A study on the differentiation of filamentous cyanobacterial isolates using DNA fingerprinting approach
337-345The present study investigates the implication of DNA fingerprinting in discrimination of class isolates collected from a spread of habitats in south west Khasi Hills district of Meghalaya, India. Five totally different primers i.e., M13, enterbacterial repetitive intergenic consensus (ERIC), short tandem repetitive repeats (STRRs) 1A & B and long tandem repetitive repeats (LTRRs), derived from repetitive sequences gift within the ordering of true bacteria, were utilized.
The generated knowledge was then evaluated for fingerprinting exploitation procedure and unweighted pairgroup method of arithmetic average (UPGMA) based dendrograms. Clear and variable stripe patterns were obtained for every of the targeted genomic regions used, LTRR being the sole exception. The stripe patterns delineate the existence of 2 basic distinct populations of true bacteria among the 19 isolates. The UPGMA cluster analyses in addition, disclosed the presence of numerous lineages among the isolates. On examination the resolution capability of the four genomic regions studied, it had been seen that, with the exception of LTRR, all the opposite regions hold smart resolution and therefore the ability to differentiate between numerous class populations. The results of the current study indicate the incidence of genetic variants among the 19 morphologically identical isolates of filiform cyanobacteria. Additionally, the study additionally explicates the quality of DNA fingerprinting supplemented with UPGMA cluster analysis as a speedy, economical and value effective tool for differentiating among morphologically identical populations
Spin-phonon coupling suppressing the structural transition in perovskite-like oxide
Multifunctional properties in quantum systems require the interaction between
different degrees of freedom. As such, spin-phonon coupling emerges as an ideal
mechanism to tune multiferroicity, magnetism, and magnetoelectric response. In
this letter, we demonstrate and explain, based on theoretical and experimental
analyses, an unusual manifestation of spin-phonon coupling, i.e., prevention of
a ferroelastic structural transition, and locking of high-temperature R-3m
phase in a magnetically frustrated perovskite-like oxide Ba2NiTeO6. We present
Ba2NiTeO6 as a prototype example among its family where long-range
antiferromagnetic structure couples with a low-frequency Eg mode (at 55 cm-1)
that exhibits a large anharmonicity. Our findings establish that spin-phonon
coupling clearly suppresses the phonon anharmonicity preventing the structural
phase transition from the R-3m to the C2/m phase in Ba2NiTeO6

301-305The present work aimed to increase the thermostability
and pH stability of lipase produced from Penicillium chrysogenum SNP5 using different anionic
carriers. The immobilized lipase from the fungus on 1.25% sodium alginate beads
and 12% polyacrylamide beads retained 37.28 and 59.75% immobilization
efficiency, respectively, on 3 mm bead size. Immobilized lipase on calcium
alginate and polyacrylamide beads retained 56.16 and 91.11% of its initial activity after 3rd use, whereas significant loss in lipase activity was observed
after 5th and 7th reuse, respectively. Properties
like optimum pH, Km, Vmax and thermostability of
immobilized lipase were changed with respect to free lipase. Optimum pH shifted
from 7 to 7.5 in calcium alginate and from 7 to 9 in polyacrylamide beads,
whereas t1/2 increased from 71 min to 205 min in calcium alginate
and from 71 min to 380 min in polyacrylamide beads at 45°C. Km and Vmax
values at free state lipase were observed as 0.30 mM and 31.948 μM min-1
mL-1, whereas after immobilization on calcium alginate and
polyacrylamide beads values changed to 0.188 mM and 27.24 μM min-1
mL-1, and 0.24 mM and 30.96 μmol min-1 mL-1,
respectively. Thus, on immobilization, lipase
has shown enhanced thermostability, specificity and working range of pH, which
would be more beneficial for its industrial application.

Modulatory effects by neodymium-doped yttrium aluminum garnet laser on fibroblast attachment to single rooted tooth surfaces following ultrasonic scaling and root planning: An in vitro study
Context: One of the most important goals of periodontal therapy is connective tissue reattachment to previously diseased root surfaces. In the recent years, laser therapy has been considered as an important tool in improving the treatment of periodontal disease. Aims: To evaluate the neodymium-doped yttrium aluminum garnet (Nd: YAG) lasers effects on root surfaces affected by periodontal disease and compare this treatment with scaling and root planning (SRP) in terms of fibroblast attachment. Materials and Methods: A sample of 30 single-rooted human teeth extracted because of advanced periodontal disease was used in this study. Sixty specimens obtained by longitudinal sectioning were randomly divided in three groups. Group A control (untreated); Group B SRP; Group C laser (Nd: YAG) and ultrasonic scaling. All specimens were incubated with fibroblast suspension and then fixed and observed under scanning electron microscope. Results: With a median of 8, the control group (Group A) exhibited the least number of total fibroblasts among all the three groups. The laser and scaling - treated group (Group C) showed the highest number of fibroblasts (median = 49, mean ± standard deviation [SD] = 48.28 ± 17.18), followed by SRP only (Group B, median = 22, mean ± SD = 22.24 ± 8.67). Conclusions: Nd: YAG laser irradiation at specific energy densities can be used as a useful tool to condition the root surfaces, enhancing fibroblast attachment. Hence aiding in re-establishment of the connective tissue attachment to the root surfaces of previously diseased teeth
