New Stability-Indicating RP-HPLC Method for Determination of Diclofenac Potassium and Metaxalone from their Combined Dosage Form
A simple, precise and accurate isocratic RP-HPLC stability-indicating assay method has been developed to determine diclofenac potassium and metaxalone in their combined dosage forms. Isocratic separation was achieved on a Hibar-C18, Lichrosphere-100® (250 mm × 4.6 mm i.d., particle size 5 μm) column at room temperature in isocratic mode, the mobile phase consists of methanol: water (80:20, v/v) at a flow rate of 1.0 ml/min, the injection volume was 20 μl and UV detection was carried out at 280nm. The drug was subjected to acid and alkali hydrolysis, oxidation, photolysis and heat as stress conditions. The method was validated for specificity, linearity, precision, accuracy, robustness and system suitability. The method was linear in the drug concentration range of 2.5–30 μg/ml and 20–240 μg/ml for diclofenac potassium and metaxalone, respectively. The precision (RSD) of six samples was 0.83 and 0.93% for repeatability, and the intermediate precision (RSD) among six-sample preparation was 1.63 and 0.49% for diclofenac potassium and metaxalone, respectively. The mean recoveries were between 100.99–102.58% and 99.97–100.01% for diclofenac potassium and metaxalone, respectively. The proposed method can be used successfully for routine analysis of the drug in bulk and combined pharmaceutical dosage forms
Stability-indicating RP-HPLC method for simultaneous estimation of levosalbutamol sulfate and theophylline in combined dosage form
A novel, simple, accurate and precise RP-HPLC method for simultaneous determination of levosalbutamol sulfate and theophylline has been developed and validated. Separation was achieved on a Phenomenex; C18 column (250 mm × 4.6 mm i.d., 5 µm) using methanol: 10 mM TBAHS(tetrabutyl ammonium hydrogen sulfate) (50:50, v/v) as mobile phase at flow rate of 1.0 mL.min-1. The UV detection wavelength was 274 nm. The linearity is obeyed over a concentration range of 0.5-150 µg.mL-1 with correlation coefficient of 0.999 for both the drugs. The proposed method was validated by determining accuracy, precision, stability and system suitability parameters. The method was found to be robust. Specificity of the method was determined by subjecting the drugs to various stress conditions like acid, alkali, oxidation, thermal and photolytic degradation. The method was used successfully for the simultaneous determination of levosalbutamol sulfate and theophylline in syrup dosage form.Desenvolveu-se e validou-se método de RP-HPLC novo, simples, exato e preciso de determinação simultânea do sulfato de levossalbutamol e teofilina.. O comprimento de onda de detecção no UV foi 274 nm. Observou-se linearidade na faixa de concentração de 0,5-150 µg mL-1, com coeficiente de correlação de 0,999 para ambos os fármacos. O método proposto foi validado determinando-se exatidão, precisão, estabilidade e parâmetros de adequação do sistema. O método mostrou-se robusto. A especificidade do método foi determinada submetendo os fármacos a várias condições de estresse, como ácido, álcali, oxidação, degradação térmica e fotolítica. O método foi usado com sucesso para a determinação simultânea do sulfato de levossalbutamol e teofilina na forma de xarope
Clinical Assessment of the Abutment Teeth of Children Undergoing Space Maintainer Therapy- A Randomised Controlled Trial
Introduction:
The band and loop space maintainer are one of the most commonly used fixed space maintainers in children. However, prolonged use of appliances in the oral cavity may lead to changes in gingival health and an increased risk of caries. This study aims to evaluate the clinical parameters of the abutment teeth, specifically the banded teeth and the tooth to which the loop is extended.
Materials And Methods:
This prospective clinical study included 35 children between the age group of 5 to 9 years. The abutment teeth were evaluated on bleeding on probing, pocket depth, gingival index, and occurrence of primary/ secondary caries.
Results:
There is a significant difference in the bleeding on probing, pocket depth, gingival index, and occurrence of primary/ secondary caries (p\u3c0.05).
