    Comparison of fluorgestone and medroxyprogesterone intravaginal sponges for oestrus synchronization in Saanen does during the transition period

    The efficiency of medroxyprogesterone acetate (MAP) and fluorogestone acetate (FGA) sponges for synchronizing oestrus in lactating Saanen goats was investigated during the transition from non-breeding to natural breeding season. Does were treated for 11 days with 60 mg MAP (n=19) or 40 mg FGA (n=24) sponges. All does received intramuscular injections of 750 IU pregnant mare serum gonadotrophin (PMSG) and 125 mg cloprostenol (PGF2a) 48 h prior to sponge removal. Cervical artificial insemination (AI) with fresh diluted semen was performed at a fixed time (36 and 48 h) followed progestagen withdrawal. The two progestagen treatments showed no significant difference in oestrous response (100% both for MAP and FGA groups), time to the onset (15.8+09 and 15.0+06 h for the MAP and FGA groups, respectively), duration (30.5+1.9 and 34.0+1.4 h for MAP and FGA, respectively) and cessation (42.32+1.6 and 43.25+1.3 h for MAP and FGA, respectively) of the induced oestrus period. No significant difference was observed with respect to pregnancy rates determined at the 53rd day after AI (52.6 and 50.0% for MAP and FGA, respectively). These dates indicate that the use of MAP and FGA intravaginal progestagen treatments are equally efficient in synchronizing oestrus in lactating goats during the transition from the non-breeding to the natural breeding season.     Allocation in Practice

    How do we allocate scarcere sources? How do we fairly allocate costs? These are two pressing challenges facing society today. I discuss two recent projects at NICTA concerning resource and cost allocation. In the first, we have been working with FoodBank Local, a social startup working in collaboration with food bank charities around the world to optimise the logistics of collecting and distributing donated food. Before we can distribute this food, we must decide how to allocate it to different charities and food kitchens. This gives rise to a fair division problem with several new dimensions, rarely considered in the literature. In the second, we have been looking at cost allocation within the distribution network of a large multinational company.     Hemoglobin is inversely related to flow-mediated dilatation in chronic kidney disease

    The microcirculation is regulated by oxygen gradients and by endothelial release of nitric oxide, which can react with hemoglobin to form S-nitroso derivatives. Here we induced flow-mediated dilatation of the brachial artery in response to ischemia in 141 non-diabetic patients with stage 3–4 chronic kidney disease who had no history of smoking, cardiovascular events or use of erythropoietin-based agents. Patients with hemoglobin concentrations above the cohort median of 11.6 g/dl were found to have significant reductions in flow-mediated dilatation compared to those below the median. This inverse relationship remained significant after adjustment for potential confounders, including insulin sensitivity, glomerular filtration rate, proteinuria, body mass index, serum urate, etiology of underlying renal disease, treatment with anti-hypertensive drugs, and traditional Framingham risk factors. Given that hemoglobin can act as an important nitric oxide carrier and buffer, our studies suggest that the mechanism by which hemoglobin influences the endothelium-dependent microcirculation requires its nitrosylation; however, more direct studies need to be performed

    FGF-23 and vascular dysfunction in patients with stage 3 and 4 chronic kidney disease

    Studies in animals show that fibroblast growth factor (FGF)-23 interferes with vascular reactivity induced by the nitric oxide (NO) system. To investigate the relationship between circulating FGF-23 levels and the response of forearm blood flow to ischemia (flow-mediated vasodilatation, FMD) and nitroglycerin, we tested 183 patients with stage 3-4 chronic kidney disease (CKD). None of them had cardiovascular complications or were taking drugs interfering with vascular function. Patients with FGF-23 levels above the median had significantly lower glomerular filtration rate, FMD, and fetuin-A levels (an anti-inflammatory molecule and potent inhibitor of calcification). They also had higher proteinuria and phosphate levels when compared to patients whose FGF-23 levels were below the median. The response to nitroglycerin was not different between the two groups. Multiple regression analysis showed that the relationship between FGF-23 and FMD was only modestly sensitive to adjustment for classical risk factors, biomarkers of bone mineral metabolism, high-sensitivity C-reactive protein, and homeostatic model assessment index. Adjustment for asymmetrical dimethyl arginine (ADMA) weakened the strength of this link; however, it remained highly significant. There was no independent association between FGF-23 and nitroglycerin. Thus, attenuation of FMD by ADMA suggests that this endogenous inhibitor of NO synthase may, in part, mediate the vascular effects of FGF-23 in patients with CKD. © 2010 International Society of Nephrology

