold flow software. From the simulation and analysis, the software for flow simulation provides sufficient information regarding filling time, injection pressure, defects like air traps, weld lines, sink mark, warpage etc. With these results, users can avoid the defect of the plastic in actual injection. The analysis will help the to design a mold with minimum modifications and which will also reduce the time and cost. The design will be validated by producing the component with the help of the designed mold

    Background: Osteoarthritis (OA) of knee is a common progressive multifactorial joint disorder affecting the quality of life, and surgical repair is the final option which has substantial impact on healthcare costs. This real-world study evaluates the efficacy and safety of an oral formulation containing UC-II and aflapin (Boswellia serrata extract enriched in 3-O-acetyl-11-keto-beta-boswellic acid) for treatment of OA of knee.Methods: Data of 505 ambulatory adult patients (study duration-Jul-21 to Jul-22) of either gender (227 M, 278 F) having OA of knee, and who received study treatment (capsule HAPID®, Wockhardt, India) once daily for a period of up to 90 days were included for the study after obtaining informed written consent. Primary outcomes were mean change in Western Ontario and McMaster universities OA index (WOMAC) scores from baseline through day 90 (total and sub-scales for joint pain, joint stiffness, and physical function), and change in 0-10 visual analogue scale (VAS) score for pain.Results: About 285 (56.4%) patients were newly diagnosed, majority (63.4%) were having grade 2 severity of OA (Kellgren and Lawrence grade). The mean (SD) baseline total WOMAC scores improved from 60.94 (23.60) at baseline to 26.42 (22.19) on day 90. Significant improvements were seen starting from day 5 (p=0.023) and progressively up to day 90 (p<0.0001).Conclusions: The excellent safety and efficacy profile of combination therapy with aflapin and UC-II makes it a desirable pharmacological treatment modality for management of patients of knee OA

    Influence of mesoscale eddies on a commercial fishery in the coastal waters of the Andaman and Nicobar Islands, India

    Mesoscale eddies enhance the productivity in a stratified coastal environment by upwelling. The seas around the Andaman and Nicobar Islands have been found to have frequent mesoscale eddy activity. Commercial fishing grounds coincide with upwelling areas associated with cyclonic and anticyclonic eddies and also with areas between two adjacent eddies. There are different eddy zones supporting different types of fishing gears and fish. The current study aims at identifying the different zones of mesoscale eddies in the Andaman Sea and compares the productivity and fishing activity in each of them. Data collected from 454 commercial fishing trips in the Andaman Sea along with maps of sea level anomaly and Moderate Resolution Imaging Spectroradiometer (MODIS) global level 3 mapped thermal infrared (IR) daytime sea surface temperature (SST) from the Aqua and Terra satellites were used for the study. Known upwelling areas such as the periphery of anticyclonic and the core of cyclonic eddies showed higher catches in longlines, ring seines among the fishing gears, and among all the fish species groups. Downwelling areas such as the periphery of cyclonic and the core of anticyclonic eddies showed lower catches with ring seines and the fish species groups. Areas in between adjacent eddies were explored in this study and the fish captures in such areas were found to be different with types of fishing and the target fish group. The study shows results that link eddy activity with the performance of a fishery

    Genome wide analysis of gene expression changes in skin from patients with type 2 diabetes

    Non-healing chronic ulcers are a serious complication of diabetes and are a major healthcare problem. While a host of treatments have been explored to heal or prevent these ulcers from forming, these treatments have not been found to be consistently effective in clinical trials. An understanding of the changes in gene expression in the skin of diabetic patients may provide insight into the processes and mechanisms that precede the formation of non-healing ulcers. In this study, we investigated genome wide changes in gene expression in skin between patients with type 2 diabetes and non-diabetic patients using next generation sequencing. We compared the gene expression in skin samples taken from 27 patients (13 with type 2 diabetes and 14 non-diabetic). This information may be useful in identifying the causal factors and potential therapeutic targets for the prevention and treatment of diabetic related diseases
