Anti-cyanobacterial activity of Moringa oleifera seeds
Filtrates from crushed Moringa oleifera seeds were tested for their effects on growth and Photosystem II efficiency of the common bloom-forming cyanobacterium Microcystis aeruginosa. M. aeruginosa populations exhibited good growth in controls and treatments with 4- and 8-mg crushed Moringa seeds per liter, having similar growth rates of 0.50 (±0.01) per day. In exposures of 20- to 160-mg crushed Moringa seeds Lâ1, growth rates were negative and on average â0.23 (±0.05) .dayâ1. Presumably, in the higher doses of 20- to 160-mg crushed seeds per liter, the cyanobacteria died, which was supported by a rapid drop in the Photosystem II efficiency (ΊPSII), while the ΊPSII was high and unaffected in 0, 4, and 8 mg Lâ1. High-density populations of M. aeruginosa (chlorophyll-a concentrations of âŒ270 ”g Lâ1) were reduced to very low levels within 2 weeks of exposure to â„80-mg crushed seeds per liter. At the highest dosage of 160 mg Lâ1, the ΊPSII dropped to zero rapidly and remained nil during the course of the experiment (14 days). Hence, under laboratory conditions, a complete wipeout of the bloom could be achieved. This is the first study that yielded evidence for cyanobactericidal activity of filtrate from crushed Moringa seeds, suggesting that Moringa seed extracts might have a potential as an effect-oriented measure lessening cyanobacterial nuisance
Transcriptional responses underlying the hormetic and detrimental effects of the plant secondary metabolite gossypol on the generalist herbivore Helicoverpa armigera
<p>Abstract</p> <p>Background</p> <p>Hormesis is a biphasic biological response characterized by the stimulatory effect at relatively low amounts of chemical compounds which are otherwise detrimental at higher concentrations. A hormetic response in larval growth rates has been observed in cotton-feeding insects in response to increasing concentrations of gossypol, a toxic metabolite found in the pigment glands of some plants in the family Malvaceae. We investigated the developmental effect of gossypol in the cotton bollworm, <it>Helicoverpa armigera</it>, an important heliothine pest species, by exposing larvae to different doses of this metabolite in their diet. In addition, we sought to determine the underlying transcriptional responses to different gossypol doses.</p> <p>Results</p> <p>Larval weight gain, pupal weight and larval development time were measured in feeding experiments and a hormetic response was seen for the first two characters. On the basis of net larval weight gain responses to gossypol, three concentrations (0%, 0.016% and 0.16%) were selected for transcript profiling in the gut and the rest of the body in a two-color double reference design microarray experiment. Hormesis could be observed at the transcript level, since at the low gossypol dose, genes involved in energy acquisition such as ÎČ-fructofuranosidases were up-regulated in the gut, and genes involved in cell adhesion were down-regulated in the body. Genes with products predicted to be integral to the membrane or associated with the proteasome core complex were significantly affected by the detrimental dose treatment in the body. Oxidoreductase activity-related genes were observed to be significantly altered in both tissues at the highest gossypol dose.</p> <p>Conclusions</p> <p>This study represents the first transcriptional profiling approach investigating the effects of different concentrations of gossypol in a lepidopteran species. <it>H. armigera</it>'s transcriptional response to gossypol feeding is tissue- and dose-dependent and involves diverse detoxifying mechanisms not only to alleviate direct effects of gossypol but also indirect damage such as pH disturbance and oxygen radical formation. Genes discovered through this transcriptional approach may be additional candidates for understanding gossypol detoxification and coping with gossypol-induced stress. In a generalist herbivore that has evolved transcriptionally-regulated responses to a variety of different plant compounds, hormesis may be due to a lower induction threshold of growth-promoting, stress-coping responses and a higher induction threshold of detoxification pathways that are costly and cause collateral damage to the cell.</p
Do acid-tolerant picocyanobacteria exist? A study of two strains isolated from humic lakes in Poland
The significance of genome-wide transcriptional regulation in the evolution of stress tolerance.
It is widely recognized that stress plays an important role in directing the adaptive adjustment of an organism to changing environments. However, very little is known about the evolution of mechanisms that promote stress-induced variation. Adaptive transcriptional responses have been implicated in the evolution of tolerance to natural and anthropogenic stressors in the environment. Recent technological advances in transcriptomics provide a mechanistic understanding of biological pathways or processes involved in stress-induced phenotypic change. Furthermore, these studies are (semi) quantitative and provide insight into the reaction norms of identified target genes in response to specific stressors. We argue that plasticity in gene expression reaction norms may be important in the evolution of stress tolerance and adaptation to environmental stress. This review highlights the consequences of transcriptional plasticity of stress responses within a single generation and concludes that gene promoters containing a TATA box are more capable of rapid and variable responses than TATA-less genes. In addition, the consequences of plastic transcriptional responses to stress over multiple generations are discussed. Based on examples from the literature, we show that constitutive over expression of specific stress response genes results in stress adapted phenotypes. However, organisms with an innate capacity to buffer stress display plastic transcriptional responses. Finally, we call for an improved integration of the concept of phenotypic plasticity with studies that focus on the regulation of transcription. © Springer Science+Business Media B.V. 2010
