Devising a more robust automated detection method using audio frequency spectra
The space shuttle, Columbia had a suitcase piece of the SOFI strike the wing of the shuttle causing its demise. Detection methods were evaluated to detect defects in the foam. One of the methods being free field acoustics was explored. Previous works were done in this area, and it showed promise as it was able to detect defects. However all defects or the defects that had significance to the eye was not detected. The method developed before is looked into and was improved upon addressing the problems. Developing a baseline curve and region to compare the data would be two of things needed to be implemented. In essence an automated process (program) was developed to be more sensitive in the detection of flaws in the sample. Also developing stopping conditions is important for the process to properly detect the defects. Due to the improvements, all the defects on the mapping of the sample were located
Transcriptional Regulation of Gene Expression in \u3cem\u3eTetrahymena thermophila\u3c/em\u3e
The only well-characterized study of gene expression in Tetrahymena thermophila (1) demonstrates that the temperature dependent expression of the Ser H3 gene is regulated at the level of mRNA stability. A run-on transcription assay was developed to determine if regulation of RNA stability was a major mechanism regulating gene expression in Tetrahymena or if transcriptional regulation dominates. The relative transcriptional activities of 14 Tetrahymena genes were determined in different physiological/developmental states (growing, starved and conjugating) in which many of the genes showed striking differences in RNA abundance. In every case except Ser H3, changes in transcription accompanied changes in RNA abundance. Thus differential transcription, not differential RNA degradation, is the major mechanism regulating RNA abundance in Tetrahymena
Therapeutic Relationship and Outcome Effectiveness: Implications for Counselor Educators
Counselor and client pairs from a university training clinic were analyzed, and therapeutic relationship was the strongest predictor of counseling outcome effectiveness as it progressed across time. In this quantitative study, therapeutic relationship accounted for 25% of the overall variance in outcome effectiveness. The results of this study support the idea that the therapeutic relationship is positively related to counseling outcomes and can be intentionally improved across time

The Transition of Poised RNA Polymerase II to an Actively Elongating State Is a âComplexâ Affair
The initial discovery of the occupancy of RNA polymerase II at certain genes prior to their transcriptional activation occurred a quarter century ago in Drosophila. The preloading of these poised complexes in this inactive state is now apparent in many different organisms across the evolutionary spectrum and occurs at a broad and diverse set of genes. In this paper, we discuss the genetic and biochemical efforts in S. cerevisiae to describe the conversion of these poised transcription complexes to the active state for productive elongation. The accumulated evidence demonstrates that a multitude of coactivators and chromatin remodeling complexes are essential for this transition
A Qualitative Study of Supervisors' Reflections on Providing Sanctioned Supervision
Sanctioned supervision, sometimes referred to as mandated supervision or professional monitoring, is intended to protect the public, reduce further counselor ethical/legal violations, and improve the professional practice of the counselor adjudicated for unprofessional behavior. Sanctioned supervision is a common remediation intervention required by state regulatory board. However, there is a lack of research on the practice of sanctioned supervision and the perceptions of the sanctioned supervisors. A qualitative research approach was used to better understand the experiences of four supervisors who provided sanctioned supervision within the past year as part of a state regulatory board remediation process. The main themes from the qualitative study included the following: supervisors finding the supervision process to be unique from traditional supervision, and supervisors experience ambivalence about the sanctioned supervision process. Practice considerations for supervisors producing sanctioned supervision are discussed
The Removal of the Multiaxial System in the DSM-5: Implications and Practice Suggestions for Counselors
With the advent of the DSM-5 in 2013, the American Psychiatric Association eliminated the longstanding multiaxial system for mental disorders. The removal of the multiaxial system has implications for counselorsâ diagnostic practices. In this article, the removal of the multiaxial system in the DSM-5 is discussed, and counselor practice suggestions related to each of the five Axes are provided. Additionally, ways in which counselors can sustain their current diagnostic skills while developing updated practices that align with the new streamlined system will be discussed
Partnerships to Address School Safety through a Student Support Lens
School safety is a primary concern of school leaders, employees, parents, and a variety of community stakeholders. Attempts to mitigate and prevent school safety concerns often focus on strategies around school climate assessment, emergency communication, school safety plan development, and school resource officer employment (U.S. DHS et al., 2018). Involvement of key stakeholders, such as school social workers, school counselors, and school-based mental health professionals is emphasized in creating and assessing school safety in a wholistic manner. This article provides an overview of a Trainings to Increase School Safety grant program that was implemented with public school stakeholders through partnerships between a university and five public school districts in the Southeastern North Carolina region

This presentation is designed to provide an overview of the The UNCP School Safety Training Program. The UNCP School Safety Training Program, developed by UNCP Social Work and Counseling faculty and funded through the NC DPI School Safety Grant, provides a variety of trainings related to addressing school safety from a student support standpoint. Data from training participants' workshop evaluations will also be highlighted
Nonradioactive, ultrasensitive site-specific proteinâprotein photocrosslinking: interactions of Îą-helix 2 of TATA-binding protein with general transcription factor TFIIA and transcriptional repressor NC2
We have developed an approach that enables nonradioactive, ultrasensitive (attamole sensitivity) site-specific proteinâprotein photocrosslinking, and we have applied the approach to the analysis of interactions of Îą-helix 2 (H2) of human TATA-element binding protein (TBP) with general transcription factor TFIIA and transcriptional repressor NC2. We have found that TBP H2 can be crosslinked to TFIIA in the TFIIAâTBPâDNA complex and in higher order transcriptionâinitiation complexes, and we have mapped the crosslink to the âconnectorâ region of the TFIIA Îą/β subunit (TFIIAÎą/β). We further have found that TBP H2 can be crosslinked to NC2 in the NC2âTBPâDNA complex, and we have mapped the crosslink to the C-terminal âtailâ of the NC2 Îą-subunit (NC2Îą). Interactions of TBP H2 with the TFIIAÎą/β connector and the NC2Îą C-terminal tail were not observed in crystal structures of TFIIAâTBPâDNA and NC2âTBPâDNA complexes, since relevant segments of TFIIA and NC2 were not present in truncated TFIIA and NC2 derivatives used for crystallization. We propose that interactions of TBP H2 with the TFIIAÎą/β connector and the NC2Îą C-terminal tail provide an explanation for genetic results suggesting importance of TBP H2 in TBPâTFIIA interactions and TBPâNC2 interactions, and provide an explanationâsteric exclusionâfor competition between TFIIA and NC2
