    Edward Stevens is the new Deputy Grand Worthy Patriarch for the Monroe and Amanda Divisions of the Sons of Temperance. He sends a report to Finley, who is currently serving as Grand Worthy Patriarch. Stevens reports that there is apathy in his region regarding the cause of temperance. He shares several ideas with Finley about improving the efficacy of the Sons of Temperance -- advocating limits regarding the sale of liquor, paying closer attention to the beautiful ceremonial rites of the order, adopting an interchange of officers, and the universal dissemination of temperance tracts.     Separation of bacterial spores from flowing water in macro-scale cavities by ultrasonic standing waves

    The separation of micron-sized bacterial spores (Bacillus cereus) from a steady flow of water through the use of ultrasonic standing waves is demonstrated. An ultrasonic resonator with cross-section of 0.0254 m x 0.0254 m has been designed with a flow inlet and outlet for a water stream that ensures laminar flow conditions into and out of the resonator section of the flow tube. A 0.01905-m diameter PZT-4, nominal 2-MHz transducer is used to generate ultrasonic standing waves in the resonator. The acoustic resonator is 0.0356 m from transducer face to the opposite reflector wall with the acoustic field in a direction orthogonal to the water flow direction. At fixed frequency excitation, spores are concentrated at the stable locations of the acoustic radiation force and trapped in the resonator region. The effect of the transducer voltage and frequency on the efficiency of spore capture in the resonator has been investigated. Successful separation of B. cereus spores from water with typical volume flow rates of 40-250 ml/min has been achieved with 15% efficiency in a single pass at 40 ml/min.Comment: 11 pages, 6 figure

    Hemodynamic effects of intravenous amiodarone

    Amiodarone is a potent antiarrhythmic agent that is effective in controlling both atrial and ventricular arrhythmias. Recently, intravenous administration was demonstrated to be effective in the acute management of rhythm disorders and, in addition, appeared to shorten the loading period normally required for oral drug administration. This investigation examined the hemodynamic effects of amiodarone after both acute intravenous bolus and continuous intravenous administration. Patients with a left ventricular ejection fraction greater than 0.35 experienced improved cardiac performance due to both acute and chronic peripheral vasodilation. However, patients with a lower ejection fraction developed a 20% decrease in cardiac index and clinically significant elevation of right heart pressures after acute bolus administration; these changes were variably compensated for by peripheral vasodilation when the drug was administered intravenously over 3 to 5 days continuously. Therefore, intravenous amiodarone can result in significant impairment of left ventricular performance in patients with preexisting left ventricular dysfunction

    An assessment of the correlation between robust CT-derived ventilation and pulmonary function test in a cohort with no respiratory symptoms

    OBJECTIVE: To evaluate CT-ventilation imaging (CTVI) within a well-characterized, healthy cohort with no respiratory symptoms and examine the correlation between CTVI and concurrent pulmonary function test (PFT). METHODS: CT scans and PFTs from 77 Caucasian participants in the NORM dataset (clinicaltrials.gov NCT00848406) were analyzed. CTVI was generated using the robust Integrated Jacobian Formulation (IJF) method. IJF estimated total lung capacity (TLC) was computed from CTVI. Bias-adjusted Pearson’s correlation between PFT and IJF-based TLC was computed. RESULTS: IJF- and PFT-measured TLC showed a good correlation for both males and females [males: 0.657, 95% CI (0.438–0.797); females: 0.667, 95% CI (0.416–0.817)]. When adjusting for age, height, smoking, and abnormal CT scan, correlation moderated [males: 0.432, 95% CI (0.129–0.655); females: 0.540, 95% CI (0.207–0.753)]. Visual inspection of CTVI revealed participants who had functional defects, despite the fact that all participant had normal high-resolution CT scan. CONCLUSION: In this study, we demonstrate that IJF computed CTVI has good correlation with concurrent PFT in a well-validated patient cohort with no respiratory symptoms. ADVANCES IN KNOWLEDGE: IJF-computed CTVI’s overall numerical robustness and consistency with PFT support its potential as a method for providing spatiotemporal assessment of high and low function areas on volumetric non-contrast CT scan

