Enlèvement des colloïdes associés aux dioxines et furanes d'un lixiviat de bassin d'entreposage de bois traité
RÉSUMÉ Les poteaux de bois utilisés pour le transport de lignes électriques sont traités par des produits de préservation du bois tels que l’arséniate de cuivre chromaté (ACC) et le pentachlorophénol (PCP). Les impuretés du PCP, les polychlorodibenzo-p-dioxines (PCDD) et polychlorodibenzofuranes (PCDF), aussi appelés dioxines et furanes, sont particulièrement nocifs pour la santé humaine et l’environnement. Ces produits (ACC et PCP) peuvent se solubiliser dans l’eau de ruissellement lorsque des poteaux neufs ou en fin de vie utile sont entreposés en lot à l’extérieur. Cette eau de ruissellement contient une contamination mixte (organique et inorganique) qui est traitée par une technologie de marais filtrants dans le but de répondre aux normes environnementales applicables.----------ABSTRACT Wood poles used for power lines are treated with wood preservatives such as chromated copper arsenate (ACC) and pentachlorophenol (PCP). Treatment of a mixed wood preservative leachate by a hybrid constructed wetland and a willow planted filter
The performance and removal mechanisms of a hybrid constructed wetland (HCW) followed by a willow planted filter (WPF) were evaluated for the treatment of a leachate contaminated by wood pole preservatives (pentachlorophenol (PCP) and chromated copper arsenate) to reach the storm sewer discharge limits. The HCW aimed to dechlorinate the PCP and polychlorodibenzo-p-dioxins/polychlorodibenzofuran (PCDD/F) and to remove metals by adsorption and precipitation. The HCW was efficient in removing PCP (>98.6%), oil, arsenic (99.4%), chromium (>99.2%), copper (>99.6%%) and iron (29%) to under their discharge limits, but it was unable to reach those of Mn and PCDD/F, with
residual concentrations of 0.11 mg Mn/L and 0.32 pg TEQ/L. Iron and manganese could be removed but were subsequently released by the HCW due to low redox conditions. No dechlorination of PCDD/F was observed since its chlorination profile remained the same in the different sections of the HCW. Adsorption was the most probable removal mechanism of PCDD/F. The WPF was able to remove some residual contamination, but it released Mn at a gradually decreasing rate. Total evapotranspiration of the leachate by a larger fertilized WPF and the construction of an underground retention basin are proposed to prevent any discharge of PCDD/F traces in the environment
High-Resolution Melting System to Perform Multilocus Sequence Typing of Campylobacter jejuni
Multi-locus sequence typing (MLST) has emerged as the state-of-the-art method for resolving bacterial population genetics but it is expensive and time consuming. We evaluated the potential of high resolution melting (HRM) to identify known MLST alleles of Campylobacter jejuni at reduced cost and time. Each MLST locus was amplified in two or three sub fragments, which were analyzed by HRM. The approach was investigated using 47 C. jejuni isolates, previously characterized by classical MLST, representing isolates from diverse environmental, animal and clinical sources and including the six most prevalent sequence types (ST) and the most frequent alleles. HRM was then applied to a validation set of 84 additional C. jejuni isolates from chickens; 92% of the alleles were resolved in 35 hours of laboratory time and the cost of reagents per isolate was 100 for sequence-based typing. HRM has the potential to complement sequence-based methods for resolving SNPs and to facilitate a wide range of genotyping studies
Transforming Growth Factor: β Signaling Is Essential for Limb Regeneration in Axolotls
