    Free radical theory of autoimmunity

    BACKGROUND: Despite great advances in clinical oncology, the molecular mechanisms underlying the failure of chemotherapeutic intervention in treating lymphoproliferative and related disorders are not well understood. HYPOTHESIS: A hypothetical scheme to explain the damage induced by chemotherapy and associated chronic oxidative stress is proposed on the basis of published literature, experimental data and anecdotal observations. Brief accounts of multidrug resistance, lymphoid malignancy, the cellular and molecular basis of autoimmunity and chronic oxidative stress are assembled to form a basis for the hypothesis and to indicate the likelihood that it is valid in vivo. CONCLUSION: The argument set forward in this article suggests a possible mechanism for the development of autoimmunity. According to this view, the various sorts of damage induced by chemotherapy have a role in the pattern of drug resistance, which is associated with the initiation of autoimmunity

    BACKGROUND: It is known that sensitivity or resistance of tumor cells to a given chemotherapeutic agent is an acquired characteristic(s), depending on the heterogeneity of the tumor mass subjected to the treatment. The clinical success of a chemotherapeutic regimen depends on the ratio of sensitive to resistant cell populations. RESULTS: Based on findings from clinical and experimental studies, a unifying model is proposed to delineate the potential mechanism by which tumor cells progress towards multi drug resistance, resulting in failure of chemotherapy. CONCLUSION: It is suggested that the evolution of multi drug resistance is a developmentally orchestrated event. Identifying stage-specific time windows during this process would help to identify valid therapeutic targets for the effective elimination of malignancy

    A rare case of conjoined twins presenting as dicephalus parapagus: a case report

    Conjoined twinning is a congenital abnormality resulting in various presentations of the fetuses in utero. It is a rare embryological developmental disorder occurring in one in 100,000 births. We present a case of conjoined twinning, which is characterized by the presence of two heads, single vertebral column and single symphysis pubis, with single genitor-urinary tract and single gastro intestinal tract. The present condition, referred to as dicephalus parapagus, is a rare entity among the conjoined twinning and has seldom positive outcomes. The mechanism of occurrence of conjoined twinning has been proposed either through fusion or fission of the embryos. In either case, surgical separation of the conjoined twins is often unsuccessful and results in increased morbidity and mortality. However, it is essential to screen for the presence of such congenital anomalies early during pregnancy, in order to terminate and provide parental counselling

    Context Based Classification of Reviews Using Association Rule Mining, Fuzzy Logics and Ontology

    The Internet has facilitated the growth of recommendation system owing to the ease of sharing customer experiences online. It is a challenging task to summarize and streamline the online textual reviews. In this paper, we propose a new framework called Fuzzy based contextual recommendation system. For classification of customer reviews we extract the information from the reviews based on the context given by users. We use text mining techniques to tag the review and extract context. Then we find out the relationship between the contexts from the ontological database. We incorporate fuzzy based semantic analyzer to find the relationship between the review and the context when they are not found therein. The sentence based classification predicts the relevant reviews, whereas the fuzzy based context method predicts the relevant instances among the relevant reviews. Textual analysis is carried out with the combination of association rules and ontology mining. The relationship between review and their context is compared using the semantic analyzer which is based on the fuzzy rules

    Establishment of targeted mutagenesis in soybean protoplasts using CRISPR/Cas9 RNP delivery via electro−transfection

    The soybean (Glycine max L.) is an important crop with high agronomic value. The improvement of agronomic traits through gene editing techniques has broad application prospects in soybean. The polyethylene glycol (PEG)-mediated cell transfection has been successfully used to deliver the CRISPR/Cas9-based ribonucleoprotein (RNP) into soybean protoplasts. However, several downstream analyses or further cell regeneration protocols might be hampered by PEG contamination within the samples. Here in this study, we attempted to transfect CRISPR/Cas9 RNPs into trifoliate leaf-derived soybean protoplasts using Neon electroporation to overcome the need for PEG transfection for the first time. We investigated different electroporation parameters including pulsing voltage (V), strength and duration of pulses regarding protoplast morphology, viability, and delivery of CRISPR/Cas9. Electroporation at various pulsing voltages with 3 pulses and 10 ms per pulse was found optimal for protoplast electro-transfection. Following electro-transfection at various pulsing voltages (500 V, 700 V, 1,000 V, and 1,300 V), intact protoplasts were observed at all treatments. However, the relative frequency of cell viability and initial cell divisions decreased with increasing voltages. Confocal laser scanning microscopy (CLSM) confirmed that the green fluorescent protein (GFP)-tagged Cas9 was successfully internalized into the protoplasts. Targeted deep sequencing results revealed that on-target insertion/deletion (InDel) frequencies were increased with increasing voltages in protoplasts electro-transfected with CRISPR/Cas9 RNPs targeting constitutive pathogen response 5 (CPR5). InDel patterns ranged from +1 bp to −6 bp at three different target sites in CPR5 locus with frequencies ranging from 3.8% to 8.1% following electro-transfection at 1,300 V and 2.1% to 3.8% for 700 V and 1,000 V, respectively. Taken together, our results demonstrate that the CRISPR/Cas9 RNP system can be delivered into soybean protoplasts by the Neon electroporation system for efficient and effective gene editing. The electro-transfection system developed in this study would also further facilitate and serve as an alternative delivery method for DNA-free genome editing of soybean and other related species for genetic screens and potential trait improvement.publishedVersio

