    Young Netherlands Twin Register (Y-NTR): A longitudinal multiple informant study of problem behavior.

    The Netherlands Twin Register (NTR) was established around 1987 at the Vrije Universiteit in Amsterdam, the Netherlands. The current article summarizes the longitudinal genetic analyses of maternal and paternal ratings of twins' behavior as a function of the sex of the children for the traits of aggression (AGG), attention problems (AP), anxious/depression (ANX), internalizing behavior (INT) and externalizing behavior (EXT). We found that genetic influences are the most important factor in explaining individual differences in these traits. For most phenotypes, influences of genetic factors fluctuate throughout development, with the exception of AP, for which genetic influences remain of similar magnitude. Changes in genetic influences parallel those in shared environmental influences, while nonshared environmental influences remain relatively constant. Around 10% to 20% of the variance is accounted for by parent-specific shared environment, which includes rater bias. For all phenotypes, stability throughout childhood is accounted for by genetic and shared environmental factors, while nonshared environmental influences are mainly age/measurement specific. About 15% of the phenotypic stability is accounted for by rater-specific shared environmental influences, which include rater bias. In conclusion, between ages 3 and 12 genetic factors are the most important cause of individual differences in emotional and behavioral problem

    Schoolsucces als de scholen dicht zijn?: Een onderzoek naar de rol van ondersteuning door leerkrachten en ouders tijdens de coronacrisis

    In dit vragenlijst-onderzoek onderzochten we het ervaren schoolsucces van leerlingen in de bovenbouw van het basisonderwijs (N = 667) en hoe dat samenhing met de ondersteuning die ze ontvingen van leraren en ouders tijdens de scholensluiting in het voorjaar van 2020. Ook vroegen we een deel van de ouders (N = 174) hoe zij de relatie met leerkrachten hebben ervaren tijdens het afstandsonderwijs. Ervaren schoolsucces vatten we op als de motivatie, zelfregulatie en ervaren schoolprestaties (“hoeveel heb ik geleerd?”). De resultaten van het onderzoek laten zien dat leerkrachtondersteuning samenhing met motivatie en zelfregulatie. Ouderbegeleiding hing samen met alle drie de indicatoren van schoolsucces. Leerkracht-ouderrelatie, ten slotte, hing alleen samen met motivatie. Het is opvallend dat leerlingen over het algemeen aangaven evenveel te hebben geleerd als in de normale situatie, terwijl eerder onderzoek naar de effecten van de scholensluiting op toetsresultaten een gemiddeld leerverlies liet zien. Ten slotte rapporteerden oudere leerlingen (groep 7 en 8) lagere prestaties evenals een lager niveau van motivatie dan de jongere groep leerlingen (groep 6). We bespreken implicaties van onze uitkomsten voor het afstandsonderwijs zelf, maar ook voor de periode daarna. Het is onder meer interessant dat leerkrachtondersteuning belangrijk blijkt voor motivatie en zelfregulatie, zelfs als er geen fysiek contact is. De resultaten impliceren bovendien dat het belangrijk is ruimte en voldoende ondersteuning te bieden aan ouders, zodat zij hun kinderen goed kunnen begeleiden.Education and Child Studie

    In Vivo Evaluation of Indium-111-Labeled 800CW as a Necrosis-Avid Contrast Agent.

    Current clinical measurements for tumor treatment efficiency rely often on changes in tumor volume measured as shrinkage by CT or MRI, which become apparent after multiple lines of treatment and pose a physical and psychological burden on the patient. Detection of therapy-induced cell death in the tumor can be a fast measure for treatment efficiency. However, there are no reliable clinical tools for detection of tumor necrosis. Previously, we studied the necrosis avidity of cyanine-based fluorescent dyes, which suffered long circulation times before tumor necrosis could be imaged due to low hydrophilicity. We now present the application of radiolabeled 800CW, a commercially available cyanine with high hydrophilicity, to image tumor necrosis in a mouse model. We conjugated 800CW to DOTA via a PEG linker, for labeling with single-photon emission-computed tomography isotope indium-111, yielding [ <sup>111</sup> In]In-DOTA-PEG <sub>4</sub> -800CW. We then investigated specific [ <sup>111</sup> In]In-DOTA-PEG <sub>4</sub> -800CW uptake by dead cells in vitro, using both fluorescence and radioactivity as detection modalities. Finally, we investigated [ <sup>111</sup> In]In-DOTA-PEG <sub>4</sub> -800CW uptake into necrotic tumor regions of a 4T1 breast tumor model in mice. We successfully prepared a precursor and developed a reliable procedure for labeling 800CW with indium-111. We detected specific [ <sup>111</sup> In]In-DOTA-PEG <sub>4</sub> -800CW uptake by dead cells, using both fluorescence and radioactivity. Albeit with a tumor uptake of only 0.37%ID/g at 6 h post injection, we were able to image tumor necrosis with a tumor to background ratio of 7:4. Fluorescence and radioactivity in cryosections from the dissected tumors were colocalized with tumor necrosis, confirmed by TUNEL staining. [ <sup>111</sup> In]In-DOTA-PEG <sub>4</sub> -800CW can be used to image tumor necrosis in vitro and in vivo. Further research will elucidate the application of [ <sup>111</sup> In]In-DOTA-PEG <sub>4</sub> -800CW or other radiolabeled hydrophilic cyanines for the detection of necrosis caused by chemotherapy or other anti-cancer therapies. This can provide valuable prognostic information in treatment of solid tumors

    Necrosis binding of Ac-Lys<sup>0</sup>(IRDye800CW)-Tyr<sup>3</sup>-octreotate: a consequence from cyanine-labeling of small molecules.

