    Chemotherapy-induced muscle wasting: Association with NF-κB and cancer cachexia

    A compounding feature of greater than 50% of all cancers is the high incidence of the cachexia syndrome, a complex metabolic disorder characterized by extreme weight loss due mainly to the gross depletion of skeletal muscle tissue. Although studies into the cause of cancer cachexia has spanned over multiple decades, little is known about the effects of various cancer treatments themselves on cachexia. For example, chemotherapy agents induce side effects such as nausea and anorexia, but these symptoms do not fully account for the changes seen with cancer cachexia. In this study we examine the effects of chemotherapeutic compounds, specifically, cisplatin in the colon-26 adenocarcinoma model of cancer cachexia. We find that although cisplatin is able to reduce tumor burden as expected, muscle wasting in mice nevertheless persists. Strikingly, cisplatin alone was seen to regulate muscle atrophy, which was independent of the commonly implicated ubiquitin proteasome system. Finally, we show that cisplatin is able to induce NF-κB activity in both mouse muscles and myotube cultures, suggesting that an additional side effect of cancer treatment is the regulation of muscle wasting that may be mediated through activation of the NF-κB signaling pathway

    The effects of grain shape and frustration in a granular column near jamming

    We investigate the full phase diagram of a column of grains near jamming, as a function of varying levels of frustration. Frustration is modelled by the effect of two opposing fields on a grain, due respectively to grains above and below it. The resulting four dynamical regimes (ballistic, logarithmic, activated and glassy) are characterised by means of the jamming time of zero-temperature dynamics, and of the statistics of attractors reached by the latter. Shape effects are most pronounced in the cases of strong and weak frustration, and essentially disappear around a mean-field point.Comment: 17 pages, 19 figure

    Daldinia eschscholzii (Ascomycota, Xylariaceae) isolado na Amazônia brasileira: características taxonômicas e condições de crescimento micelial

    The Amazon has a high diversity of fungi, including species of the genus Daldinia (Ascomycota, Xylariaceae), which produce secondary metabolites with recognized nematicidal and antimicrobial activity. The ecological role of Daldinia is important, as stromata serve as refuges to many insects and arthropodes, and the fungi contribute to the degradation of vegetable organic matter. The aim of this study was to analyze the taxonomic features and mycelial growth conditions in vitro of a Daldinia specimen collected in the Brazilian Amazon. Morphological and molecular studies of the fungus identified it as D. eschscholtzii. To evaluate mycelial growth, we cultivated the fungus at 20, 25, 30, 35, and 40 °C in malt extract-peptone agar (MEPA), malt extract-peptone (MEP), potato dextrose (PD), and minimum medium (MM). The best mycelial growth occurred at 35 °C, although the greatest amount of biomass was obtained at 25 °C and 30 °C. PD proved to be the best medium for biomass production.A Amazônia apresenta alta diversidade de fungos, incluindo Daldinia (Ascomycota, Xylariaceae), cujas espécies produzem metabólitos secundários com reconhecida atividade antimicrobiana e nematicida. O papel ecológico é importante, visto que estromas servem de abrigo para muitos insetos e artrópodes, além de contribuir na degradação da matéria orgânica vegetal. O objetivo desse estudo foi analizar as características taxonômicas e as condições do crescimento micelial in vitro de um espécime de Daldinia coletado na Amazônia brasileira. Estudos morfológicos e moleculares do fungo o indetificaram como D. eschscholtzii. Para avaliação do crescimento micelial o fungo foi cultivado nas temperaturas de 20, 25, 30, 35 e 40 °C e nos meios de cultura extrato de malte-peptona ágar (EMPA), extrato de malte-peptona (EMP), batata dextrose (BD) e meio mínimo (MM). O melhor crescimento micelial ocorreu a 35 °C, entretanto, a maior quantidade de biomassa foi obtida a 25 e 30 °C. O meio BD provou ser o melhor meio para produção de biomassa

    Spread of a SARS-CoV-2 variant through Europe in the summer of 2020.

    Following its emergence in late 2019, the spread of SARS-CoV-21,2 has been tracked by phylogenetic analysis of viral genome sequences in unprecedented detail3–5. Although the virus spread globally in early 2020 before borders closed, intercontinental travel has since been greatly reduced. However, travel within Europe resumed in the summer of 2020. Here we report on a SARS-CoV-2 variant, 20E (EU1), that was identified in Spain in early summer 2020 and subsequently spread across Europe. We find no evidence that this variant has increased transmissibility, but instead demonstrate how rising incidence in Spain, resumption of travel, and lack of effective screening and containment may explain the variant’s success. Despite travel restrictions, we estimate that 20E (EU1) was introduced hundreds of times to European countries by summertime travellers, which is likely to have undermined local efforts to minimize infection with SARS-CoV-2. Our results illustrate how a variant can rapidly become dominant even in the absence of a substantial transmission advantage in favourable epidemiological settings. Genomic surveillance is critical for understanding how travel can affect transmission of SARS-CoV-2, and thus for informing future containment strategies as travel resumes. © 2021, The Author(s), under exclusive licence to Springer Nature Limited

    Molecular Dynamics Simulations of Laser Induced Ablation for Micro Propulsion

    A new concept of micro propulsion based upon laser ablation MICROLAS was introduced by the Institute of Technical Physics (ITP) of DLR Stuttgart. Pulsed lasers are used for material removal of a target. The amount of removed material should be variable due to the tunability of input laser energy and repetition rate, resulting in well defined impulse bits and low small thrusts down to the sub-µ�N scale. We present a modeling approach of laser ablation in order to calculate important figures of merit in aerospace engineering. The program applied is IMD (http://imd.itap.uni-stuttgart.de), an open source molecular dynamics package of the Institute of Functional Materials Quantum Technologies (FMQ). Results are compared with a hydrodynamic code, VLL (http://vll.ihed.ras.ru), as well as with experimental investigations

    Auditable Privacy: On Tamper-Evident Mix Networks

    We introduce the notion of tamper-evidence for mix networks in order to defend against attacks aimed at covertly leaking secret information held by corrupted mix servers. This is achieved by letting observers (which need not be trusted) verify the absence of covert channels by means of techniques we introduce herein. Our tamper-evident mix network is a type of re-encryption mixnet in which a server proves that the permutation and re-encryption factors that it uses are correctly derived from a random seed to which the server is committed
