    Comprehensive analysis of the membrane phosphoproteome regulated by oligogalacturonides in Arabidopsis thaliana

    Early changes in the Arabidopsis thaliana membrane phosphoproteome in response to oligogalacturonides (OGs), a class of plant damage-associated molecular patterns (DAMPs), were analyzed by two complementary proteomic approaches. Differentially phosphorylated sites were determined through phosphopeptide enrichment followed by LC-MS/MS using label-free quantification; differentially phosphorylated proteins were identified by 2D-DIGE combined with phospho-specific fluorescent staining (phospho-DIGE). This large-scale phosphoproteome analysis of early OG-signaling enabled us to determine 100 regulated phosphosites using LC-MS/MS and 46 differential spots corresponding to 34 pdhosphoproteins using phospho-DIGE. Functional classification showed that the OG-responsive phosphoproteins include kinases, phosphatases and receptor-like kinases, heat shock proteins (HSPs), reactive oxygen species (ROS) scavenging enzymes, proteins related to cellular trafficking, transport, defense and signaling as well as novel candidates for a role in immunity, for which elicitor-induced phosphorylation changes have not been shown before. A comparison with previously identified elicitor-regulated phosphosites shows only a very limited overlap, uncovering the immune-related regulation of 70 phosphorylation sites and revealing novel potential players in the regulation of elicitor-dependent immunity

    Changes in the microsomal proteome of tomato fruit during ripening

    The variations in the membrane proteome of tomato fruit pericarp during ripening have been investigated by mass spectrometry-based label-free proteomics. Mature green (MG30) and red ripe (R45) stages were chosen because they are pivotal in the ripening process: MG30 corresponds to the end of cellular expansion, when fruit growth has stopped and fruit starts ripening, whereas R45 corresponds to the mature fruit. Protein patterns were markedly different: among the 1315 proteins identified with at least two unique peptides, 145 significantly varied in abundance in the process of fruit ripening. The subcellular and biochemical fractionation resulted in GO term enrichment for organelle proteins in our dataset, and allowed the detection of low-abundance proteins that were not detected in previous proteomic studies on tomato fruits. Functional annotation showed that the largest proportion of identified proteins were involved in cell wall metabolism, vesicle-mediated transport, hormone biosynthesis, secondary metabolism, lipid metabolism, protein synthesis and degradation, carbohydrate metabolic processes, signalling and response to stress

    The ultraviolet habitable zone of exoplanets

    The dozens of rocky exoplanets discovered in the Circumstellar Habitable Zone (CHZ) currently represent the most suitable places to host life as we know it outside the Solar System. However, the presumed presence of liquid water on the CHZ planets does not guarantee suitable environments for the emergence of life. According to experimental studies, the building blocks of life are most likely produced photochemically in presence of a minimum ultraviolet (UV) flux. On the other hand, high UV flux can be life-threatening, leading to atmospheric erosion and damaging biomolecules essential to life. These arguments raise questions about the actual habitability of CHZ planets around stars other than Solar-type ones, with different UV to bolometric luminosity ratios. By combining the "principle of mediocricy" and recent experimental studies, we define UV boundary conditions (UV-habitable Zone, UHZ) within which life can possibly emerge and evolve. We investigate whether exoplanets discovered in CHZs do indeed experience such conditions. By analysing Swift-UV/Optical Telescope data, we measure the near ultraviolet (NUV) luminosities of 17 stars harbouring 23 planets in their CHZ. We derive an empirical relation between NUV luminosity and stellar effective temperature. We find that eighteen of the CHZ exoplanets actually orbit outside the UHZ, i.e., the NUV luminosity of their M-dwarf hosts is decisively too low to trigger abiogenesis - through cyanosulfidic chemistry - on them. Only stars with effective temperature >3900 K illuminate their CHZ planets with enough NUV radiation to trigger abiogenesis.     Harvesting techniques for non-industrial SRF biomass plantations on farmland

    <p>The goal of this study was to compare the technical and economic performance of terrain chipping and roadside chipping, applied to short rotation biomass plantations. The null hypothesis was that no significant difference are in the performance of the two work systems, when applied to short rotation coppices. Those systems especially designed for non-industrial SRF plantations, were used for conventional logging operations. The difference on the above mentioned systems consisted especially in the chipping location: chipping was performed directly to the field (containers reach the chipper in the field) or at the field’s edge (roadside chipping). Both systems were tested on two of the most common SRF poplar clones in Italy, namely: AF2 and Monviso. Plots were allocated randomly to the two treatment levels ( roadside or field chipping) than blocked for two main clone types ( AF2 and Monviso ) so that each of the 4 treatments level and clone types has a minimum repetition plot of 6 times (total of 24 replications). The Plot were identified with paint markings at the stump so each plot area could be identified at the ground. Net weight of each charge was obtained by a certified weighbridge, so each plot has its own productivity in terms of weight and time consumption. Results were encouraging: harvesting cost varied from 16.3 to 23.2 tonne-1, and was lower for terrain chipping and for the most productive clone (Monviso). Despite its higher cost, roadside chipping was preferred for its better terrain capability and for the superior storage quality of uncomminuted biomass. Both systems were suboptimal in their current configurations. They could offer a better performance, subject to minor improvements.</p

