    Genetic Fidelity in Micropropagated Plantlets of Pimpinella tirupatiensis-an Endemic and Threatened Medicinal Plant Using RAPD and ISSR Markers

    Tissue cultured Pimpinella tirupatiensis plantlets were subjected to assessment of genetic stability considering the fact that associated in vitro stress might result in breakdown of control mechanism causing instability of the genome. We have used two DNA based molecular markers to assess the genetic fidelity of in vitro regenerated Pimpinella tirupatiensis through shoot tip from in vitro raised seedlings. The shoot tips upon transfer to MS medium containing different concentrations and combinations of 6- benzyl aminopurine (BAP) kinetin (KN), 2- isopentyl adenine (2- ip), α- napthalene acetic acid (NAA) and indole 3- acetic acid (IAA). The best morphogenic response was observed on MS medium fortified with BA (13.31 µM) and NAA (2.69 µM) which exhibited the highest regeneration frequency (90%), the maximum number of shoots/explants (6.50 ± 0.91) and shoot length (3.20 ± 0.20) within 5 weeks. Rooting was achieved within 15 days of shoot implantation on ½ strength MS media fortified with BAP (13.31 µM) and IBA (9.8 µM). The rooted plantlets were successfully acclimatized with 85% survival rate. Out of 20 RAPD and 3 ISSR primers screened, only 6 random amplified polymorphic DNA (RAPD) and all three inter simple sequence repeats (ISSR) primers produced clear reproducible and scorable bands. All banding profiles from micropropagated plants were monomorphic and similar to the mother plant indicating an absence of noticeable genetic variation in the regenerated plantlets. This study is of high significance as these could be commercially utilized for large scale production of true-to-type plantlets in Pimpinella tirupatiensis

    Ultralow-Power and Secure S-Box Circuit Using FinFET Based ECRL Adiabatic Logic

    Advanced Encryption Standard (AES) is the widely used technique in critical cyber security applications. In AES architecture S-box is the most important block. However, the power consumed by      S-box is 75% of the total AES design. The   S-box is also prone to Differential Power Analysis (DPA) attack which is one of the most threatening types of attacks in cryptographic systems. In this paper, a     three-stage positive polarity Reed-Muller (PPRM) S-box is implemented with 45nm FinFET using Efficient Charge Recovery Logic (ECRL) to reduce power consumption. The simulation results indicate up to 66% power savings for FinFET based S-box as compared to CMOS design. Further, the FinFET ECRL 8-bit     S-box circuit is evaluated for transitional energy fluctuations and peak current traces to compare its resistance against side-channel attacks. The lower energy variations and uniform current trace exhibit the improved security performance of the circuit to withstand DPA and Differential Electromagnetic Radiation Attacks (DEMA)

    1206-1210The excess molar volumes (VmE) for binary liquid mixtures of cyclohexane+2-ethoxyethanol have been calculated from the experimental values of density and ultrasonic velocity at temperatures 303.15, 308.15, 313.15 and 318.15 K and atmospheric pressure over the entire composition range. The results have been used to estimate the partial molar volumes m,i of the components. The excess values have been fitted to Redlich-Kister polynomial equation. The changes of VmE and with composition and temperature have been discussed with reference to the nature of interactions between the component molecules

    An audit of the use of hydroxychloroquine in rheumatology clinics

    Objectives To audit the use, indications, complications and patient information regarding hydroxychloroquine (HCQ) treatment in rheumatology clinics in a tertiary referral centre. Methods During a 9-month period, we identified all patients prescribed HCQ and attending rheumatology clinics in one hospital. We established: (1) the indication for HCQ (2) the prevalence of HCQ overdosing based on absolute body weight (ABW) (3) documentation of warning of risk of retinal toxicity (4) systemic and ocular co-morbidities (5) ocular symptoms during treatment (6) reasons for stopping HCQ. Results We identified 427 patients (104 male, 323 female). The cumulative dose of HCQ was lower in rheumatoid arthritis (RA; median 365 g; range 6-1752 g) compared to systemic lupus erythematosus (SLE; 450 g; 66-1788 g) (p = 0.105). The median duration of HCQ therapy was four years (range 0.1-13); 28% of patients with RA and 29% with SLE continued HCQ beyond five years. After adjusting for ABW and renal function, 10% (31/312) had been prescribed doses exceeding recommendations. Formal documentation of counselling on ocular complications was only found in one third of patients. Three cases of HCQ retinopathy were identified (all of whom had RA). Conclusion HCQ therapy is being used for more than five years in 29% of patients with rheumatic diseases, with higher than recommended doses in approximately 10% of patients. We recommend more rigorous scrutiny of the use of HCQ to reduce the risk of retinopathy

    Evaluation of an integrated management package to reduced preharvest seed infection by Aspergillus flavus in groundnut

