    Bacteriophage Mu integration in yeast and mammalian genomes

    Genomic parasites have evolved distinctive lifestyles to optimize replication in the context of the genomes they inhabit. Here, we introduced new DNA into eukaryotic cells using bacteriophage Mu DNA transposition complexes, termed ‘transpososomes’. Following electroporation of transpososomes and selection for marker gene expression, efficient integration was verified in yeast, mouse and human genomes. Although Mu has evolved in prokaryotes, strong biases were seen in the target site distributions in eukaryotic genomes, and these biases differed between yeast and mammals. In Saccharomyces cerevisiae transposons accumulated outside of genes, consistent with selection against gene disruption. In mouse and human cells, transposons accumulated within genes, which previous work suggests is a favorable location for efficient expression of selectable markers. Naturally occurring transposons and viruses in yeast and mammals show related, but more extreme, targeting biases, suggesting that they are responding to the same pressures. These data help clarify the constraints exerted by genome structure on genomic parasites, and illustrate the wide utility of the Mu transpososome technology for gene transfer in eukaryotic cells

    A Test Of A Crime And Justice Infotainment Measure

    In recent years, a new term which combined aspects of both news entertainment into a single format had been introduced into the crime and justice media vocabulary-infotainment. Media history reflected information and entertainment as usually presented separate and distinct. Early television programming, for example, could easily be recognized and classified as news or entertainment, but recently, the line between news and entertainment had become blurred. Beginning in the late 1980s, contemporary infotainment programs like Geraldo or Unsolved Mysteries began to appear on television and the genre had increased steadily in popularity. Since the 1980s, it had become more difficult for television viewers and media consumers in general to identify what was news and what was entertainment. This article presents a brief history of crime and justice infotainment and develops and tests a measurement refinement of crime and justice infotainment programming. Additionally, it offers research directions for exploring the impact of such programming. © 2002 Elsevier Science Ltd. All rights reserved

    SIMULATION AND VISUALIZATION OF GENETIC REGULATORY NETWORKS

    Gene regulation networks are a significant biological research area. Simulations and visualizations of genetic processes are being created as biologists grapple with the vast amounts of new genetic information. We present a genetic network simulation environment that visualizes protein-gene interactions and concentrations as they occur during the simulation. In addition, the layout of this genetic simulation can be changed into a visualization of a conceptual model of the simulated genetic network. The visual simulation and network visualization are integrated by animating the change between the simulation view and the visualization view. The flagella system of Escherichia coli has been used to verify the results of this tool and to provide a working model. This particle-based real-time visual simulations of genetic networks allows for virtual experimentation using similar methodologies of live experiments.We are currently acquiring citations for the work deposited into this collection.     Social Worker Perceptions Of The Portrayal Of The Profession In The News And Entertainment Media: An Exploratory Study

    This exploratory study describes social workers’ perceptions of the depiction ofthe social work profession found in the news and entertainment media. A randomsample of 665 MSW social workers who were members of the FloridaChapter of the National Association of Social Workers were surveyed regardinghow they felt the profession was depicted in the news and entertainment mediaon a set of paired adjectives comprising 9 semantic differential scales. Findingsshowed that these social workers perceived the profession to be depicted negativelyin both news and entertainment media.     Roles and Responsibilities: Analyzing Local Leaders\u27 Views on Jail Crowding From a Systems Perspective

    Jail crowding is a substantial concern for many local jurisdictions. Although several authors have suggested a system-wide approach to reduce crowding, relatively little is known about how top local criminal justice officials viewthis issue. Using interviews and surveys of criminal justice leaders in a large southern metropolitan county, this study examined perspectives on jail crowding and perceived interagency relationships. Consistent with prior studies, the respondents agreed that jail crowding is a problem and that the consequences of crowding extend beyond the correctional facility. Greater discord was observed on issues of agency responsibility for changing policies to effect reductions in crowding. Findings are interpreted within the context of a loose coupling framework of criminal justice organizations
