    A topological characterization of modulo-p arguments and implications for necklace splitting

    The classes PPA-p have attracted attention lately, because they are the main candidates for capturing the complexity of Necklace Splitting with p thieves, for prime p. However, these classes were not known to have complete problems of a topological nature, which impedes any progress towards settling the complexity of the Necklace Splitting problem. On the contrary, topological problems have been pivotal in obtaining completeness results for PPAD and PPA, such as the PPAD-completeness of finding a Nash equilibrium [18, 15] and the PPA-completeness of Necklace Splitting with 2 thieves [24]. In this paper, we provide the first topological characterization of the classes PPA-p. First, we show that the computational problem associated with a simple generalization of Tucker's Lemma, termed p-polygon-Tucker, as well as the associated Borsuk-Ulam-type theorem, p-polygon-Borsuk-Ulam, are PPA-p-complete. Then, we show that the computational version of the well-known BSS Theorem [8], as well as the associated BSS-Tucker problem are PPA-p-complete. Finally, using a different generalization of Tucker's Lemma (termed Zp-star-Tucker), which we prove to be PPA-p-complete, we prove that p-thief Necklace Splitting is in PPA-p. This latter result gives a new combinatorial proof for the Necklace Splitting theorem, the only proof of this nature other than that of Meunier [42]. All of our containment results are obtained through a new combinatorial proof for Zp-versions of Tucker's lemma that is a natural generalization of the standard combinatorial proof of Tucker's lemma by Freund and Todd [27]. We believe that this new proof technique is of independent interest

    Determination of the effect of collars containing 10% w/w imidacloprid and 4.5% w/w flumethrin (Seresto®) on the incidence of Leishmania and other canine vector-borne pathogen infections in Greece

    Background: The objective of this field study was to assess the effect of treating a considerable portion of a dog population naturally exposed to canine vector-borne pathogens (CVBPs) in endemic areas with a 10% w/w imidacloprid/4.5% w/w flumethrin collar (Seresto®) on the transmission of CVBPs and the resulting incidence of infection. Methods: A total of 479 dogs from two sites were enrolled in the study. Collars were placed on all dogs continuously for 21 months, with replacement of the collar every 7 months. All dogs were examined, including body weight and blood/conjunctival swab collections, every 7 months. Serum samples were analysed for the presence of antibodies against Leishmania infantum, Ehrlichia canis and Anaplasma phagocytophilum. PCR assays were also performed on blood samples and conjunctival swab collected from the dogs for the presence of L. infantum, and on blood samples only for the presence of Ehrlichia spp. and Anaplasma spp. Sand flies were collected, identified to species level and molecularly tested for L. infantum throughout two vector activity seasons. Results: The results showed that the Seresto collar was safe with continuous use. At study inclusion, 419, 370 and 453 dogs tested negative for L. infantum, Ehrlichia spp. and Anaplasma spp., respectively (353 dogs tested negative for any pathogen). Overall, 90.2% of the dogs were protected from L. infantum infection on both sites combined. The entomological survey confirmed the presence of competent vectors of L. infantum at all monitored locations, namely the sand flies Phlebotomus neglectus and Phlebotomus tobbi, both of which are regarded as the most important competent vectors in the Mediterranean basin. All captured sand flies tested negative for L. infantum. Protection against ticks and fleas was high, with only two dogs showing a low number of ticks and seven dogs having low numbers of fleas at single evaluation time points. Across the entire study population, a number of dogs became infected with tick-transmitted pathogens, but prevention of transmission was 93% for E. canis and 87.2% for Anaplasma spp. when all cases from both sites were combined. Conclusions: The Seresto® (10% w/w imidacloprid/4.5% w/w flumethrin) collar significantly reduced the risk of CVBP transmission when compared to previously observed incidences of CVBP infections in two highly endemic areas under field conditions

    Morphology of the first instar larva of obligatory traumatic myiasis agents (Diptera: Calliphoridae, Sarcophagidae)