Axolotls (urodele amphibians) have the unique ability, among vertebrates, to perfectly regenerate many parts of their body including limbs, tail, jaw and spinal cord following injury or amputation. The axolotl limb is the most widely used structure as an experimental model to study tissue regeneration. The process is well characterized, requiring multiple cellular and molecular mechanisms. The preparation phase represents the first part of the regeneration process which includes wound healing, cellular migration, dedifferentiation and proliferation. The redevelopment phase represents the second part when dedifferentiated cells stop proliferating and redifferentiate to give rise to all missing structures. In the axolotl, when a limb is amputated, the missing or wounded part is regenerated perfectly without scar formation between the stump and the regenerated structure. Multiple authors have recently highlighted the similarities between the early phases of mammalian wound healing and urodele limb regeneration. In mammals, one very important family of growth factors implicated in the control of almost all aspects of wound healing is the transforming growth factor-beta family (TGF-β). In the present study, the full length sequence of the axolotl TGF-β1 cDNA was isolated. The spatio-temporal expression pattern of TGF-β1 in regenerating limbs shows that this gene is up-regulated during the preparation phase of regeneration. Our results also demonstrate the presence of multiple components of the TGF-β signaling machinery in axolotl cells. By using a specific pharmacological inhibitor of TGF-β type I receptor, SB-431542, we show that TGF-β signaling is required for axolotl limb regeneration. Treatment of regenerating limbs with SB-431542 reveals that cellular proliferation during limb regeneration as well as the expression of genes directly dependent on TGF-β signaling are down-regulated. These data directly implicate TGF-β signaling in the initiation and control of the regeneration process in axolotls
Les indicateurs de motivation qui incitent les enseignants débutants à s'engager dans leur travail et à poursuivre leur carrière dans l'enseignement au secondaire
RÉSUMÉ: L'entrée dans la profession enseignante représente une étape cruciale dans la carrière
de l'enseignant débutant. Cette période d'insertion dans la profession entraîne non
seulement une Ă©nergie positive, mais aussi une panoplie d'Ă©motions, lorsque l'enseignant
novice fait face à la complexité de son nouveau rôle professionnel (Lamarre, 2003). Alors
qu'il s'était imaginé une vision exemplaire de l'enseignement, il se trouve fréquemment
précipité dans le milieu de pratique, embauché à la dernière minute, récoltant des tâches
fragmentées, responsable d'enseigner des matières avec lesquelles il est peu ou pas familier
et parfois assigné aux groupes les plus difficiles (Mukamurera, 1998, 2005; Comité
d'orientation de la formation du personnel enseignant (COFPE), 2002; Fédération des
syndicats de l'enseignement (FSE), 2003). Souvent laissé à lui-même dans cette phase
d'entrée dans la carrière, l'enseignant peut être envahi par un sentiment d'insécurité face
aux tâches complexes à accomplir (COFPE, 2002). Il doit alors faire preuve d'autonomie et
d'une grande capacité d'adaptation pour surmonter sereinement cette période.
Alors que certains poursuivent leur carrière en enseignement et vivent l'étape de
l'entrée dans la profession comme une expérience constructive dans leur développement
professionnel, celle-ci peut s'avérer plus difficile pour d'autres. Cette situation d'instabilité
où le novice doit jongler avec plusieurs facteurs nouveaux et imprévus peut engendrer des
difficultés diverses et même susciter une remise en question quant à son avenir dans la
profession (Mukamurera, 2005). Au Québec, les statistiques montrent qu'un enseignant sur
cinq, soit près de 20 % des enseignants, décroche au cours des cinq premières années dans
la carrière (FSE, 2008). Plusieurs chercheurs associent ce phénomène d'abandon de la
profession aux difficultés vécues par les nouveaux enseignants lors de l'insertion
professionnelle (Veenman, 1984; Huberman, 1989; COFPE, 2002), aux conditions
d'embauche précaires et instables (Mukamurera, 1998, 2005) ou encore aux modalités
d'accompagnement et de soutien peu présentes dans les milieux scolaires (Gervais, 1999;
COFPE, 2002).
Au-delĂ de ces statistiques peu reluisantes, il y a tout de mĂŞme plus de 80 % des
enseignants québécois qui persistent dans la profession, et ce, malgré les péripéties
rencontrées en début de carrière. Depuis plus de quarante ans, les études portant sur
l'insertion professionnelle des enseignants ont davantage abordé les difficultés vécues chez
les novices, au dĂ©triment de dĂ©voiler les Ă©lĂ©ments qui les motivent Ă persĂ©vĂ©rer et Ă
s'engager dans leur travail malgré les péripéties parfois déroutantes (Lamarre, 2003).