Conclusion: Precautions and thorough knowledge about the consequences of the delivery of appliances can prevent and overcome the causes. Oral health education and constant motivation should be provided to the children on dental hygiene and regular follow-ups to avoid any extensive invasive management
Cryptococcal infection causing longitudinal extensive transverse myelitis in an immunocompetent individual: Case report and literature review
Cryptococcal CNS infections in immunocompetent individuals are occasionally reported in literature. The spinal manifestations of cryptococcal CNS infections are epidural abscess, chronic arachnoiditis, intramedullary granuloma, myelitis and vasculitis. We report a rare case of CNS cryptococcal infection presenting as a longitudinal extensive transverse myelitis (LETM) in an immunocompetent male. This report highlights cryptococcus as an important etiology among infectious causes in acute LETM patients in-spite of the immunocompetent status of the patient and the utility of CRAG (cryptococcal antigen) for diagnosis in such patients. We also present a literature review of all reported cases of cryptococcal myelitis
FORMULATION AND DEVELOPMENT OF KETOROLAC TROMETHAMINE OPHTHALMIC SOLUTION
An acetic acid derivative ketorolac tromethmine has found its applicability in both gram positive and gram negative bacterial ocular infection and used commonly associated with multiple doses. Ophthalmic medication stored in multiple dose containers is required by the U.S. Food and Drug Administration to contain a preservative so that patients are provided with microbe free medication. Benzalkonium chloride in concentrations from 0.1% to 0.0001% induced dose-dependent growth arrest and conjunctiva epithelial cell death, either delayed or immediately after administration. In such case, a preservative Benzalkonium chloride must be used within reasonable bound. Benzalkonium chloride can provide more help than harm. Hence the present study focusing on to formulate a formulation for ketorolac tromethmine (0.5%) ophthalmic solution using different concentration of Benzalkonium chloride as preservative. Structure-based assortment of herbal analogues against spike protein to restrict COVID-19 entry through hACE2 receptor : an in-silico approach
On-going global pandemic COVID-19 has spread all over the world and has led to more than 1.97 million deaths till date. Natural compounds may be useful to protecting health in this perilous condition. Mechanism of shuttle entry of SARS-COV-2 virus is by interaction with viral spike protein with human angiotensin-converting enzyme-2 (ACE-2) receptor. To explore potential natural therapeutics, 213 important phytochemi-cals of nine medicinal plants Aconitum heterophyllum, Cassia angustifolia, Cymbopogon flexuosus, Cymbopogon martinii, Nux vomica, Phyllanthus urinaria, Swertia chirayita, Justicia adhatoda, Vetiveria zizanioides were selected for in-silico molecular docking against the spike protein of SARS-COV-2 and compared with recently prescribed drug chloroquine, ramdesivir, lopinavir and hydroxychloroquine. Results revealed that rhamnocitrin of P. urinaria, 1,5-dihydroxy-3,8-dimethoxyxanthone of S. chirayita and laevojunenol of V. zizanioides potentially binds with the receptor binding site of SARS-COV-2 spike glycoprotein and more robustly destabilized the RBD-ACE-2 binding over chloroquine, ramdesivir, lopinavir and hydroxychloroquine. It was also found that laevojunenol, rhamnocitrin, and 1,5-dihydroxy-3,8-dimethoxyxanthone qualified the criteria for drug-likeness as per Lipinski rule. After attachment of the selected phytochemical with the spike protein the affinity of the later towards ACE-2 was minimized and the effect of 1,5-dihydroxy-3,8-dimethoxyxanthone and laevojunenol was superior. Hence, rhamnocitrin of P. urinaria, 1,5-dihydroxy-3,8-dimethoxyxanthone of S. chirayita and laevojunenol of V. zizanioides, are potential therapeutic molecules for SARS-COV-2, which upon binding with spike protein changes the affinity of the spike towards ACE-2 and therefore restrict the entry of the virus into a human cell. Subsequent clinical validation is needed to confirm these phytochemicals as drugs to combat COVID-19
Design and Fabrication of Liquid Pressure Sensor using FBG Sensor through Seesaw Hinge Mechanism
Pressure sensors are used in various industrial applications assisting in preventing unintended disasters. This paper presents the design and fabrication of a novel Seesaw device incorporating a diaphragm and Fiber Bragg Grating (FBG) sensor to measure the pressure of liquids. The designed sensor has been tested in a static water column. The proposed design enables the user to easily make and modify the diaphragm based on the required pressure range without interfering with the FBG sensor. The developed pressure sensor produces improved accuracy and sensitivity to applied liquid pressure in both low and high-pressure ranges without requiring sophisticated sensor construction. A finite element analysis has been performed on the diaphragm and on the entire structure at 10 bar pressure. The deformation of the diaphragm is comparable to theoretical deformation levels. A copper diaphragm with a thickness of 0.25 mm is used in the experiments. All experiments are performed in the elastic region of the diaphragm. The sensor’s sensitivity as 19.244 nm/MPa with the linearity of 99.64% is obtained based on the experiments.