    There are only three fly species that are obligate agents of traumatic myiasis of humans and livestock: a single species of flesh fly, Wohlfahrtia magnifica (Sarcophagidae), and two species of blow flies, Chrysomya bezziana and Cochliomyia hominivorax (Calliphoridae). The morphology of their first instar larvae is thoroughly and consistently documented here with light microscopy photographs and scanning electron microscopy micrographs. The following morphological structures are documented: pseudocephalon, antennal complex, maxillary palpus, oral ridges, thoracic and abdominal spinulation, spiracular field, posterior spiracles and cephaloskeleton. New diagnostic features drawn from the cephaloskeleton and the spinulation of abdominal segments, including the anal pad, are discovered and extensively described. Earlier descriptions in the literature are revisited, and major discrepancies between these and the results of the current study are discussed. The present results allow clarification, correction and, especially, complementation of information provided by earlier authors. The relatively distant taxonomic position of all three species is evidence that obligatory myiasis has arisen independently, and the extensively similar morphology in the first instar larvae of Chrysomya bezziana, Cochliomyia hominivorax and W. magnifica in comparison to necrophagous species, especially the enhancement of the anterior part of the cephaloskeleton and the segmental spinulation, is therefore best interpreted as homoplasic adaptations to a life strategy as obligate vertebrate parasites.     Genotyping of European Toxoplasma gondii strains by a new high-resolution next-generation sequencing-based method

    Purpose: A new high-resolution next-generation sequencing (NGS)-based method was established to type closely related European type II Toxoplasma gondii strains. Methods: T. gondii field isolates were collected from different parts of Europe and assessed by whole genome sequencing (WGS). In comparison to ME49 (a type II reference strain), highly polymorphic regions (HPRs) were identified, showing a considerable number of single nucleotide polymorphisms (SNPs). After confirmation by Sanger sequencing, 18 HPRs were used to design a primer panel for multiplex PCR to establish a multilocus Ion AmpliSeq typing method. Toxoplasma gondii isolates and T. gondii present in clinical samples were typed with the new method. The sensitivity of the method was tested with serially diluted reference DNA samples. Results: Among type II specimens, the method could differentiate the same number of haplotypes as the reference standard, microsatellite (MS) typing. Passages of the same isolates and specimens originating from abortion outbreaks were identified as identical. In addition, seven different genotypes, two atypical and two recombinant specimens were clearly distinguished from each other by the method. Furthermore, almost all SNPs detected by the Ion AmpliSeq method corresponded to those expected based on WGS. By testing serially diluted DNA samples, the method exhibited a similar analytical sensitivity as MS typing. Conclusion: The new method can distinguish different T. gondii genotypes and detect intra-genotype variability among European type II T. gondii strains. Furthermore, with WGS data additional target regions can be added to the method to potentially increase typing resolution

    An assessment of snail-farm systems based on land use and farm components

    In this study, the structural and management characteristics of snail farms in Greece were analyzed to maximize sustainable food production. Objectives, such as the classification of farming systems and assessing the effects of various annual production parameters, were investigated. Data were collected (2017) via a questionnaire, and sampling was conducted in 29 snail farms dispersed in six different regions (Thrace, Central Macedonia, West Macedonia, Thessaly, Western Greece, and the Attica Islands). Descriptive statistics for continuous variables and frequencies for categorical variables were calculated. The similarity between farms was analyzed using nonmetric multidimensional scaling (nMDS). The average farm operation duration exceeded eight months and the mean annual production was 1597 kg of fresh, live snails. Results recorded five farming systems: elevated sections (7%), net-covered greenhouse (38%), a mixed system with a net-covered greenhouse (10%), open field (38%), and mixed system with an open field (7%). Snail farms differ in the type of substrate, available facilities, and equipment (60% similarity between most of the open field farms). The geographical location of a farms’ settlement affects productivity but also influences the duration of operation, especially in open field farms, due to their operation under a wide assortment of climatic types. © 2021 by the authors.