    Mapping the research output of Medicinal Mushroom: A Scientometric Study

    This paper attempts to show up the growth trends on Medicinal Mushroom literature and make the quantitative and qualitative assessment by analyzing various features of research output and Citations impact based on the Scopus database. A total of 3513 publications were published on Medicinal Mushroom during 1990-2020. Year 2019 has highest publication of 363 whereas the year 2015 scores the highest citation impact of 5612. The People\u27s Republic of China has the highest share 1125 of publications and received 19435 Citations followed by Taiwan with 5311 Citation for 288 publications, India with 3087 Citations for 275 publications. It’s noteworthy that USA moves the Taiwan to third place through citations wise analysis with less number of publications compared to Taiwan and India. The highly productive Institutions were: Chinese Academy of Science with 152 publications having citation of 2921 followed by University Malaya with 1849 citations having 110 records and National Chung Hsing University with 83 publications. The highly productive journals are INTERNATIONAL JOURNAL OF MEDICINAL MUSHROOMS with 1004 Publications and having citation of 7798, followed by JOURNAL OF ETHNOPHARMACOLOGY and MOLECULES with 65 publications having citation of 2035 and 912 respectively. There were 126 single Author publications by the scientists and remaining were multi-Author collaborative publications. The Collaboration Index is 3.30. The average citation per document is 17.7 and an average citation per year per document is 2.06

    Etiology and risk factors among young patients presenting with stroke in a tertiary care hospital in South India

    Background: Stroke is one of the leading causes of disease and death throughout the world with the incidence rising steeply with age. The occurrence of stroke in young is rare but once it occurs the outcome is severe. Limited studies have been performed in the Indian population to describe the clinical presentation and etiology of young adults with stroke. The objective of present study is to assess the etiology, risk factors and clinical presentation among patients who visit a tertiary care centre with a clinical diagnosis of stroke in individuals less than 45 years of age.Methods: Consecutive patients visiting the Dept of General Medicine & Neurology, SRM Hospital aged between 15-45 years with abrupt onset of focal or global neurological deficit attributable to vascular cause and persisting for more than 24 hours were included into the study. Patients with traumatic injury and transient ischemic attack were excluded. The clinical features, laboratory and radiological investigations were obtained from the patient’s records.Results: The mean age of the study group was 36.42±6.92 years. The most common risk factor amongst study subjects was smoking (36%). Hemiplegia was more commonly seen in males (38 vs. 14 %, p= 0.26) than in females. Seizures were more commonly seen among subjects with hemorrhagic stroke and cortical venous thrombosis (p= 0.001). Among   subjects with ischemic stroke, the most common etiology was hypercoagulation. The most common arterial territory involved was Middle cerebral artery territory 84.84% with the  Left MCA (53.5%) being  more common than right MCA (46.4%).Conclusions: The clinical presentation and etiology among young patients with stroke appear to remain consistent with that reported earlier in the literature. The paucity of rare presentations of stroke in our study could be attributable to the limited sample size of our study.

    Methane and n-hexane ignition in a newly developed diaphragmless shock tube

    Shock tubes have been routinely used to generate reliable chemical kinetic data for gas-phase chemistry. The conventional diaphragm-rupture mode for shock tube operation presents many challenges that may ultimately affect the quality of chemical kinetics data. Numerous diaphragmless concepts have been developed to overcome the drawbacks of using diaphragms. Most of these diaphragmless designs require significant alterations in the driver section of the shock tube and, in some cases, fail to match the performance of the diaphragm-mode of operation. In the present work, an existing diaphragm-type shock tube is retrofitted with a fast-acting valve, and the performance of the diaphragmless shock tube is evaluated for investigating the ignition of methane and n-hexane. The diaphragmless shock tube reported here presents many advantages, such as eliminating the use of diaphragms, avoiding substantial manual effort during experiments, automating the shock tube facility, having good control over driver conditions, and obtaining good repeatability for reliable gas-phase chemical kinetic studies. Ignition delay time measurements have been performed in the diaphragmless shock tube for three methane mixtures and two n-hexane mixtures at P5P_5 = 10 - 20 bar and T5T_5 = 738 - 1537 K. The results obtained for fuel-rich, fuel-lean, and oxygen-rich (undiluted) mixtures show very good agreement with previously reported experimental data and literature kinetic models (AramcoMech 3.0 [1] for methane and Zhang et al. mechanism [2] for n-hexane). The study presents an easy and simple method to upgrade conventional shock tubes to a diaphragmless mode of operation and opens new possibilities for reliable chemical kinetics investigations.Comment: 22 pages, 9 figure
