    Combining scanning haptic microscopy and fibre optic Raman spectroscopy for tissue characterization

    The tactile resonance method (TRM) and Raman spectroscopy (RS) are promising for tissue characterization in vivo. Our goal is to combine these techniques into one instrument, to use TRM for swift scanning, and RS for increasing the diagnostic power. The aim of this study was to determine the classification accuracy, using support vector machines, for measurements on porcine tissue and also produce preliminary data on human prostate tissue. This was done by developing a new experimental set-up combining micro-scale TRM—scanning haptic microscopy (SHM)—for assessing stiffness on a micro-scale, with fibre optic RS measurements for assessing biochemical content. We compared the accuracy using SHM alone versus SHM combined with RS, for different degrees of tissue homogeneity. The cross-validation classification accuracy for healthy porcine tissue types using SHM alone was 65–81%, and when RS was added it increased to 81–87%. The accuracy for healthy and cancerous human tissue was 67–70% when only SHM was used, and increased to 72–77% for the combined measurements. This shows that the potential for swift and accurate classification of healthy and cancerous prostate tissue is high. This is promising for developing a tool for probing the surgical margins during prostate cancer surgery

    Are the results of open randomised controlled trials comparing antipsychotic drugs in schizophrenia biased?:Exploratory meta- and subgroup analysis

    A recent meta-epidemiological study did not reveal major differences between the results of blinded and open randomised-controlled trials (RCTs). Fewer patients may consent to double-blind RCTs than to open RCTs, compromising generalisability, making this question very important. However, the issue has not been addressed in schizophrenia. We used a database of randomised, acute-phase antipsychotic drug trials. Whenever at least one open and one blinded RCT was available for a comparison of two drugs, we contrasted the results by random-effects meta-analysis with subgroup tests. The primary outcome was overall symptoms as measured by the Positive and Negative Syndrome Scale, supplemented by seven secondary efficacy and side-effect outcomes. We also examined whether open RCTs were biased in favour of more recently introduced antipsychotics, less efficacious or more prone to side-effects antipsychotics, and pharmaceutical sponsors. 183 RCTs (155 blinded and 28 open) with 34715 participants comparing two active drugs were available. The results did not suggest general differences between open and blinded RCTs, which examined two active drugs. Only 12 out of 122 subgroup tests had a p-value below 0.1, four below 0.05, and if a Bonferroni correction for multiple tests had been applied, only one would have been significant. There were some exceptions which, however, did not always confirm the originally hypothesized direction of bias. Due to the relatively small number of open RCTs, our analysis is exploratory, but this fundamental question should be given more scientific attention. Currently, open RCTs should be excluded from meta-analyses, at least in sensitivity analyses.</p

    Omega-3 polyunsaturated fatty acids and bronchial inflammation in grass pollen allergy after allergen challenge

    SummaryRatioAsthma is a major public health problem, with bronchial inflammation as the therapeutic target. The role of dietary fish oil derived polyunsaturated fatty acids (PUFAs) in allergic inflammation is controversial. Most asthmatics suffer from mild disease and non-pharmacologic interventions are attractive. This study investigates the anti-inflammatory potential of nutritional PUFAs in an experimentally induced bronchial inflammation.MethodsWe examined 38 grass pollen allergic asthmatics and 19 controls. History of dietary PUFA intake was compared with levels of PUFAs in erythrocyte membranes, and stratified according to low (25th quartile; Q25) and high (75th quartile; Q75) ratios of omega-3 (n-3) to omega-6 (n-6) PUFAs as a surrogate for anti-inflammatory (Q75) or proinflammatory (Q25) effects. Bronchial inflammation was simulated with one-step inhalation of grass pollen. Bronchial response (exhaled nitric monoxide, eNO as surrogate for inflammation, decrease of FEV1) was correlated with levels of PUFAs in erythrocyte membranes.ResultsRatios of n-3/n-6 PUFA were significantly lower in asthmatics than in healthy controls. Levels of eNO were significantly higher in Q25 asthmatics than in Q75 asthmatics (p = 0.040). There was a trend of higher bronchial hyperreactivity in Q25 asthmatics (median PD20 0.27 vs. 0.14; n.s.), induced by specific bronchial challenge with grass pollen (FEV1 decrease 16.7 vs. 23.1%; n.s.).ConclusionWhen stratifying for erythrocyte membrane PUFA content as a surrogate for alimentary intake, we found mild effects on bronchial allergic inflammation. Future intervention studies with pharmacological PUFA doses appear suitable to clarify dietary PUFA role as an adjunctive intervention to the established treatment of asthma.ClinicalTrials.gov No. NCT00519740