    Janus: Launch of a NASA SmallSat Mission to Near-Earth Binary Asteroids

    Janus is a two-spacecraft SmallSat mission to fly by two different pairs of binary near Earth asteroids, (175706) 1996 FG3 and (35107) 1991 VH. The two identical Janus spacecraft are scheduled to launch during a launch period opening 1 August 2022 as secondary payloads with the NASA Psyche mission, on a SpaceX Falcon Heavy launch vehicle. Janus is led by principal investigator Dr. Dan Scheeres at the University of Colorado Boulder and managed, built, and operated by Lockheed Martin. These planetary SmallSats share many deep space challenges similar to larger missions: Janus must execute deep space maneuvers to achieve hundreds of meters per second ΔV to reach its destinations, close a telecommunication link at ranges up to 2.4 AU, autonomously manage a several-month-long telecommunications blackout during solar conjunction, operate at a maximum Sun range of 1.62 AU, and survive for approximately four years in interplanetary space before encountering their target asteroids. During the encounters, the spacecraft will return high resolution visible and infra-red images of the asteroids. In getting Janus to the pad, the implementation team successfully managed an aggressive mission schedule despite COVID-19 related supply chain impacts and work environments, all while remaining on target for the SIMPLEx-2 cost cap. Janus is a pathfinder for achievable and affordable SmallSat science missions and demonstrates the valuable partnership between an experienced deep space mission engineering team, the SmallSat commercial component industry, and a forward- looking NASA model for Class-D science missions

    Rhodium(II) Proximity-Labeling Identifies a Novel Target Site on STAT3 for Inhibitors with Potent Anti-Leukemia Activity

    Nearly 40 % of children with acute myeloid leukemia (AML) suffer relapse arising from chemoresistance, often involving upregulation of the oncoprotein STAT3 (signal transducer and activator of transcription 3). Herein, rhodium(II)-catalyzed, proximity-driven modification identifies the STAT3 coiled-coil domain (CCD) as a novel ligand-binding site, and we describe a new naphthalene sulfonamide inhibitor that targets the CCD, blocks STAT3 function, and halts its disease-promoting effects in vitro, in tumor growth models, and in a leukemia mouse model, validating this new therapeutic target for resistant AML

    Correlations between life-detection techniques and implications for sampling site selection in planetary analog missions

    We conducted an analog sampling expedition under simulated mission constraints to areas dominated by basaltic tephra of the Eldfell and Fimmvörðuháls lava fields (Iceland). Sites were selected to be “homogeneous” at a coarse remote sensing resolution (10–100 m) in apparent color, morphology, moisture, and grain size, with best-effort realism in numbers of locations and replicates. Three different biomarker assays (counting of nucleic-acid-stained cells via fluorescent microscopy, a luciferin/luciferase assay for adenosine triphosphate, and quantitative polymerase chain reaction (qPCR) to detect DNA associated with bacteria, archaea, and fungi) were characterized at four nested spatial scales (1 m, 10 m, 100 m, and >1 km) by using five common metrics for sample site representativeness (sample mean variance, group F tests, pairwise t tests, and the distribution-free rank sum H and u tests). Correlations between all assays were characterized with Spearman's rank test. The bioluminescence assay showed the most variance across the sites, followed by qPCR for bacterial and archaeal DNA; these results could not be considered representative at the finest resolution tested (1 m). Cell concentration and fungal DNA also had significant local variation, but they were homogeneous over scales of >1 km. These results show that the selection of life detection assays and the number, distribution, and location of sampling sites in a low biomass environment with limited a priori characterization can yield both contrasting and complementary results, and that their interdependence must be given due consideration to maximize science return in future biomarker sampling expeditions