    Partial-thickness macular hole in vitreomacular traction syndrome: a case report and review of the literature

    <p>Abstract</p> <p>Introduction</p> <p>Vitreomacular traction syndrome has recently been recognized as a distinct clinical condition. It may lead to many complications, such as cystoid macular edema, macular pucker formation, tractional macular detachment, and full-thickness macular hole formation.</p> <p>Case presentation</p> <p>We report a case of vitreomacular traction syndrome with eccentric traction at the macula and a partial-thickness macular hole in a 63-year-old Pakistani Punjabi man. The patient was evaluated using optical coherence tomography, and he underwent a successful pars plana vitrectomy. After the operation, his foveal contour regained normal configuration, and his visual acuity improved from 20/60 to 20/30.</p> <p>Conclusions</p> <p>Pars plana vitrectomy prevents the progression of a partial thickness macular hole in vitreomacular traction syndrome. The relief of traction by vitrectomy restores foveal anatomy and visual acuity in this condition.</p

    A Performance Analysis on Pressure Loss and Airflow Diffusion in a Chamber with Perforated V-Profile Diffuser Designed for Air Handling Units (AHUs)

    Outlet cross-sectional area of fans used in air handling units is smaller than cross-sectional area of chambers which are located next to the fan. In order to ensure efficiently operating of the air handling units, it is required that the air flows through a perforated diffuser to create a uniform air diffusion from fan outlet to following chamber with a minimum pressure loss and uniform velocity distribution. In this concept, numerical simulations and experiments were performed for the chamber with perforated V-profile diffuser, which is often used in air handling units because of its simple geometry and easy manufacturing. Pressure losses were firstly obtained experimentally for different air velocities in the chamber. Then a performance analysis on the air flow diffusion and pressure losses inside chamber with perforated V-profile diffuser for different geometric parameters such as entry length, apex angle, geometry and pattern of hole, plate thickness, porosity and surface roughness has been carried out numerically. It is seen that the experimental results validated with the numerical turbulence model results

    A global optimization approach applied to structural dynamic updating

    In this paper, the application of stochastic global optimization tech- niques, in particular the GlobalSearch and MultiStart solvers from MatLab®, to improve the updating of a structural dynamic model, are presented. For com- parative purposes, the efficiency of these global methods relatively to the local search method previously used in a Finite Element Model Updating program is evaluated. The obtained solutions showed that the GlobalSearch and MultiStart solvers are able to achieve a better solution than the local solver previously used, in the updating of a structural dynamic model. The results show also that the GlobalSearch solver is more efficient than the MultiStart, since requires less computational effort to obtain the global solution.Fundação para a Ciência e a Tecnologia (FCT

    Endothelial function in patients with familial Mediterranean fever-related amyloidosis and association with cardiovascular events