    Complex matching of RDF datatype properties

    Property mapping is a fundamental component of ontology matching, and yet there is little support that goes beyond the identification of single property matches. Real data often requires some degree of composition, trivially exemplified by the mapping of "first name" and "last name" to "full name" on one end, to complex matchings, such as parsing and pairing symbol/digit strings to SSN numbers, at the other end of the spectrum. In this paper, we propose a two-phase instance-based technique for complex datatype property matching. Phase 1 computes the Estimate Mutual Information matrix of the property values to (1) find simple, 1:1 matches, and (2) compute a list of possible complex matches. Phase 2 applies Genetic Programming to the much reduced search space of candidate matches to find complex matches. We conclude with experimental results that illustrate how the technique works. Furthermore, we show that the proposed technique greatly improves results over those obtained if the Estimate Mutual Information matrix or the Genetic Programming techniques were to be used independently.     ADAM10 is expressed in human podocytes and found in urinary vesicles of patients with glomerular kidney diseases

    <p>Abstract</p> <p>Background</p> <p>The importance of the Notch signaling in the development of glomerular diseases has been recently described. Therefore we analyzed in podocytes the expression and activity of ADAM10, one important component of the Notch signaling complex.</p> <p>Methods</p> <p>By Western blot, immunofluorescence and immunohistochemistry analysis we characterized the expression of ADAM10 in human podocytes, human urine and human renal tissue.</p> <p>Results</p> <p>We present evidence, that differentiated human podocytes possessed increased amounts of mature ADAM10 and released elevated levels of L1 adhesion molecule, one well known substrate of ADAM10. By using specific siRNA and metalloproteinase inhibitors we demonstrate that ADAM10 is involved in the cleavage of L1 in human podocytes. Injury of podocytes enhanced the ADAM10 mediated cleavage of L1. In addition, we detected ADAM10 in urinary podocytes from patients with kidney diseases and in tissue sections of normal human kidney. Finally, we found elevated levels of ADAM10 in urinary vesicles of patients with glomerular kidney diseases.</p> <p>Conclusions</p> <p>The activity of ADAM10 in human podocytes may play an important role in the development of glomerular kidney diseases.</p

    Development and testing of a pyro-driven launcher for harpoon-based comet sample acquisition

    The CORSAIR (COmet Rendezvous, Sample Acquisition, Investigation, and Return) mission is a proposal for the fourth NASA New Frontiers program. It belongs to the Comet Surface Sample Return mission theme which focuses on acquiring and returning to Earth a macroscopic sample from the surface of a comet nucleus. CORSAIR uses a harpoon-based Sample Acquisition System (SAS) with the spacecraft hovering several meters above the comet surface. This stand-off strategy overcomes disadvantages of systems using drills or shovels. Since comets are low gravity objects, these techniques would require anchoring before sampling, which is not necessary here. Moreover, the harpoon-based system allows for acquiring several samples from different locations on the comet maximizing the scientifc output of the mission. Each SAS assembly consists of a pyro-driven launcher, a Sample Acquisition and Retrieval Projectile (SARP) and a retraction system using a deployable composite boom structure. In order to collect enough cometary material, the launcher has to provide the required kinetic energy to the SARP. Due to high energy densities, pyrotechnically actuated devices ultimately reduce the overall system mass and dimensions. Here, an overview of the development, design and testing of the launcher is given.
