    QUALITY OF LIFE AND COGNITION IN PATIENTS WITH PARKINSON\u27S DISEASE DURING THE COVID-19 PANDEMIC

    Introduction: Restrictive isolation contributes to the containment of the virus, but it also has negative consequences for mental health. This study aimed to assess the quality of life, during the pandemic, in patients with Parkinson\u27s disease (PD), compare it with measures before the pandemic, and identify factors that may be associated with non-compliance with cognitive intervention activities. Methods: The PDQ-39 questionnaire was reapplied in a sample of PD patients previously followed up in rehabilitation program. In addition, a questionnaire to assess the follow-up of cognitive training activities was elaborated. Results: Twenty-two individuals with mean age of 62.72(7.49) years and a mean duration of the disease of 8.7(3.30) years participated in this study. There was a statistically significant difference in the total score of the PDQ-39(p=0.048), as well as in the mobility (p=0.038), stigma (p=0.035), social support(p=0.045), and cognition(p=0.026) dimensions. When analyzing the questionnaire, it was observed that most of the sample was able to follow the guidelines received during the cognitive training group (77.3%), mainly reading activities (41%), games (35%), and applications (35%). However, 77.3% reported worsening cognition, with attention (50%) and memory (34%) being highlighted, as well as sadness, discouragement, and indisposition (72%), anxiety (77.27%), and difficulty falling asleep (68%). A total of 86.36% perceived the need for professional support in mental health. Conclusion: This study showed the impact of the pandemic on individuals with PD, leading to a worse perception of quality of life and subjective complaints related to sleep disturbance, cognitive impairment, and neuropsychiatric symptoms (depression and anxiety)

    An integrative genomic approach reveals coordinated expression of intronic miR-335, miR-342, and miR-561 with deregulated host genes in multiple myeloma

    <p>Abstract</p> <p>Background</p> <p>The role of microRNAs (miRNAs) in multiple myeloma (MM) has yet to be fully elucidated. To identify miRNAs that are potentially deregulated in MM, we investigated those mapping within transcription units, based on evidence that intronic miRNAs are frequently coexpressed with their host genes. To this end, we monitored host transcript expression values in a panel of 20 human MM cell lines (HMCLs) and focused on transcripts whose expression varied significantly across the dataset.</p> <p>Methods</p> <p>miRNA expression was quantified by Quantitative Real-Time PCR. Gene expression and genome profiling data were generated on Affymetrix oligonucleotide microarrays. Significant Analysis of Microarrays algorithm was used to investigate differentially expressed transcripts. Conventional statistics were used to test correlations for significance. Public libraries were queried to predict putative miRNA targets.</p> <p>Results</p> <p>We identified transcripts specific to six miRNA host genes (<it>CCPG1</it>, <it>GULP1</it>, <it>EVL</it>, <it>TACSTD1</it>, <it>MEST</it>, and <it>TNIK</it>) whose average changes in expression varied at least 2-fold from the mean of the examined dataset. We evaluated the expression levels of the corresponding intronic miRNAs and identified a significant correlation between the expression levels of <it>MEST</it>, <it>EVL</it>, and <it>GULP1 </it>and those of the corresponding miRNAs miR-335, miR-342-3p, and miR-561, respectively. Genome-wide profiling of the 20 HMCLs indicated that the increased expression of the three host genes and their corresponding intronic miRNAs was not correlated with local copy number variations. Notably, miRNAs and their host genes were overexpressed in a fraction of primary tumors with respect to normal plasma cells; however, this finding was not correlated with known molecular myeloma groups. The predicted putative miRNA targets and the transcriptional profiles associated with the primary tumors suggest that <it>MEST</it>/miR-335 and <it>EVL/</it>miR-342-3p may play a role in plasma cell homing and/or interactions with the bone marrow microenvironment.</p> <p>Conclusion</p> <p>Our data support the idea that intronic miRNAs and their host genes are regulated dependently, and may contribute to the understanding of their biological roles in cancer. To our knowledge, this is the first evidence of deregulated miRNA expression in MM, providing insights that may lead to the identification of new biomarkers and altered molecular pathways of the disease.</p

    El objetivo del estudio fue desarrollar una revisión sistemática de literatura sobre la competencia digital en estudiantes universitarios, con el fin de orientar futuras investigaciones. Se realizó una investigación cualitativa documental de dos fases: heurística y hermenéutica. Los artículos mayoritariamente utilizan el enfoque cuantitativo y alcance descriptivo, revelando información valiosa sobre la competencia digital de estudiantes universitarios. Sin embargo, dada la naturaleza descriptiva de los estudios, no se profundiza lo suficiente en las variables que pudieran explicar los resultados obtenidos. Se concluye que se requiere realizar investigaciones con alcances explicativos y correlacionales para comprender mejor la competencia digital en estudiantes universitarios, considerando factores como evaluaciones del desempeño y creencias personales

    Tracking Down of a Selected Panel of Parabens: A Validated Method to Evaluate Their Occurrence in Skin Layers