    The cardiovascular risk of young women with polycystic ovary syndrome: an observational, analytical, prospective case-control study

    To evaluate the cardiovascular risk of polycystic ovary syndrome (PCOS), we investigated lipid profile, metabolic pattern, and echocardiography in 30 young women with PCOS and 30 healthy age- and body mass index (BMI)-matched women. PCOS women had higher fasting glucose and insulin levels, homeostasis model assessment score of insulin sensitivity, total cholesterol (TC) and low density lipoprotein cholesterol (LDL-C) levels, and TC/high density lipoprotein cholesterol (HDL-C) ratio and lower HDL-C levels than controls. Additionally, PCOS women had higher left atrium size (32.0 +/- 4.9 vs. 27.4 +/- 2.1 mm; P &lt; 0.0001) and left ventricular mass index (80.5 +/- 18.1 vs. 56.1 +/- 5.4 g/m(2); P &lt; 0.0001) and lower left ventricular ejection fraction (64.4 +/- 4.1 vs. 67.1 +/- 2.6%; P = 0.003) and early to late mitral flow velocity ratio (1.6 +/- 0.4 vs. 2.1 +/- 0.2; P &lt; 0.0001) than controls. When patients and controls were grouped according to BMI [normal weight (BMI, &gt;18 and &lt;25 kg/m(2)), overweight (BMI, 25.1-30 kg/m(2)), and obese (BMI, &gt;30 kg/m(2))], the differences between PCOS women and controls were maintained in overweight and obese women. In normal weight PCOS women, a significant increase in left ventricular mass index and a decrease in diastolic filling were observed, notwithstanding no change in TC, LDL-C, HDL-C, TC/HDL-C ratio, and TG compared with controls. In conclusion, our data show the detrimental effect of PCOS on the cardiovascular system even in young women asymptomatic for cardiac disease

    Superior Vena Cava Syndrome in a Patient with Polycytemia Vera: Diagnosis and Treatment

    Polycythemia vera is a myeloproliferative disorder characterized by thrombotic complications both in the arterial and venous systems. We report the case of a 55-year-old patient affected by polycythemia vera, presenting with acute superior vena cava syndrome due to thrombosis of the upper part of the superior vena cava. Diagnosis was done clinically and by computed tomography scan and showed an unusual finding: an air bubble trapped in the brachiocephalic venous trunk. The patient underwent emergency surgery.     Top-down cracking in Italian motorway pavements: A case study

    Abstract Top-down cracking (TDC) is a distress affecting asphalt pavements and consists of longitudinal cracks that initiate on the pavement surface and propagate downwards. In general, TDC is more critical in the case of thick pavements with open-graded friction course (OGFC), which are the typical characteristics of Italian motorway pavements. Recent surveys showed the presence of many longitudinal cracks potentially ascribable to TDC on Italian motorways. Within this context, this study has two main objectives: 1) to define reliable identification criteria allowing to distinguish between TDC and the other types of longitudinal cracks observed and 2) based on the developed criteria, to quantify TDC in Italian motorway pavements. In this regard, a 200 km long trial network (400 km considering both directions) was studied, taking into account the effect of several variables (e.g. geometric characteristics, traffic level, wearing layer type and climate). For this purpose, images of the trial network acquired during pavement monitoring were visually analysed and some control cores were taken. Specific criteria (which can be used in a pavement management system, PMS) were developed to distinguish between the main types of longitudinal cracks observed on the trial network, i.e. TDC, cracks due to heavy vehicles tire blowout and construction joints, based on their geometric features on the pavement surface. It was found that TDC can affect up to 20–30 % of the slow traffic lane. Specifically, the highest TDC concentrations were observed for high traffic levels and OGFC, whereas TDC was absent in the case of a dense-graded wearing layer. Finally, surprisingly the concentration of tire blowout cracks was even higher than TDC. This study provides evidence on the fact that, for thick pavements with OGFC, TDC has to be considered a priority problem to be addressed in both pavement design and maintenance