Ce bref constat met en évidence la pertinence de nous intéresser, non pas à ceux qui
quittent l'enseignement pour différents motifs, mais plutôt à ceux qui choisissent de
poursuivre leur carrière dans ce domaine. Le fait d'explorer la situation des enseignants lors
de leurs premières années d'expérience a permis de déterminer des indices de motivation
qui incitent les enseignants en début de carrière à s'engager dans leur travail en plus de
cibler les facteurs favorisant la réussite de l'insertion professionnelle ainsi que la rétention
dans la profession. La recension des écrits révèle que peu d'études ont été menées sur
l'engagement et la motivation des enseignants qui amorcent leur carrière en enseignement
au QuĂ©bec (Duchesne, 2004). De plus, ces Ă©tudes portent sur la situation des enseignants Ă
la mi-carrière et non sur celle des enseignants en début de parcours.
Dans une perspective qualitative interprétative, cette recherche vise à cibler les
indicateurs de motivation qui incitent les enseignants novices Ă s'engager dans leur travail
et à persévérer, malgré les débuts périlleux de la carrière (Veenman, 1984; COFPE, 2002 ;
Nault, 1993 ; Boutin, 2002). Il s'agit de découvrir ce qui favorise la motivation (Deci et
Ryan ,1985; Valle rand et coll., 1989,1992; Blais et coll., 1993),l'engagement (Reyes,
1990; Duchesne, 2004; Guillemette, 2006) et la rétention dans la profession enseignante. La
théorie de l' autodétermination de Deci et Ryan (1985) a été retenue pour l'analyse
conceptuelle, car elle pem1et d'explorer le phénomène de la motivation humaine, selon
qu'elle provienne de facteurs relevant de l'intérieur de soi (intrinsèques) ou de l'extérieur
de soi (extrinsèques).
Plus spécifiquement, l'étude porte sur l'expérience vécue de cinq enseignants en
exercice au secondaire, ayant de 5 à 7 années d 'expérience et une tâche de 80 % ou plus. Il
s'agit d'explorer comment ces enseignants ont vécu leur insertion professionnelle afin de
faire Ă©merger les facteurs de motivation qui les incitent Ă s'engager dans leur travail et Ă
poursuivre leur carrière dans l'enseignement. La question qui oriente le processus de
recherche s'énonce de la façon suivante: Quels sont les indicateurs de motivation qui
incitent les enseignants débutants à s'engager dans leur travail et à poursuivre leur carrière
dans l'enseignement au secondaire? Par conséquent, l'objectif de recherche permet de
déterminer les facteurs de motivation qui incitent les enseignants débutants à s'engager
dans leur travail et à poursuivre leur carrière dans l'enseignement au secondaire. Cette
recherche contribue Ă fournir un Ă©clairage nouveau dans le domaine de la motivation et de
l'engagement des enseignants débutants, tout en ciblant les facteurs de réussites favorisant
une insertion professionnelle harmonieuse ainsi que la persistance dans la profession
enseignante. -- ABSTRACT: Starting out in the teaching profession is a critical step in the careers of new teachers.
In Quebec, several statistics show that one in five teachers, nearly 20%, quit in the first five
years in their chosen career (FSE, 2008). Many researchers associate the trend of teachers
abandoning the profession to the difficulties experienced by new teachers during
professional integration (Veenman, 1984; Heberman, 1989; COFPE, 2002), to precarious
and unstable conditions of employment (Mukamurera, 1998, 2005) and to the lack of
support and collaboration in schools (Gervais, 1999; COFPE, 2002).
