Interaction imaging with amplitude-dependence force spectroscopy
Knowledge of surface forces is the key to understanding a large number of
processes in fields ranging from physics to material science and biology. The
most common method to study surfaces is dynamic atomic force microscopy (AFM).
Dynamic AFM has been enormously successful in imaging surface topography, even
to atomic resolution, but the force between the AFM tip and the surface remains
unknown during imaging. Here, we present a new approach that combines high
accuracy force measurements and high resolution scanning. The method, called
amplitude-dependence force spectroscopy (ADFS) is based on the
amplitude-dependence of the cantilever's response near resonance and allows for
separate determination of both conservative and dissipative tip-surface
interactions. We use ADFS to quantitatively study and map the nano-mechanical
interaction between the AFM tip and heterogeneous polymer surfaces. ADFS is
compatible with commercial atomic force microscopes and we anticipate its
wide-spread use in taking AFM toward quantitative microscopy

Cytotoxicity, chemical stability, and surface properties of ferroelectric ceramics for biomaterials
© 2017 The American Ceramic Society Surface chemistry and topo-physical properties determine the interactions of biomaterials with their physiological environment. Ferroelectrics hold great promise as the next generation of scaffolds for tissue repair since they feature tunable surface electrical charges, piezoelectricity, and sensing capabilities. We investigate the topography, wettability, chemical stability, and cytotoxicity in salient ferroelectric systems such as (1âx) (Na1/2Bi1/2)TiO3âxBaTiO3, (1âx)Ba(Zr0.2Ti0.8)O3âx(Ba0.7Ca0.3)TiO3, and Pb(Zr,Ti)O3 to test their suitability as biomaterials. The lead-free ferroelectrics promote in vitro cell viability and proliferation to a considerably high extent. 0.94 mol % (Na1/2Bi1/2)TiO3â0.06 mol% BaTiO3 showed the greatest potential leading to a cell viability of (149 ± 30)% and DNA synthesis of (299 ± 85)% in comparison to the reference. Lead leaching from Pb(Zr,Ti)O3 negatively affected the cultured cells. Wettability and chemical stability are key factors that determine the cytotoxicity of ferroelectrics. These variables have to be considered in the design of novel electroactive scaffolds based on ferroelectric ceramics
Formation of regulatory modules by local sequence duplication
Turnover of regulatory sequence and function is an important part of
molecular evolution. But what are the modes of sequence evolution leading to
rapid formation and loss of regulatory sites? Here, we show that a large
fraction of neighboring transcription factor binding sites in the fly genome
have formed from a common sequence origin by local duplications. This mode of
evolution is found to produce regulatory information: duplications can seed new
sites in the neighborhood of existing sites. Duplicate seeds evolve
subsequently by point mutations, often towards binding a different factor than
their ancestral neighbor sites. These results are based on a statistical
analysis of 346 cis-regulatory modules in the Drosophila melanogaster genome,
and a comparison set of intergenic regulatory sequence in Saccharomyces
cerevisiae. In fly regulatory modules, pairs of binding sites show
significantly enhanced sequence similarity up to distances of about 50 bp. We
analyze these data in terms of an evolutionary model with two distinct modes of
site formation: (i) evolution from independent sequence origin and (ii)
divergent evolution following duplication of a common ancestor sequence. Our
results suggest that pervasive formation of binding sites by local sequence
duplications distinguishes the complex regulatory architecture of higher
eukaryotes from the simpler architecture of unicellular organisms

Levels of State and Trait Anxiety in Patients Referred to Ophthalmology by Primary Care Clinicians: A Cross Sectional Study
Purpose There is a high level of over-referral from primary eye care leading to significant numbers of people without ocular pathology (false positives) being referred to secondary eye care. The present study used a psychometric instrument to determine whether there is a psychological burden on patients due to referral to secondary eye care, and used Rasch analysis to convert the data from an ordinal to an interval scale. Design Cross sectional study. Participants and Controls 322 participants and 80 control participants. Methods State (i.e. current) and trait (i.e. propensity to) anxiety were measured in a group of patients referred to a hospital eye department in the UK and in a control group who have had a sight test but were not referred. Response category analysis plus infit and outfit Rasch statistics and person separation indices were used to determine the usefulness of individual items and the response categories. Principal components analysis was used to determine dimensionality. Main Outcome Measure Levels of state and trait anxiety measured using the State-Trait Anxiety Inventory. Results State anxiety scores were significantly higher in the patients referred to secondary eye care than the controls (p0.1). Rasch analysis highlighted that the questionnaire results needed to be split into âanxiety-absentâ and âanxiety-presentâ items for both state and trait anxiety, but both subscales showed the same profile of results between patients and controls. Conclusions State anxiety was shown to be higher in patients referred to secondary eye care than the controls, and at similar levels to people with moderate to high perceived susceptibility to breast cancer. This suggests that referral from primary to secondary eye care can result in a significant psychological burden on some patients
An experimental test of noncontextuality without unphysical idealizations
To make precise the sense in which nature fails to respect classical physics, one requires a formal notion of classicality. Ideally, such a notion should be defined operationally, so that it can be subject to direct experimental test, and it should be applicable in a wide variety of experimental scenarios so that it can cover the breadth of phenomena thought to defy classical understanding. Bellâs notion of local causality fulfils the first criterion but not the second. The notion of noncontextuality fulfils the second criterion, but it is a long-standing question whether it can be made to fulfil the first. Previous attempts to test noncontextuality have all assumed idealizations that real experiments cannot achieve, namely noiseless measurements and exact operational equivalences. Here we show how to devise tests that are free of these idealizations. We perform a photonic implementation of one such test, ruling out noncontextual models with high confidence