    Objectives. Secondary amyloidosis is the most important complication of FMF and endothelial function is more severely impaired. Elevated asymmetric dimethyl arginine (ADMA) may mediate the excess cardiovascular disease (CVD) risk of this group. We aimed to compare endothelial function characteristics, including ADMA, in patients with FMF-related amyloidosis and primary glomerulopathies and to define risk factors for a CVD event. Methods. We undertook a cross-sectional study with prospective follow-up including consecutive patients with FMF-related amyloidosis (n = 98) or other non-diabetic glomerulopathies (n = 102). All patients had nephrotic-range proteinuria and normal glomerular filtration rate. Flow-mediated dilatation (FMD) was assessedand ADMA levels, CRP and pentraxin 3 (PTX3) were determined. Patients were followed for cardiovascular events. Results. Amyloidosis patients secondary to FMF showed higher levels of ADMA, CRP and PTX3 and lower FMD as compared with patients with other glomerulopathies. Cardiovascular events (n = 54) were registered during 3 years of follow-up. Increased ADMA levels and lower FMD were observed in patients with cardiovascular risk in both groups, but especially in individuals with amyloidosis.Conclusion. Patients with FMF-related amyloidosis have increased CVD event risk, probably related to the high ADMA levels, elevated inflammatory markers and decreased FMD measures observed in these patients

    Evolutionary Sequence Modeling for Discovery of Peptide Hormones

    There are currently a large number of “orphan” G-protein-coupled receptors (GPCRs) whose endogenous ligands (peptide hormones) are unknown. Identification of these peptide hormones is a difficult and important problem. We describe a computational framework that models spatial structure along the genomic sequence simultaneously with the temporal evolutionary path structure across species and show how such models can be used to discover new functional molecules, in particular peptide hormones, via cross-genomic sequence comparisons. The computational framework incorporates a priori high-level knowledge of structural and evolutionary constraints into a hierarchical grammar of evolutionary probabilistic models. This computational method was used for identifying novel prohormones and the processed peptide sites by producing sequence alignments across many species at the functional-element level. Experimental results with an initial implementation of the algorithm were used to identify potential prohormones by comparing the human and non-human proteins in the Swiss-Prot database of known annotated proteins. In this proof of concept, we identified 45 out of 54 prohormones with only 44 false positives. The comparison of known and hypothetical human and mouse proteins resulted in the identification of a novel putative prohormone with at least four potential neuropeptides. Finally, in order to validate the computational methodology, we present the basic molecular biological characterization of the novel putative peptide hormone, including its identification and regional localization in the brain. This species comparison, HMM-based computational approach succeeded in identifying a previously undiscovered neuropeptide from whole genome protein sequences. This novel putative peptide hormone is found in discreet brain regions as well as other organs. The success of this approach will have a great impact on our understanding of GPCRs and associated pathways and help to identify new targets for drug development

    Developing an ontological framework for effective data quality assessment and knowledge modelling

    Big data has become a major challenge in the 21st century, with research being carried out to classify, mine and extract knowledge from data obtained from disparate sources. Abundant data sources with non-standard structures complicate even more the arduous process of data integration. Currently, the major requirement is to understand the data available and detect data quality issues, with research being conducted to establish data quality assessment methods. Further, the focus is to improve data quality and maturity so that early onset of problems can be predicted and handled effectively. However, the literature highlights that comprehensive analysis, and research of data quality standards and assessment methods are still lacking. To handle these challenges, this paper presents a structured framework to standardise the process of assessing the quality of data and modelling the knowledge obtained from such an assessment by implementing an ontology. The main steps of the framework are: (i) identify user’s requirements; (ii) measure the quality of data considering data quality issues, dimensions and their metrics, and visualise this information into a data quality assessment (DQA) report; and (iii) capture the knowledge from the DQA report using an ontology that models the DQA insights in a standard reusable way. Following the proposed framework, an Excel-based tool to measure the quality of data and identify emerging issues is developed. An ontology, created in Protégé, provides a standard structure to model the data quality insights obtained from the assessment, while it is frequently updated to enrich captured knowledge, reducing time and costs for future projects. An industrial case study in the context of Through life Engineering Services, using operational data of high value engineering assets, is employed to validate the proposed ontological framework and tool; the results show a well-structured guide that can effectively assess data quality and model knowledge