    A method was set up and validated to identify and quantify seven parabens in each of the three skin layers, i.e., Stratum Corneum, Epidermis, and Dermis, because, even if only some analogues are legally allowed in Europe, forbidden parabens are also detected in many personal care products and therefore can be absorbed by the skin. A solid/liquid extraction followed by a gradient elution chromatographic separation method was performed and validated according to European guidelines. Our validated method afforded the detection of all seven parabens with limit of detection values ranging from 0.026 to 0.090 μg mL−1 and recoveries ranging from 61.80 to 105.73 μg mL−1 at high and low concentration values (50.0–5.0 μg mL−1), respectively. The proposed method can help assess paraben’s skin bioaccumulation since people are repeatedly exposed to consumer goods containing parabens in their daily routine, posing a chronic risk to human health

    Parabens Permeation through Biological Membranes: A Comparative Study Using Franz Cell Diffusion System and Biomimetic Liquid Chromatography

    Parabens (PBs) are used as preservatives to extend the shelf life of various foodstuffs, and pharmaceutical and cosmetic preparations. In this work, the membrane barrier passage potential of a subset of seven parabens, i.e., methyl-, ethyl-, propyl- isopropyl, butyl, isobutyl, and benzyl paraben, along with their parent compound, p-hydroxy benzoic acid, were studied. Thus, the Franz cell diffusion (FDC) method, biomimetic liquid chromatography (BLC), and in silico prediction were performed to evaluate the soundness of both describing their permeation through the skin. While BLC allowed the achievement of a full scale of affinity for membrane phospholipids of the PBs under research, the permeation of parabens through Franz diffusion cells having a carbon chain > ethyl could not be measured in a fully aqueous medium, i.e., permeation enhancer-free conditions. Our results support that BLC and in silico prediction alone can occasionally be misleading in the permeability potential assessment of these preservatives, emphasizing the need for a multi-technique and integrated experimental approach

    The VEI 2 Christmas 2018 Etna Eruption: A Small But Intense Eruptive Event or the Starting Phase of a Larger One?

    The Etna flank eruption that started on 24 December 2018 lasted a few days and involved the opening of an eruptive fissure, accompanied by a seismic swarm and shallow earthquakes, significant SO2 flux release, and by large and widespread ground deformation, especially on the eastern flank of the volcano. Lava fountains and ash plumes from the uppermost eruptive fissure accompanied the opening stage, causing disruption to Catania International Airport, and were followed by a quiet lava effusion within the barren Valle del Bove depression until 27 December. This was the first flank eruption to occur at Etna in the last decade, during which eruptive activity was confined to the summit craters and resulted in lava fountains and lava flow output from the crater rims. In this paper, we used ground and satellite remote sensing techniques to describe the sequence of events, quantify the erupted volumes of lava, gas, and tephra, and assess volcanic hazards.Publishedid 9056V.     Targeting COPZ1 non-oncogene addiction counteracts the viability of thyroid tumor cells

    Thyroid carcinoma is generally associated with good prognosis, but no effective treatments are currently available for aggressive forms not cured by standard therapy. To find novel therapeutic targets for this tumor type, we had previously performed a siRNA-based functional screening to identify genes essential for sustaining the oncogenic phenotype of thyroid tumor cells, but not required to the same extent for the viability of normal cells (non-oncogene addiction paradigm). Among those, we found the coatomer protein complex ζ1 (COPZ1) gene, which is involved in intracellular traffic, autophagy and lipid homeostasis. In this paper, we investigated the mechanisms through which COPZ1 depletion leads to thyroid tumor cell death. We showed that siRNA-mediated COPZ1 depletion causes abortive autophagy, endoplasmic reticulum stress, unfolded protein response and apoptosis. Interestingly, we observed that mouse tumor xenografts, locally treated with siRNA targeting COPZ1, showed a significant reduction of tumor growth. On the whole, we demonstrated for the first time the crucial role of COPZ1 in the viability of thyroid tumor cells, suggesting that it may be considered an attractive target for novel therapeutic approaches for thyroid cancer

    Monitoring of Pollutants Content in Bottled and Tap Drinking Water in Italy

    The concentration levels of thirteen organic pollutants and selected heavy metals were investigated in 40 plastics bottled and tap water samples. Some of the selected contaminants have an ascertained or suspected endocrine disrupting activity, such as Bisphenol A (BPA) and its analogs, and Bis 2-ethylhexyl phthalate (DEHP), which are used by industries as plasticizers. The most frequently detected pollutants were Bisphenol AF (BPAF) (detection frequency (DF) = 67.5%, mean 387.21 ng L−1), DEHP (DF = 62.5%, mean 46.19 µg L−1) and BPA (DF = 60.0%, mean 458.57 ng L−1), with higher concentration levels found in tap waters. Furthermore, a possible level of exposure to thirteen pollutants via drinking water intake was calculated. Our findings show that, even though the occurrence of contaminants and heavy metals in drinking waters does not pose an immediate, acute health risk for the population, their levels should be constantly monitored and “hard-wired” into everyday practice. Indeed, the health impact to the continuous and simultaneous intake of a huge variety of xenobiotics from various sources by humans is complex and still not fully understood