    Two treatments, an integrated aflatoxin management package (IAMP) and normal farmers' practice were compared in Alfisol fields at ICRISAT, Patancheru, Andhra Pradesh, India during kharif 2001. The IAMP consisted of summer ploughing of the field, seed treatment with carbendazim (Bavistin 50 WP) at 4 g/kg seed, furrow application of Trichoderma harzianum at 50 g culture mixed in 50 kg FYM before sowing (to make a final population of 1 × 106 CFU/g soil), spray of Nimbucidin (250 ml in 50 litres water) and a second spray of carbendazim (50 g) plus Dithane M-45 [mancozeb] (250 g) in 50 litres of water, if required, harvesting at 75% maturity, drying by inverted windrows for 3 days, and removal of damaged and diseased pods. The farmers' practice was summer ploughing, harvesting at full maturity, drying by leaving them in the field and removal of damaged pods. No chemical treatments were involved. For both treatments A. flavus strain Af 11-4 inoculum was applied at 50% flowering stage. Soil was sampled prior to sowing, before inoculum application and at harvest. Colonization of seeds by A. flavus was determined and aflatoxin contamination estimated by ELISA. Soil sample analysis showed unremitting growth of A. flavus in soils where farmers' practice was followed. Seed infection studies revealed predominance of A. flavus infection in plots under farmers' practice (10%) compared with that under IAMP (2%). No aflatoxin contamination was recorded in seed samples from plots subjected to either practice

    A Kallikrein 15 (KLK15) single nucleotide polymorphism located close to a novel exon shows evidence of association with poor ovarian cancer survival.

    BACKGROUND: KLK15 over-expression is reported to be a significant predictor of reduced progression-free survival and overall survival in ovarian cancer. Our aim was to analyse the KLK15 gene for putative functional single nucleotide polymorphisms (SNPs) and assess the association of these and KLK15 HapMap tag SNPs with ovarian cancer survival. RESULTS: In silico analysis was performed to identify KLK15 regulatory elements and to classify potentially functional SNPs in these regions. After SNP validation and identification by DNA sequencing of ovarian cancer cell lines and aggressive ovarian cancer patients, 9 SNPs were shortlisted and genotyped using the Sequenom iPLEX Mass Array platform in a cohort of Australian ovarian cancer patients (N = 319). In the Australian dataset we observed significantly worse survival for the KLK15 rs266851 SNP in a dominant model (Hazard Ratio (HR) 1.42, 95% CI 1.02-1.96). This association was observed in the same direction in two independent datasets, with a combined HR for the three studies of 1.16 (1.00-1.34). This SNP lies 15 bp downstream of a novel exon and is predicted to be involved in mRNA splicing. The mutant allele is also predicted to abrogate an HSF-2 binding site. CONCLUSIONS: We provide evidence of association for the SNP rs266851 with ovarian cancer survival. Our results provide the impetus for downstream functional assays and additional independent validation studies to assess the role of KLK15 regulatory SNPs and KLK15 isoforms with alternative intracellular functional roles in ovarian cancer survival.RIGHTS : This article is licensed under the BioMed Central licence at http://www.biomedcentral.com/about/license which is similar to the 'Creative Commons Attribution Licence'.     Association Analysis of a Microsatellite Repeat in the TRIB1 Gene With Prostate Cancer Risk, Aggressiveness and Survival

    With an estimated 1.1 million men worldwide diagnosed with prostate cancer yearly, effective and more specific biomarkers for early diagnosis could lead to better patient outcome. As such, novel genetic markers are sought for this purpose. The tribbles homologue 1 gene (TRIB1) has recently shown to have a role in prostate tumorigenesis and data-mining of prostate cancer expression data confirmed clinical significance of TRIB1 in prostate cancer. For the first time, a polymorphic microsatellite in this gene was studied for its potential association with prostate cancer risk and aggressiveness. Genomic DNA was extracted from a cohort of 1,152 prostate cancer patients and 1,196 cancer-free controls and the TTTTG-TRIB1 microsatellite was genotyped. The socio-demographic and clinical characteristics were analyzed using the non-parametric t-test and two-way ANOVA. Association of the TTTTG-TRIB1 microsatellite and prostate cancer risk and aggressiveness were analyzed by binary logistic regression and confirmed by bootstrapping. Total and prostate cancer mortality was analyzed using the Kaplan Meier test. Genotype and allele correlation with TRIB1 mRNA levels was analyzed using the non-parametric Kolmogorov–Smirnov test. To predict the effect that the TTTTG-TRIB1 polymorphisms had on the mRNA structure, the in silico RNA folding predictor tool, mfold, was used. By analyzing the publicly available data, we confirmed a significant over-expression of TRIB1 in prostate cancer compared to other cancer types, and an over-expression in prostate cancerous tissue compared to adjacent benign. Three alleles (three–five repeats) were observed for TTTTG-TRIB1. The three-repeat allele was associated with prostate cancer risk at the allele (OR = 1.16; P = 0.044) and genotypic levels (OR = 1.70; P = 0.006) and this association was age-independent. The four-repeat allele was inversely associated with prosatet cancer risk (OR = 0.57; P < 0.0001). TRIB1 expression was upregulated in tumors when compared to adjacent cancer-free tissue but was not allele specific. In silico analysis suggested that the TTTTG-TRIB1 alleles may alter TRIB1 mRNA structure. In summary, the three-repeat allele was significantly associated with prostate cancer risk, suggesting a biomarker potential for this microsatellite to predict prostate cancer. Further studies are needed to elucidate the functional role of this microsatellite in regulating TRIB1 expression, perhaps by affecting the TRIB1 mRNA structure and stability