Beyond these b1eak statistics, 80% of Quebec teachers continue in the profession
despite the vicissitudes encountered early in their careers. For over forty years, studies on
the integration of teachers have focussed primarily on the difficulties experienced by those starting out in the field and less on determining elements that motivate them to persevere
and to invest more in their work despite the ups and downs. (Lamarre, 2003). This brief
analysis shows that it is important that we focus, not on those who leave teaching for
various reasons, but rather on those who choose to pursue their career in this field.
This study discusses the positive aspects of starting a career in teaching. Specifically,
this analysis focuses on factors that motivate and encourage new teachers to engage in their
work and to persevere in the profession, des pite the difficulties experienced early in their
careers. From a practical perspective, five high school teachers, having between 5 and 7
years of experience, have been interviewed to explore their experience in terms of
professional integration and to determine the motivators which have encouraged them to
pursue their careers and become invested in the profession. Deci and Ryan's selfdetermination
theory (1985) was selected for the conceptual analysis because it allows us to
explore the phenomenon of human motivation originating from intrinsic or extrinsic
factors. The question that guides the research process reads as follows: What motivation
encourages new teachers to engage in their work and to pursue careers in teaching at the
high schoollevel? Consequently, the objective of the study is to determine the motivating
factors that encourage new teachers to engage in their work and pursue their careers in
secondary education. This research helps provide new insights in the field of motivation
and commitment of new teachers, while focusing on factors promoting harmonious
professional integration and persistence in the teaching profession
Changing arctic and subarctic marine environments and implications for human health
Arctic and Subarctic marine environments are experiencing rapid and significant changes associated with climate warming. The drastic decrease in sea ice extent and thickness has important ramifications for physical, chemical and biological processes and energy dynamics of food webs. Changes in sea ice and the amount of light entering the upper ocean can significantly affect the growth of autotrophic single-celled algae and therefore the availability of essential biomolecules that are eventually incorporated into new biomass at higher trophic levels. Ice-free warmer waters and consequent changes in ocean currents have also been shown to drive the northward expansion of boreal species and the availability of essential fatty acids through novel interactions occurring in both bottom-up and top-down interactions. These changes in marine food webs and associated contaminant dynamics can lead to cascading effects on northern coastal communities that rely on country foods of high nutritional quality. This chapter gathers a selection of research results from the Sentinel North program that address interdisciplinary and ecosystem-level research questions that pertain to climate-related changes in primary production and food web dynamics, notably the availability and quality of essentials compounds; the trophic flux of energy through the major biota of Arctic and Subarctic marine ecosystems; and the role of marine country foods in contaminants exposure and human health

Les environnements marins arctiques et subarctiques subissent des changements rapides et importants liés au réchauffement climatique. La diminution drastique du couvert et du volume de glace de mer a des répercussions importantes sur les processus physiques, chimiques et biologiques, ainsi que sur la dynamique énergétique des réseaux trophiques. Les changements dans la glace de mer et la quantité de lumière qui pénètre dans la partie supérieure de l'océan peuvent affecter de manière significative la croissance des algues unicellulaires autotrophes et, par conséquent, la disponibilité de biomolécules essentielles qui sont ensuite incorporées à des niveaux trophiques supérieurs. Il a également été démontré que les eaux chaudes libres de glace et les modifications des courants océaniques qui en découlent favorisent l'expansion vers le nord d’espèces boréales et modifient la disponibilité des acides gras essentiels, en raison de nouvelles interactions ascendantes (bottom-up) et descendantes (top-down). Ces changements dans les réseaux trophiques marins et la dynamique des contaminants qui y est associée peuvent avoir des effets en cascade sur les communautés nordiques qui dépendent d'aliments traditionnels issus de la mer. Ce chapitre rassemble une sélection de résultats du programme Sentinelle Nord qui abordent des questions de recherche interdisciplinaire qui concernent les changements liés au climat dans la production primaire et la dynamique des réseaux trophiques, notamment la disponibilité et la qualité de composés essentiels ; le flux d'énergie à travers les principaux organismes des écosystèmes marins arctiques et subarctiques ; et le rôle des aliments traditionnels marins dans l'exposition aux contaminants et la santé humaine
