    Evaluation and registration of plant protection inputs for organic farming

    Organic farmers may only use those plant protection products which are listed in Annex II B of the ‘Organic Regulation’ 2092/91 and also registered for use at national level. At present, new compounds may only be listed in Reg. 2092/91, if they do not come into direct contact with the crop (although in perennial crops, contact outside the growing season of the edible parts is possible), which is a major obstacle for progress. The EU-funded ORGANIC INPUTS EVALUATION project aims at making Reg. 2092/91 more flexible with regard to new products, while safeguarding the principles of organic farming. Registra-tion under the ‘Pesticide Directive’ 91/414 represents a considerable financial hurdle. The REBECA project will propose registration requirements which are better adapted for biological control agents, plant extracts and other low-risk products. The aim is to accelerate registration and make it more cost-effective, without compromises to the level of safety

    An identifying signature is proposed for the existence and location of the neutral line in the magnetotail. The signature, abrupt density, and temperature changes in the Earthtail direction, was first discovered in test particle simulations. Such temperature variations have been observed in ISEE data (Huang et. al. 1992), but their connection to the possible existence of a neutral line in the tail has not yet been established. The proposed signature develops earlier than the ion velocity space ridge of Martin and Speiser (1988), but can only be seen by spacecraft in the vicinity of the neutral line, while the latter can locate a neutral line remotely

    Translocation of phosphonate from frigoplants to fruits in strawberries

    We analysed the phosphonate contents in commercially available strawberry frigoplants and in the fruits grown on these plants. The results show that phosphonates can be translocated from frigoplants to fruits. Thus, strawberry growers may be confronted with phosphonate residues even if they have not applied phosphonates themselves

    Phenotypic and functional cell properties are usually analyzed at the level of defined cell populations but not single cells. Yet, large differences between individual cells may have important functional consequences. It is likely that T-cell-mediated immunity depends on the polyfunctionality of individual T cells, rather than the sum of functions of responding T-cell subpopulations. We performed highly sensitive single-cell gene expression profiling, allowing the direct ex vivo characterization of individual virus-specific and tumor-specific T cells from healthy donors and melanoma patients. We have previously shown that vaccination with the natural tumor peptide Melan-A-induced T cells with superior effector functions as compared with vaccination with the analog peptide optimized for enhanced HLA-A*0201 binding. Here we found that natural peptide vaccination induced tumor-reactive CD8 T cells with frequent coexpression of both memory/homing-associated genes (CD27, IL7R, EOMES, CXCR3, and CCR5) and effector-related genes (IFNG, KLRD1, PRF1, and GZMB), comparable with protective Epstein-Barr virus-specific and cytomegalovirus-specific T cells. In contrast, memory/homing-associated and effector-associated genes were less frequently coexpressed after vaccination with the analog peptide. Remarkably, these findings reveal a previously unknown level of gene expression diversity among vaccine-specific and virus-specific T cells with the simultaneous coexpression of multiple memory/homing-related and effector-related genes by the same cell. Such broad functional gene expression signatures within antigen-specific T cells may be critical for mounting efficient responses to pathogens or tumors. In summary, direct ex vivo high-resolution molecular characterization of individual T cells provides key insights into the processes shaping the functional properties of tumor-specific and virus-specific T cells

    Immune protection from infectious diseases and cancer is mediated by individual T cells of different clonal origin. Their functions are tightly regulated but not yet fully characterized. Understanding the contribution of each T cell will improve the prediction of immune protection based on laboratory assessment of T-cell responses. Here we developed techniques for simultaneous molecular and functional assessment of single CD8 T cells directly ex vivo. We studied two groups of patients with melanoma after vaccination with two closely related tumor antigenic peptides. Vaccination induced T cells with strong memory and effector functions, as found in virtually all T cells of the first patient group, and fractions of T cells in the second group. Interestingly, high functionality was not restricted to dominant clonotypes. Rather, dominant and nondominant clonotypes acquired equal functional competence. In parallel, this was also found for EBV- and CMV-specific T cells. Thus, the nondominant clonotypes may contribute similarly to immunity as their dominant counterparts

    Evaluating inputs for organic farming – a new system. Proposals of the ORGANIC INPUTS EVALUATION project

    This volume contains proposals for criteria for evaluation of plant protection products, fertilisers and soil conditioners1 to be used in organic agriculture. These ideas were developed in the course of the European Union (EU) Concerted Action project ‘ORGANIC INPUTS EVALUATION’ (QLK5-CT-2002-02565). For more information on this project see the end of this volume or visit the project website www.organicinputs.org. The documents in this volume are proposals elaborated by the project consortium and external experts. They were discussed with a broader audience at a public conference held in Brussels on October 13, 2005, and have been amended accordingly. Our proposals also include a “criteria matrix”, which is in Microsoft Excel format, and therefore stands as a separate file. The criteria matrix is discussed in section 5, but we strongly recommend that you consult the original document. To illustrate the use of the matrix, we have further prepared two case studies, which are also separate Excel files. All of these files are contained on the CD, and can also be downloaded from the project website. Currently, Regulation 2092/91 is under revision. We hope that our ideas can be incorporated into the regulation during this revision! In addition, we strongly encourage national institutions to make use of our proposals at the national level

    Facilitating the registration of biocontrol organisms, plant extracts and semiochemicals in Europe

    The legal regulation of plant protection products (Dir. 91/414/ EEC) is a bottleneck in the market introduction of new microbial biocontrol agents, plant extracts and pheromones. In contrast, invertebrate biocontrol agents (“beneficials”) are not registered at EU level. The EU-funded project REBECA suggested improvements to accelerate the regulation process and make it more cost-effective, without compromises to the level of safety. Representatives of all stakeholder groups participated in the REBECA workshops. The EU Commission and Member States are encouraged to improve the registration of biocontrol organisms, plant extracts and semiochemicals at EU and/or national level.     Congenital adrenal hyperplasia

    Congenital adrenal hyperplasia consists of a heterogenous group of inherited disorders due to enzymatic defects in the biosynthetic pathway of cortisol and/or aldosterone. This results in glucocorticoid deficiency, mineralocorticoid deficiency, and androgen excess. 95% of CAH cases are due to 21-hydroxylase deficiency. Clinical forms range from the severe, classical CAH associated with complete loss of enzyme function, to milder, non-classical forms (NCAH). Androgen excess affects the pilosebaceous unit, causing cutaneous manifestations such as acne, androgenetic alopecia and hirsutism. Clinical differential diagnosis between NCAH and polycystic ovary syndrome may be difficult. In this review, the evaluation of patients with suspected CAH, the clinical presentation of CAH forms, with emphasis on the cutaneous manifestations of the disease, and available treatment options, will be discussed