    There is a growing body of nuclear contrast agents that are repurposed for fluorescence-guided surgery. New contrast agents are obtained by substituting the radioactive tag with, or adding a fluorescent cyanine to the molecular structure of antibodies or peptides. This enables intra-operative fluorescent detection of cancerous tissue, leading to more complete tumor resection. However, these fluorescent cyanines can have a remarkable influence on pharmacokinetics and tumor uptake, especially when labeled to smaller targeting vectors such as peptides. Here we demonstrate the effect of cyanine-mediated dead cell-binding of Ac-Lys &lt;sup&gt;0&lt;/sup&gt; (IRDye800CW)-Tyr &lt;sup&gt;3&lt;/sup&gt; -octreotate (800CW-TATE) and how this can be used as an advantage for fluorescence-guided surgery. Binding of 800CW-TATE could be blocked with DOTA &lt;sup&gt;0&lt;/sup&gt; -Tyr &lt;sup&gt;3&lt;/sup&gt; -octreotate (DOTA-TATE) on cultured SSTR &lt;sub&gt;2&lt;/sub&gt; -positive U2OS cells and was absent in SSTR &lt;sub&gt;2&lt;/sub&gt; negative U2OS cells. However, strong binding was observed to dead cells, which could not be blocked with DOTA-TATE and was also present in dead SSTR &lt;sub&gt;2&lt;/sub&gt; negative cells. No SSTR &lt;sub&gt;2&lt;/sub&gt; -mediated binding was observed in frozen tumor sections, possibly due to disruption of the cells in the process of sectioning the tissue before exposure to the contrast agent. DOTA-TATE blocking resulted in an incomplete reduction of 61.5 ± 5.8% fluorescence uptake by NCI-H69-tumors in mice. Near-infrared imaging and dead cell staining on paraffin sections from resected tumors revealed that fluorescence uptake persisted in necrotic regions upon blocking with DOTA-TATE. This study shows that labeling peptides with cyanines can result in dead cell binding. This does not hamper the ultimate purpose of fluorescence-guided surgery, as necrotic tissue appears in most solid tumors. Hence, the necrosis binding can increase the overall tumor uptake. Moreover, necrotic tissue should be removed as much as possible: it cannot be salvaged, causes inflammation, and is tumorigenic. However, when performing binding experiments to cells with disrupted membrane integrity, which is routinely done with nuclear probes, this dead cell-binding can resemble non-specific binding.     Design and implementation of a twin-family database for behavior genetics and genomics studies.

    In this article we describe the design and implementation of a database for extended twin families. The database does not focus on probands or on index twins, as this approach becomes problematic when larger multigenerational families are included, when more than one set of multiples is present within a family, or when families turn out to be part of a larger pedigree. Instead, we present an alternative approach that uses a highly flexible notion of persons and relations. The relations among the subjects in the database have a one-to-many structure, are user-definable and extendible and support arbitrarily complicated pedigrees. Some additional characteristics of the database are highlighted, such as the storage of historical data, predefined expressions for advanced queries, output facilities for individuals and relations among individuals and an easy-to-use multi-step wizard for contacting participants. This solution presents a flexible approach to accommodate pedigrees of arbitrary size, multiple biological and nonbiological relationships among participants and dynamic changes in these relations that occur over time, which can be implemented for any type of multigenerational family study

    Evaluation of Ac-Lys(0)(IRDye800CW)Tyr(3)-octreotate as a novel tracer for SSTR2-targeted molecular fluorescence guided surgery in meningioma

    PURPOSE: Meningioma recurrence rates can be reduced by optimizing surgical resection with the use of intraoperative molecular fluorescence guided surgery (MFGS). We evaluated the potential of the fluorescent tracer 800CW-TATE for MFGS using in vitro and in vivo models. It targets somatostatin receptor subtype 2 (SSTR(2)), which is overexpressed in all meningiomas. METHODS: Binding affinity of 800CW-TATE was evaluated using [(177)Lu] Lu-DOTA-Tyr(3)-octreotate displacement assays. Tumor uptake was determined by injecting 800CW-TATE in (SSTR(2)-positive) NCI-H69 or (SSTR(2)-negative) CH-157MN xenograft bearing mice and FMT2500 imaging. SSTR(2)-specific binding was measured by comparing tumor uptake in NCI-H69 and CH-157MN xenografts, blocking experiments and non-targeted IRDye800CW-carboxylate binding. Tracer distribution was analyzed ex vivo, and the tumor-to-background ratio (TBR) was calculated. SSTR(2) expression was determined by immunohistochemistry (IHC). Lastly, 800CW-TATE was incubated on frozen and fresh meningioma specimens and analyzed by microscopy. RESULTS: 800CW-TATE binding affinity assays showed an IC(50) value of 72 nM. NCI-H69 xenografted mice showed a TBR of 21.1. 800CW-TATE detection was reduced after co-administration of non-fluorescent DOTA-Tyr(3)-octreotate or administration of IRDye800CW. CH-157MN had no tumor specific tracer staining due to absence of SSTR(2) expression, thereby serving as a negative control. The tracer bound specifically to SSTR(2)-positive meningioma tissues representing all WHO grades. CONCLUSION: 800CW-TATE demonstrated sufficient binding affinity, specific SSTR(2)-mediated tumor uptake, a favorable biodistribution, and high TBR. These features make this tracer very promising for use in MFGS and could potentially aid in safer and a more complete meningioma resection, especially in high-grade meningiomas or those at complex anatomical localizations.
